    A multi-scale study of 3D printed Co-Al2O3 catalyst monoliths versus spheres

    This study demonstrates the characteristics of two model packing configurations: 3D printed (3DP) catalyst monoliths on the one hand, and their conventional counterparts, packed beds of spheres, on the other. Cobalt deposited on alumina is selected as a convenient model system for this work, due to its wide spread use in many catalytic reactions. 3DP constructs were produced from alumina powder impregnated with cobalt nitrate while the alumina spheres were directly impregnated with the same cobalt nitrate precursor. The form of the catalyst, the impregnation process, as well as the thermal history, were found to have a significant effect on the resulting cobalt phases. Probing the catalyst bodies in situ by XRD-CT indicated that the level of dispersion of identified Co phases (Co3O4 reduced to CoO) across the support is maintained under reduction conditions. The packed bed of spheres exhibits a non-uniform distribution of cobalt phases, including a core-shell morphology with an average crystallite size of 10–14 nm across the sphere, while the 3DP monolith exhibits a uniform distribution of cobalt phases with an average crystallite size of 5–12 nm upon reduction from Co3O4 to CoO. Computational Fluid Dynamics (CFD) modelling was carried out to develop digital twins and assess the effect of the geometry of both configurations on the pressure drop and velocity profiles. Finally, the activity of both Cobalt-based catalyst geometries was assessed in terms of their conversion, selectivity and turn over frequencies under model multiphase (selective oxidation) reaction conditions, which showed that the desired 3D printed monolithic geometries can offer distinct advantages to the reactor design

    Characterization of the Molecular Determinants of Primary HIV-1 Vpr Proteins: Impact of the Q65R and R77Q Substitutions on Vpr Functions

    Although HIV-1 Vpr displays several functions in vitro, limited information exists concerning their relevance during infection. Here, we characterized Vpr variants isolated from a rapid and a long-term non-progressor (LTNP). Interestingly, vpr alleles isolated from longitudinal samples of the LTNP revealed a dominant sequence that subsequently led to diversity similar to that observed in the progressor patient. Most of primary Vpr proteins accumulated at the nuclear envelope and interacted with host-cell partners of Vpr. They displayed cytostatic and proapoptotic activities, although a LTNP allele, harboring the Q65R substitution, failed to bind the DCAF1 subunit of the Cul4a/DDB1 E3 ligase and was inactive. This Q65R substitution correlated with impairment of Vpr docking at the nuclear envelope, raising the possibility of a functional link between this property and the Vpr cytostatic activity. In contradiction with published results, the R77Q substitution, found in LTNP alleles, did not influence Vpr proapoptotic activity

    What Does Ecological Farming Mean for Farm Labour?

    Summary: Ecological farming, such as organic and low‐input farming, is gaining popularity in the public discourse. One question is how this type of farming may impact farm labour from a socio‐economic point of view. The article first discusses how low‐input farming practices (i.e. with lower reliance on inputs derived from fossil fuels) may affect the economic returns to labour, measured as the farm’s revenue per hour of labour input, on data from the Farm Accountancy Data Network (FADN) in 2004‐‐2015 for four European countries. Returns to labour appear to be highest at the two extremes – very low‐input farms and highly intensive farms. Farms in the low‐input end of the spectrum are in the minority, while the overwhelming majority of farms are intensive and have internal economic incentives to intensify further. The article also analyses how working conditions differ between organic and conventional dairy farms in two European countries based on interviews with farmers in 2019. Results show that all dimensions of working conditions are affected by being an organic farm or not, but this is not the only factor. There are many influences on working conditions, such as the production context and workforce composition

    Functional mechanisms underlying pleiotropic risk alleles at the 19p13.1 breast-ovarian cancer susceptibility locus

