    Applicability of the Caenorhabditis elegans survival, growth and reproduction test to assess the effects of biosolids used in agriculture

    Modern high productivity agriculture has led to an impoverishment of nutrients and organic carbon in field soils, and as a consequence plants nutrition complements like fertilisers or wastes have to be used to keep high yield. These complex materials can also be a source of toxicants, depending on their origin, so there is a necessity to assess their environmental effects before field use. In this study, biosolids used in agriculture were tested as a mix with standard soil at different field dose through ecotoxicity test on the nematode Caenorhabditis elegans, endpoints for this organism being survival, growth and reproduction. In order to run these tests we first followed the standardized protocol for sediment and soil testing with C. elegans (ISO 10872, 2010). These results showed that the mixtures could be significantly different according to the type of soil (e.g. two phases system with overlaying water). Indeed, soil moistening in the standardized protocol is based on soil dry weight which means that the same amount of water is added for every soil, regardless of the water holding capacity (WHC) of the samples. Moreover, some biosolids can have a high WHC what increase humidity's differences among soil and soil mixed with biosolids when moistened as recommended by this protocol. To prevent this, improvements of the standardized protocol were developed, in which water addition is based on soil WHC instead of soil dry weight. Moreover, food volume added to the samples was lowered in order to be able to include it in the volume for soil moistening. In this context and in order to validate this new method, comparisons between standardized and optimized protocols were carried out for endpoints responses in five soils with different textures. This optimized protocol was used to assess the effect of different biosolids (limed sludge, manure ...) mixed with standard soil at different field rates and an example for a limed sludge is given. Regarding these results, improvements of the standardized protocol are adapted to assess the effects of these materials on C. elegans

    Understanding signaling cascades in melanoma

    Understanding regulatory pathways involved in melanoma development and progression has advanced significantly in recent years. It is now appreciated that melanoma is the result of complex changes in multiple signaling pathways that affect growth control, metabolism, motility and the ability to escape cell death programs. Here we review the major signaling pathways currently known to be deregulated in melanoma with an implication to its development and progression. Among these pathways are Ras, B-Raf, MEK, PTEN, phosphatidylinositol-3 kinase (PI3Ks) and Akt which are constitutively activated in a significant number of melanoma tumors, in most cases due to genomic change. Other pathways discussed in this review include the [Janus kinase/signal transducer and activator of transcription (JAK/STAT), transforming growth factor-beta pathways which are also activated in melanoma, although the underlying mechanism is not yet clear. As a paradigm for remodeled signaling pathways, melanoma also offers a unique opportunity for targeted drug development.Fil: Lopez Bergami, Pablo Roberto.     Risk Factors for Food Residue after Distal Gastrectomy and a New Effective Preparation for Endoscopy: The Water-Intake Method

    Background/Aims: Food residue is frequently ob-served in the gastric remnant after distal gastrectomy, despite adequate preparation. We devised a water-in-take method to reduce food residue in the gastric remnant by drinking large quantities of water in a short time. The aims of this study were to identify the risk factors for food residue and to study the effec-tiveness of this new method for endoscopy prepa-ration. Methods: A cohort of 708 patients who under-went distal gastrectomy for gastric cancer was re-viewed prospectively. Sixty patients with large amo-unts of food residue were randomly divided into two groups: a water-intake group (n=40) and a prolonged fasting group (n=20). Results: The incidences of a large amount of food residue were 15.7%, 5.8%, 7.5%, and 2.8 % at 3, 12, 24, and 36 months, re-spectively, after distal gastrectomy. Independent risk factors for food residue were endoscopy at 3 months, diabetes mellitus, a body mass index of 19.5, and laparoscopic surgery. The proportion of successful preparations at follow-up endoscopy was higher for the water-intake group (70%) than for the prolonged fasting group (40%, p=0.025). Conclusions: The wa-ter-intake method can be recommended as a prepara-tion for endoscopy in patients who have had repetitive food residue or risk factors after distal gastrectomy

    New perspectives for the design of sustainable bioprocesses for phosphorus recovery from waste

    Phosphate rock has long been used for the production of phosphorus based chemicals. However, considering the depletion of the reservoirs and the decrease of the quality of phosphate rocks, a potential market is now emerging for the recovery of phosphate from waste and its reuse for different applications. Notably, phosphate recovery from wastewater could be included in a circular economy approach. This review focuses on the use of microbial systems for phosphorus accumulation and recovery, by considering the actual range of analytical techniques available for the monitoring of phosphorus accumulating organisms, as well as the actual biochemical and metabolic engineering toolbox available for the optimization of bioprocesses. In this context, knowledge gathered from process, system and synthetic biology could potentially lead to innovative process design

    Identification of bZIP interaction partners of viral proteins HBZ, MEQ, BZLF1, and K-bZIP using coiled-coil arrays

    Basic-region leucine-zipper transcription factors (bZIPs) contain a segment rich in basic amino acids that can bind DNA, followed by a leucine zipper that can interact with other leucine zippers to form coiled-coil homo- or heterodimers. Several viruses encode proteins containing bZIP domains, including four that encode bZIPs lacking significant homology to any human protein. We investigated the interaction specificity of these four viral bZIPs by using coiled-coil arrays to assess self-associations as well as heterointeractions with 33 representative human bZIPs. The arrays recapitulated reported viral−human interactions and also uncovered new associations. MEQ and HBZ interacted with multiple human partners and had unique interaction profiles compared to any human bZIPs, whereas K-bZIP and BZLF1 displayed homospecificity. New interactions detected included HBZ with MAFB, MAFG, ATF2, CEBPG, and CREBZF and MEQ with NFIL3. These were confirmed in solution using circular dichroism. HBZ can heteroassociate with MAFB and MAFG in the presence of MARE-site DNA, and this interaction is dependent on the basic region of HBZ. NFIL3 and MEQ have different yet overlapping DNA-binding specificities and can form a heterocomplex with DNA. Computational design considering both affinity for MEQ and specificity with respect to other undesired bZIP-type interactions was used to generate a MEQ dimerization inhibitor. This peptide, anti-MEQ, bound MEQ both stably and specifically, as assayed using coiled-coil arrays and circular dichroism in solution. Anti-MEQ also inhibited MEQ binding to DNA.     Scientific Opinion addressing the state of the science on risk assessment of plant protection products for in-soil organisms

    Following a request from EFSA, the Panel on Plant Protection Products and their Residues developed an opinion on the science behind the risk assessment of plant protection products for in-soil organisms. The current risk assessment scheme is reviewed, taking into account new regulatory frameworks and scientific developments. Proposals are made for specific protection goals for in-soil organisms being key drivers for relevant ecosystem services in agricultural landscapes such as nutrient cycling, soil structure, pest control and biodiversity. Considering the time-scales and biological processes related to the dispersal of the majority of in-soil organisms compared to terrestrial non-target arthropods living above soil, the Panel proposes that in-soil environmental risk assessments are made at in- and off-field scale considering field boundary levels. A new testing strategy which takes into account the relevant exposure routes for in-soil organisms and the potential direct and indirect effects is proposed. In order to address species recovery and long-term impacts of PPPs, the use of population models is also proposed
