APPLICATION OF KJELDAHL AND DUMAS COMBUSTION METHODS FOR NITROGEN ANALYSIS
O método padrão de Kjeldahl vem sendo substituído pelo método de combustão Dumas
na determinação do nitrogênio total (NT) em alimentos. Estes procedimentos foram utilizados para
avaliar amostras de rotina em laboratórios de nutrição animal (concentrados, forragens, excreta e
conteúdo duodenal). A covariável nitrato (N-NO3) interferiu na determinação do NT em função da
metodologia empregada. Foi observada uma correlação positiva entre os métodos utilizados para
cada grupo avaliado: ingredientes volumosos, Kjeldahl-N = 1.1661 + 0.9216 Dumas-N (R2=0.99);
alimentos energéticos, Kjeldahl-N = 0.6376 + 0.9445 Dumas-N (R2=0.72) e alimentos protéicos, Kjeldahl-
N = 6.5638 + 0.8615 Dumas-N (R2=0.97). As médias para NT dos alimentos, ajustadas para a
covariável, não diferiram entre os métodos (P > 0,05). O desvio padrão do teor de N dentro e entre os
métodos utilizados estão dentro dos intervalos permitidos pela literatura, o que, associado ao fato da
metodologia de Dumas consistir em uma técnica não poluente ao ambiente, indica ser esta metodologia
capaz de substituir com vantagens o procedimento de Kjeldahl em análises de rotina em laboratórios
de nutrição animal.
Abstract
The Kjeldahl procedure has been replaced by Dumas combustion procedure for total N
determination in feeds. These methods were compared using routine samples in the animal nutrition
laboratory (concentrates, forages, excreta and duodenal content). NO3-N covariate interfered on total
N determination according to the procedure used. A significant correlation for crude protein (CP) content
was observed between methods used for each group: grass silage component, Kjeldahl-N = 1.1661 +
0.9216 Dumas-N (R2=0.99); energy feeds, Kjeldahl-N = 0.6376 + 0.9445 Dumas-N (R2=0.72) and
protein feeds, Kjeldahl-N = 6.5638 + 0.8615 Dumas-N (R2=0.97). Means for total N contents from
components, adjusted by covariance, were not different (P > 0.05) between both methods. Considering
that the standard deviation for N content obtained by the two methods stayed within the intervals observed
in the literature, corroborated by the fact that Dumas combustion procedure using the analyzer LECOFP
528 LC is not harmful to the environment it was concluded, that this method can replace with
advantages the Kjeldahl procedure in routine animal nutrition N analyzes
APLICAÇÃO DE TÉCNICAS PARA ESTUDOS DE INGESTÃO, COMPOSIÇÃO DA DIETA E DIGESTIBILIDADE
A adequada nutrição dos animais de produção é dependente do conhecimento das
exigências dos animais e valor nutritivo dos alimentos. No entanto, em muitas situações, a obtenção
das informações necessárias não é possível ou é dificultada pela falta de recursos ou alto custo dos
procedimentos experimentais, sendo necessária a aplicação de técnicas que possam estimar de
forma precisa os parâmetros desejados. A avaliação de animais em pastagem é bastante dificultada
pela impossibilidade em se controlar a ingestão e excreção dos animais, onde a aplicação de
modelos matemáticos e a utilização de indicadores surgem como alternativas que podem reduzir
custos e aumentar a precisão das estimativas. O objetivo dessa revisão foi apresentar as principais
técnicas em uso que visam determinar os parâmetros consumo, digestibilidade e composição da
dieta em diferentes sistemas de produção assim como as particularidades, vantagens e desvantagens
dos métodos empregados. A escolha da técnica empregada depende fundamentalmente dos
objetivos a serem alcançados, dos recursos disponíveis e da precisão necessária. A utilização de
indicadores é uma ferramenta bastante apropriada, possibilitando a estimativa de consumo e
digestibilidade nos diferentes compartimentos gastrintestinais. O emprego dos alcanos tem
apresentado resultados bastante satisfatórios na avaliação da composição das dietas, em especial
em sistemas de pastagem
(Ruminal methane emission related aspects in cattle production systems
Methane is well known as a greenhouse
effect intensifier. Ruminants, during the ruminal digestive
fermentation process, transform carbohydrates available
in the diet to volatile fatty acids, with production of carbon
dioxide and methane. Therefore, ruminants are known
as important methane source to the atmosphere.