    A locus at 19p13 is associated with breast cancer (BC) and ovarian cancer (OC) risk. Here we analyse 438 SNPs in this region in 46,451 BC and 15,438 OC cases, 15,252 BRCA1 mutation carriers and 73,444 controls and identify 13 candidate causal SNPs associated with serous OC (P=9.2 × 10-20), ER-negative BC (P=1.1 × 10-13), BRCA1-associated BC (P=7.7 × 10-16) and triple negative BC (P-diff=2 × 10-5). Genotype-gene expression associations are identified for candidate target genes ANKLE1 (P=2 × 10-3) and ABHD8 (P<2 × 10-3). Chromosome conformation capture identifies interactions between four candidate SNPs and ABHD8, and luciferase assays indicate six risk alleles increased transactivation of the ADHD8 promoter. Targeted deletion of a region containing risk SNP rs56069439 in a putative enhancer induces ANKLE1 downregulation; and mRNA stability assays indicate functional effects for an ANKLE1 3′-UTR SNP. Altogether, these data suggest that multiple SNPs at 19p13 regulate ABHD8 and perhaps ANKLE1 expression, and indicate common mechanisms underlying breast and ovarian cancer risk

    The FANCM:p.Arg658* truncating variant is associated with risk of triple-negative breast cancer

    Abstract: Breast cancer is a common disease partially caused by genetic risk factors. Germline pathogenic variants in DNA repair genes BRCA1, BRCA2, PALB2, ATM, and CHEK2 are associated with breast cancer risk. FANCM, which encodes for a DNA translocase, has been proposed as a breast cancer predisposition gene, with greater effects for the ER-negative and triple-negative breast cancer (TNBC) subtypes. We tested the three recurrent protein-truncating variants FANCM:p.Arg658*, p.Gln1701*, and p.Arg1931* for association with breast cancer risk in 67,112 cases, 53,766 controls, and 26,662 carriers of pathogenic variants of BRCA1 or BRCA2. These three variants were also studied functionally by measuring survival and chromosome fragility in FANCM−/− patient-derived immortalized fibroblasts treated with diepoxybutane or olaparib. We observed that FANCM:p.Arg658* was associated with increased risk of ER-negative disease and TNBC (OR = 2.44, P = 0.034 and OR = 3.79; P = 0.009, respectively). In a country-restricted analysis, we confirmed the associations detected for FANCM:p.Arg658* and found that also FANCM:p.Arg1931* was associated with ER-negative breast cancer risk (OR = 1.96; P = 0.006). The functional results indicated that all three variants were deleterious affecting cell survival and chromosome stability with FANCM:p.Arg658* causing more severe phenotypes. In conclusion, we confirmed that the two rare FANCM deleterious variants p.Arg658* and p.Arg1931* are risk factors for ER-negative and TNBC subtypes. Overall our data suggest that the effect of truncating variants on breast cancer risk may depend on their position in the gene. These solutions allow life scientists to turn the exponentially growing amount of data into knowledge. Here, we provide an overview of SIB's resources and competence areas, with a strong focus on curated databases and SIB's most popular and widely used resources. In particular, SIB's Bioinformatics resource portal ExPASy features over 150 resources, including UniProtKB/Swiss-Prot, ENZYME, PROSITE, neXtProt, STRING, UniCarbKB, SugarBindDB, SwissRegulon, EPD, arrayMap, Bgee, SWISS-MODEL Repository, OMA, OrthoDB and other databases, which are briefly described in this article

    A case-only study to identify genetic modifiers of breast cancer risk for BRCA1/BRCA2 mutation carriers

    Abstract: Breast cancer (BC) risk for BRCA1 and BRCA2 mutation carriers varies by genetic and familial factors. About 50 common variants have been shown to modify BC risk for mutation carriers. All but three, were identified in general population studies. Other mutation carrier-specific susceptibility variants may exist but studies of mutation carriers have so far been underpowered. We conduct a novel case-only genome-wide association study comparing genotype frequencies between 60,212 general population BC cases and 13,007 cases with BRCA1 or BRCA2 mutations. We identify robust novel associations for 2 variants with BC for BRCA1 and 3 for BRCA2 mutation carriers, P < 10−8, at 5 loci, which are not associated with risk in the general population. They include rs60882887 at 11p11.2 where MADD, SP11 and EIF1, genes previously implicated in BC biology, are predicted as potential targets. These findings will contribute towards customising BC polygenic risk scores for BRCA1 and BRCA2 mutation carriers
