    Spatial monitoring of groundwater drawdown and rebound associated with quarry dewatering using automated time-lapse electrical resistivity tomography and distribution guided clustering

    Dewatering systems used for mining and quarrying operations often result in highly artificial and complex groundwater conditions, which can be difficult to characterise and monitor using borehole point sampling approaches. Here automated time-lapse electrical resistivity tomography (ALERT) is considered as a means of monitoring subsurface groundwater dynamics associated with changes in the dewatering regime in an operational sand and gravel quarry. We considered two scenarios: the first was unplanned interruption to dewatering due to a pump failure for a period of several days, which involved comparing ALERT monitoring results before and after groundwater rebound; the second involved a planned interruption to pumping over a period of 6 h, for which near-continuous ALERT monitoring of groundwater rebound and drawdown was undertaken. The results of the second test were analysed using distribution guided clustering (DGC) to provide a more quantitative and objective assessment of changes in the subsurface over time. ALERT successfully identified groundwater level changes during both monitoring scenarios. It provided a more useful indication of the rate of water level rise and maximum water levels than piezometer monitoring results. This was due to the piezometers rapidly responding to pressure changes at depth, whilst ALERT/DGC provided information of slower changes associated with the storage and delayed drainage of water within the sediment. By applying DGC we were able to automatically and quantitatively define changes in the resistivity sections, which correlated well with the direct observations of groundwater at site. For ERT monitoring applications that generate numerous time series, the use of DGC could significantly enhance the efficiency of data interpretation, and provide a means of automating groundwater monitoring through assigning alarm thresholds associated with rapid changes in groundwater conditions

    Amelioration of systemic inflammation via the display of two different decoy protein receptors on extracellular vesicles

    Extracellular vesicles (EVs) can be functionalized to display specific protein receptors on their surface. However, surface-display technology typically labels only a small fraction of the EV population. Here, we show that the joint display of two different therapeutically relevant protein receptors on EVs can be optimized by systematically screening EV-loading protein moieties. We used cytokine-binding domains derived from tumour necrosis factor receptor 1 (TNFR1) and interleukin-6 signal transducer (IL-6ST), which can act as decoy receptors for the pro-inflammatory cytokines tumour necrosis factor alpha (TNF-α) and IL-6, respectively. We found that the genetic engineering of EV-producing cells to express oligomerized exosomal sorting domains and the N-terminal fragment of syntenin (a cytosolic adaptor of the single transmembrane domain protein syndecan) increased the display efficiency and inhibitory activity of TNFR1 and IL-6ST and facilitated their joint display on EVs.     Age at first birth in women is genetically associated with increased risk of schizophrenia

    Prof. Paunio on PGC:n jäsenPrevious studies have shown an increased risk for mental health problems in children born to both younger and older parents compared to children of average-aged parents. We previously used a novel design to reveal a latent mechanism of genetic association between schizophrenia and age at first birth in women (AFB). Here, we use independent data from the UK Biobank (N = 38,892) to replicate the finding of an association between predicted genetic risk of schizophrenia and AFB in women, and to estimate the genetic correlation between schizophrenia and AFB in women stratified into younger and older groups. We find evidence for an association between predicted genetic risk of schizophrenia and AFB in women (P-value = 1.12E-05), and we show genetic heterogeneity between younger and older AFB groups (P-value = 3.45E-03). The genetic correlation between schizophrenia and AFB in the younger AFB group is -0.16 (SE = 0.04) while that between schizophrenia and AFB in the older AFB group is 0.14 (SE = 0.08). Our results suggest that early, and perhaps also late, age at first birth in women is associated with increased genetic risk for schizophrenia in the UK Biobank sample. These findings contribute new insights into factors contributing to the complex bio-social risk architecture underpinning the association between parental age and offspring mental health.Peer reviewe

    The Cholecystectomy As A Day Case (CAAD) Score: A Validated Score of Preoperative Predictors of Successful Day-Case Cholecystectomy Using the CholeS Data Set