Methane production also means animal losses of
ingested energy. Ruminal fermentation process is driven
by dry matter intake, and feed quality. Therefore,
consume of low quality diets will produce more methane
per product unit (milk or beef) compared to high
producing animals ingesting great amounts of high quality
diets. The sulphur hex fluoride gas tracer technique did
allow good methane emission measurements by grazing
or free managed animals. Results show that the
reduction of methane emission by cattle is related to
better quality diet, to improvement of forage quality of
pastures, to grain inclusion in diet, to animal breeds with
greater production potential, and other practices that
improve production efficiency, and result in shorter
production cycles.O gás metano possui reconhecidamente
um importante papel como intensificador do efeito
estufa. Pelo seu processo digestivo de fermentação
entérica, os ruminantes convertem os carboidratos
presentes na dieta em ácidos graxos voláteis,
resultando na produção de dióxido de carbono e
metano. Dessa forma, os ruminantes são
reconhecidos como importantes emissores de
metano ruminal para a atmosfera, sendo que a
produção deste gás representa ainda perdas
energéticas para os animais em relação à energia
consumida. Animais consumindo dietas de baixa
qualidade podem produzir mais metano por unidade
de produto (carne ou leite) em relação aos animais
de alta produção consumindo dietas de melhor
qualidade em maiores níveis de ingestão. Resultados
têm mostrado que a redução da emissão de metano
pela pecuária está ligada à melhoria da dieta, à
melhoria dos pastos, à suplementação alimentar, à
seleção por maior potencial genético de produção e
a outras medidas que reflitam na melhor eficiência
produtiva, resultando em menores ciclos de
produção
Expression of E-cadherin, Snail and Hakai in epithelial cells isolated from the primary tumor and from peritumoral tissue of invasive ductal breast carcinomas
Epithelial intercellular cohesion, mainly mediated by E-cadherin (CDH1) expression and function, may be deregulated during cancer cell invasion of adjacent tissues and lymphatic and vascular channels. CDH1 expression is down-modulated in invasive lobular breast carcinomas but its regulation in invasive ductal carcinomas (IDC) is less clear. CDH1 expression is repressed by transcription factors such as Snail (SNAI1) and its product is degraded after Hakai ubiquitination. We compared CDH1, SNAI1 and HAKAI mRNA expression in IDC and paired adjacent normal breast tissue and evaluated its relation with node metastasis and circulating tumor cells. Matched tumor/peritumoral and blood samples were collected from 30 patients with early IDC. Epithelial cells from each compartment (tumor/peritumoral) were recovered by an immunomagnetic method and gene expression was determined by real time RT-PCR. There were no differences in CDH1, SNAI1 and HAKAI mRNA expression between tumor and corresponding peritumoral samples and no differential tumoral gene expression according to nodal involvement. Another 30 patients with a long-term follow-up (at least 5 years) and a differential prognosis (good or poor, as defined by breast cancer death) had E-cadherin and Snail protein detected by immunohistochemistry in tumor samples. In this group, E-cadherin-positive expression, but not Snail, may be associated with a better prognosis. This is the first report simultaneously analyzing CDH1, SNAI1 and HAKAI mRNA expression in matched tumor and peritumoral samples from patients with IDC. Influence of edapho-climatic factors on the sporulation and colonization of arbuscular mycorrhizal fungi in two Amazonian native fruit species
Arbuscular mycorrhizal fungi (AMF) colonization and spore numbers in the rhizosphere of two fruit species, Paullinia cupana Mart. and Theobroma grandiflorum Schum., growing in a terra firme ecosystem in Central Amazonia were studied from August 1998 to May 2000. Climatic and edaphic factors were also determined to investigate their influence on mycorrhizal variables. Soil pH, Al, Mn and effective cation exchange capacity exhibited seasonal variations during the investigation period. Temporal variations in mycorrhizal colonization levels and spore numbers occurred, indicating seasonality. Moreover, the patterns of mycorrhizal colonization levels and spore numbers for both host species were similar during the studied period. Mycorrhizal variables were related to climatic and edaphic factors, however, the intensity and type of influence of climatic and soil characteristics on AMF development tended to vary with the season and host plant species in Central Amazonia conditions
Search for displaced vertices arising from decays of new heavy particles in 7 TeV pp collisions at ATLAS
We present the results of a search for new, heavy particles that decay at a
significant distance from their production point into a final state containing
charged hadrons in association with a high-momentum muon. The search is
conducted in a pp-collision data sample with a center-of-mass energy of 7 TeV
and an integrated luminosity of 33 pb^-1 collected in 2010 by the ATLAS
detector operating at the Large Hadron Collider. Production of such particles
is expected in various scenarios of physics beyond the standard model. We
observe no signal and place limits on the production cross-section of
supersymmetric particles in an R-parity-violating scenario as a function of the
neutralino lifetime. Limits are presented for different squark and neutralino
masses, enabling extension of the limits to a variety of other models.Comment: 8 pages plus author list (20 pages total), 8 figures, 1 table, final
