    The Structure of Clusters with Bimodal Distributions of Galaxy Radial Velocities. I. A1035

    The structure of the A1035 cluster of galaxies (10h32m +40d13', cz ~ 22000 km/s), which exhibits a bimodal distribution of galaxy radial velocities (\Delta V\approx 3000 km/s), is analyzed using three methods of determining the relative distances to clusters from early-type galaxies: the Kormendy relation corrected for the dependence of residuals on galaxy magnitude, the photometric plane, and the fundamental plane. We use the data obtained with the 1-m telescope of the Special Astrophysical Observatory of the Russian Academy of Sciences and SDSS (DR5) data to show that A1035 consists of two gravitationally unbound independent clusters. These clusters with the velocity dispersions of 566 km/s and 610 km/s and masses within R_200 equal to 2.7 10^14 and 3.5 10^14 M_sun, respectively, obey the Hubble law.Comment: 13 pages, 3 tables, 6 figure

    Effect of dexamethasone therapy on factors of adhesiveness and coagulation in acute lower limb ischemia

    Leukocyte-platelet adhesion during hypoxia, tissue damage, activation of inflammation and coagulation is associated with the expression of ICAM-1 membrane molecules and integrins by blood and tissue cells. At the same time, platelet adhesion receptors determine their adhesion to the endothelium and recruited lymphocytes. The role of platelets in the pathogenesis of ischemic cardiovascular diseases also consists in their ability to modulate both hemostasis and inflammatory reactions, which is accompanied by the secretion of inflammatory mediators and factors that promote the recruitment of leukocytes to tissue damage sites. Purpose of the study: to study the effect of the synthetic glucocorticoid dexamethasone on the expression of adhesion receptors CD18+ and CD54+ on leukocytes, the content of platelets and fibrinogen in the blood of patients with ALLI, the relationship of these indicators with the severity and outcome of the disease.To study the effect of anti-inflammatory therapy, a group of 32 patients treated with dexamethasone was formed; the comparison group was represented by 71 patients with basic therapy, the control group consisted of 15 volunteers. After revascularization, all patients received antiplatelet and anticoagulant therapy. Dexamethasone infusions were carried out in a course of 4 to 6 days after reconstructive surgery. In all patients, the content of C-reactive protein in the blood, the content of platelets and fibrinogen were determined. The number of lymphocytes expressing adhesion molecules ICAM-1 (CD54+) and integrins (CD18+) was counted using the immunocytochemical method. Studies were performed before surgery and on days 1, 3, 7, and 10 after surgery.With exacerbation of ischemia and damage to the endothelium, the accumulation of cytolysis products, the expression of adhesion molecules increases both on endotheliocytes and on inflammatory effector cells – leukocytes and platelets. Adhesion molecules conduct an activation signal inside the cell, which promotes adhesion of leukocytes and platelets to the endothelium, lymphocytic-platelet adhesion, the formation of a parietal thrombus, and possible occlusion of damaged vessels. Increased expression of adhesion molecules is associated with the activation of metabolism, inflammation, coagulation and oxidative stress, stimulates all hematopoietic lineages, including platelets. The level of involvement of cellular reactions in the pathogenesis of the disease affects the effectiveness and duration of treatment, the risk of recurrent thrombosis and death. Anti-inflammatory therapy with dexamethasone contributed to earlier remission, a decrease in the proportion of infectious complications, such as wound suppuration from 10% to 6%, the number of necessary amputations from 32% to 16%, the frequency of deaths from 31% to 6%, and a reduction in hospital stay from 13 days to 10.Inflammation, adhesiveness of effector cells and thrombosis are important factors in the pathogenesis of acute lower limb ischemia. Therapy with dexamethasone helps to reduce the level of systemic inflammatory response, the number of necessary amputations, the number of complications and adverse outcomes in the treatment of ALLI, and reduce the length of stay in the hospital

    The experience of the introduction of convective methods since the eighties of the 20th century is described.Aim: to evaluate the infl uence of convective transport supplementation on renal replacement therapy results. Methods and results. The number of convective procedures was applied: hemodiafi ltration with commercial substituted fl uid, paired fi ltration dialysis, acetate-free biofi ltration, original hybrid methods, online hemodiafi ltration. Themain trend was the magnifi cation of substituted volume. In the long run after switching from conventional dialysis to convective therapy the increase of Kt/V, the decline of phosphate, the rise of serum albumin, the decrease of CRP level and ESA consumption, QOL improvement and the drop in severity of intradialysis complications were observed. Over last six years online hemodiafi ltration has become the standard of dialysis therapy in our Center.Conclusion. The undoubted advantages of convective modalities and accessibility of online hemodiafi ltration make these methods the gold standard of dialysis therapy.Описан опыт внедрения в практику программного гемодиализа конвективных методов с 80-х годов про-шлого столетия по настоящее время.Цель: оценить влияние конвективного переноса, прежде всего в рамках гемодиафильтрации «онлайн», на результаты заместительной терапии.Методы и результаты.Применены различные конвективные методики: гемодиафильтрация с использованием коммерческого замещающего раствора, парные фильтрация-диализ, безацетатная биофильтрация, оригинальные гибридные методики, гемодиафильтрация «онлайн». Основной тенденцией при этом было увеличение объема замещения за процедуру. При наблюдении в длительной перспективе после перевода со стандартного гемодиализа при использовании конвективных методик отмечалось увеличение эффективности лечения по индексу Kt/V, снижение фосфатемии, повышение концентрации альбумина, уменьшение уровня С-реактивного белка и потребности в эритропоэзтимулирующих препаратах, повышение качества жизни и снижение выраженности интрадиализной симптоматики. В течение последних 6 лет гемодиафильтрация «онлайн» стала стандартом диализного лечения в нашем учреждении.Заключение. Несомненные преимущества конвективных методик и доступность практически неограниченного объема замещающей жидкости, характеризующая гемодиафильтрацию «онлайн», делают этот метод «золотым» стандартом диализной терапии

    Our growing awareness of the microbial world’s importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable bacterial and archaeal ribosomal RNA gene sequences to be followed across multiple studies and allow us to explore patterns of diversity at an unprecedented scale. The result is both a reference database giving global context to DNA sequence data and a framework for incorporating data from future studies, fostering increasingly complete characterization of Earth’s microbial diversity

    Multiple identification of most important waterborne protozoa in surface water used for irrigation purposes by 18S rRNA amplicon-based metagenomics

    [EN] Understanding waterborne protozoan parasites (WPPs) diversity has important implications in public health. In this study, we evaluated a NGS-based method as a detection approach to identify simultaneously most important WPPs using 18S rRNA high-throughput sequencing. A set of primers to target the V4 18S rRNA region of WPPs such as Cryptosporidium spp., Giardia sp., Blastocystis sp., Entamoeba spp, Toxoplasma sp. and free-living amoebae (FLA) was designed. In order to optimize PCR conditions before sequencing, both a mock community with a defined composition of representative WPPs and a real water sample inoculated with specific WPPs DNA were prepared. Using the method proposed in this study, we have detected the presence of Giardia intestinalis, Acanthamoeba castellanii, Toxoplasma gondii, Entamoeba histolytica and Blastocystis sp. at species level in real irrigation water samples. Our results showed that untreated surface irrigation water in open fields can provide an important source of WPPs.