    Background Day-case surgery is associated with significant patient and cost benefits. However, only 43% of cholecystectomy patients are discharged home the same day. One hypothesis is day-case cholecystectomy rates, defined as patients discharged the same day as their operation, may be improved by better assessment of patients using standard preoperative variables. Methods Data were extracted from a prospectively collected data set of cholecystectomy patients from 166 UK and Irish hospitals (CholeS). Cholecystectomies performed as elective procedures were divided into main (75%) and validation (25%) data sets. Preoperative predictors were identified, and a risk score of failed day case was devised using multivariate logistic regression. Receiver operating curve analysis was used to validate the score in the validation data set. Results Of the 7426 elective cholecystectomies performed, 49% of these were discharged home the same day. Same-day discharge following cholecystectomy was less likely with older patients (OR 0.18, 95% CI 0.15–0.23), higher ASA scores (OR 0.19, 95% CI 0.15–0.23), complicated cholelithiasis (OR 0.38, 95% CI 0.31 to 0.48), male gender (OR 0.66, 95% CI 0.58–0.74), previous acute gallstone-related admissions (OR 0.54, 95% CI 0.48–0.60) and preoperative endoscopic intervention (OR 0.40, 95% CI 0.34–0.47). The CAAD score was developed using these variables. When applied to the validation subgroup, a CAAD score of ≤5 was associated with 80.8% successful day-case cholecystectomy compared with 19.2% associated with a CAAD score >5 (p < 0.001). Conclusions The CAAD score which utilises data readily available from clinic letters and electronic sources can predict same-day discharges following cholecystectomy

    In this study, we assess the suitability of ERT for UK sand and gravel deposit assessment. To this end, we have reviewed the characteristics of deposits in terms of geological setting, thickness and heterogeneity to inform our survey design strategy. We have collated existing data on the electrical properties of UK sand and gravel, reviewed previous examples of ERT sand and gravel surveys, and undertaken detailed geophysical studies at seven potential or active sand and gravel extraction locations in East Anglia and the East Midlands

    Juazeiro (Zizyphus joazeiro Mart.) é uma árvore brasileira típica dos sertões nordestinos. Endêmica da Caatinga, ela ocorre nos diversos Estados do Nordeste do Brasil e apresenta grande potencial econômico e é importante para a região semi-árida. Assim, este estudo teve como objetivo determinar a melhor profundidade de semeadura para a emergência de plântulas de Zizyphus joazeiro. O experimento foi realizado em casa-de-vegetação, tendo como substrato areia lavada e esterilizada contida em bandejas de plástico, com 4 repetições de 25 unidades de dispersão cada uma, nas seguintes profundidades (um, dois, três, quatro e cinco centímetros). A semeadura de juazeiro em ambiente protegido deve ser feita na profundidade entre 1,0 e 1,6cm, resultando em elevado aproveitamento das unidades de dispersão. A profundidade de 1cm resultou em menor tempo para a germinação total.<br>Zizyphus joazeiro Mart. is a typical Brazilian tree of the northeastern dry lands, being endemic to the Caatinga. It occurs in various states of Northeast Brazil and provides a great economic potential and importance for this semi-arid region. This study had as objective to determine the best sowing depth of Zizyphus joazeiro. In greenhouse sand substratum, with 4 replications of 25 units of dispersion each one, in the following depths (one, too, tree, four and five centimeters). It was ended that the sowing of juazeiro in nursery should be done in the depth of 1.0 to 1.6cm. The depth of 1 cm resulted in smaller time for the total germination

    Premature ovarian failure: morphological and ultrastructural aspects

    The authors documented by means of light and transmission electron microscopy that the ovaries of women with premature ovarian failure (POF) displayed dense connective tissue and rare corpora albicantia. Eight of the ten studied cases did not present ovarian follicles; in two cases, it was verified the presence of ovarian follicles, atypical primordial follicles and in one case, a corpus luteum was identified (after stimulation with exogenous gonadotrophin). Regarding the ultrastructural analysis, it was noted that the fibroblasts were united one to each other by cellular prolongations that formed a woof, constituting a cellular syncicius.Os autores analisaram, através de microscopia de luz e de microscopia eletrônica de transmissão, os ovários de mulheres com insuficiência ovariana prematura. Observaram que são constituídos por tecido conjuntivo denso e raros corpos albicantes. Oito dos 10 casos não apresentavam folículos primordiais; em um caso verificou-se a presença de folículos ovarianos atípicos e, em outro, identificou-se corpo lúteo (após estimulação com gonadotrofina exógena).
