    Cosmic variance in [O/Fe] in the Galactic disk

    We examine the distribution of the [O/Fe] abundance ratio in stars across the Galactic disk using H-band spectra from the Apache Point Galactic Evolution Experiment (APOGEE). We minimize systematic errors by considering groups of stars with similar atmospheric parameters. The APOGEE measurements in the Sloan Digital Sky Survey data release 12 reveal that the square root of the star-to-star cosmic variance in the oxygen-to-iron ratio at a given metallicity is about 0.03–0.04 dex in both the thin and thick disk. This is about twice as high as the spread found for solar twins in the immediate solar neighborhood and the difference is probably associated to the wider range of galactocentric distances spanned by APOGEE stars. We quantify the uncertainties by examining the spread among stars with the same parameters in clusters; these errors are a function of effective temperature and metallicity, ranging between 0.005 dex at 4000 K and solar metallicity, to about 0.03 dex at 4500 K and [Fe/H] ≃ −0.6. We argue that measuring the spread in [O/Fe] and other abundance ratios provides strong constraints for models of Galactic chemical evolution

    Quality of life data as prognostic indicators of survival in cancer patients: an overview of the literature from 1982 to 2008

    <p>Abstract</p> <p>Background</p> <p>Health-related quality of life and survival are two important outcome measures in cancer research and practice. The aim of this paper is to examine the relationship between quality of life data and survival time in cancer patients.</p> <p>Methods</p> <p>A review was undertaken of all the full publications in the English language biomedical journals between 1982 and 2008. The search was limited to cancer, and included the combination of keywords 'quality of life', 'patient reported-outcomes' 'prognostic', 'predictor', 'predictive' and 'survival' that appeared in the titles of the publications. In addition, each study was examined to ensure that it used multivariate analysis. Purely psychological studies were excluded. A manual search was also performed to include additional papers of potential interest.</p> <p>Results</p> <p>A total of 451 citations were identified in this rapid and systematic review of the literature. Of these, 104 citations on the relationship between quality of life and survival were found to be relevant and were further examined. The findings are summarized under different headings: heterogeneous samples of cancer patients, lung cancer, breast cancer, gastro-oesophageal cancers, colorectal cancer, head and neck cancer, melanoma and other cancers. With few exceptions, the findings showed that quality of life data or some aspects of quality of life measures were significant independent predictors of survival duration. Global quality of life, functioning domains and symptom scores - such as appetite loss, fatigue and pain - were the most important indicators, individually or in combination, for predicting survival times in cancer patients after adjusting for one or more demographic and known clinical prognostic factors.</p> <p>Conclusion</p> <p>This review provides evidence for a positive relationship between quality of life data or some quality of life measures and the survival duration of cancer patients. Pre-treatment (baseline) quality of life data appeared to provide the most reliable information for helping clinicians to establish prognostic criteria for treating their cancer patients. It is recommended that future studies should use valid instruments, apply sound methodological approaches and adequate multivariate statistical analyses adjusted for socio-demographic characteristics and known clinical prognostic factors with a satisfactory validation strategy. This strategy is likely to yield more accurate and specific quality of life-related prognostic variables for specific cancers.</p

    L-Cysteine determination in embryo cell culture media using Co (II)-phthalocyanine modified disposable screen-printed electrodes

    Thiol-containing compounds such as L-cysteine have been demonstrated to play an important role in metabolism and cellular growth, acting as powerful antioxidants. Consequently, their analytical determination in biological media has received a considerable amount of attention. In this work, an electrochemical sensor for the accurate electroanalytical determination of L-cysteine is proposed, based upon a Co(II)-phthalocyanine bulk modified disposable screen-printed graphite electrode (CoPc-SPE). Cyclic (CV) and Square Wave (SWV) voltammetry experiments have demonstrated an excellent electrocatalytic activity towards the electrochemical oxidation of L-cysteine using CoPc-SPEs within optimum neutral or basic pH. Moreover, the SWV response of L-cysteine is found to exhibit a linear range of 2.6–200 ΌM, with a low limit of detection of 4 ΌM (S/N = 3) and a sensitivity of 0.78 ΌA cm− 2 ΌM− 1. Coefficient of variation for reproducibility and repeatability of L-cysteine determination using the CoPc-SPE are 3 and 0.4%, respectively. The effect of inherent interferences such as naturally occurring amino acids, cystine and cysteic acid has been also evaluated. Finally, the applicability of the L-cysteine electrochemical sensor based upon CoPc-SPEs has been successfully demonstrated for the first time for the assessment of L-cysteine in a complex embryo cell culture medium

    Phylogeny Disambiguates the Evolution of Heat-Shock cis-Regulatory Elements in Drosophila

    Heat-shock genes have a well-studied control mechanism for their expression that is mediated through cis-regulatory motifs known as heat-shock elements (HSEs). The evolution of important features of this control mechanism has not been investigated in detail, however. Here we exploit the genome sequencing of multiple Drosophila species, combined with a wealth of available information on the structure and function of HSEs in D. melanogaster, to undertake this investigation. We find that in single-copy heat shock genes, entire HSEs have evolved or disappeared 14 times, and the phylogenetic approach bounds the timing and direction of these evolutionary events in relation to speciation. In contrast, in the multi-copy gene Hsp70, the number of HSEs is nearly constant across species. HSEs evolve in size, position, and sequence within heat-shock promoters. In turn, functional significance of certain features is implicated by preservation despite this evolutionary change; these features include tail-to-tail arrangements of HSEs, gapped HSEs, and the presence or absence of entire HSEs. The variation among Drosophila species indicates that the cis-regulatory encoding of responsiveness to heat and other stresses is diverse. The broad dimensions of variation uncovered are particularly important as they suggest a substantial challenge for functional studies

    Anaplasma phagocytophilum MSP4 and HSP70 proteins are involved in interactions with host cells during pathogen infection

    Anaplasma phagocytophilum transmembrane and surface proteins play a role during infection and multiplication in host neutrophils and tick vector cells. Recently, A. phagocytophilum Major surface protein 4 (MSP4) and Heat shock protein 70 (HSP70) were shown to be localized on the bacterial membrane, with a possible role during pathogen infection in ticks. In this study, we hypothesized that A. phagocytophilum MSP4 and HSP70 have similar functions in tick-pathogen and host-pathogen interactions. To address this hypothesis, herein we characterized the role of these bacterial proteins in interaction and infection of vertebrate host cells. The results showed that A. phagocytophilum MSP4 and HSP70 are involved in host-pathogen interactions, with a role for HSP70 during pathogen infection. The analysis of the potential protective capacity of MSP4 and MSP4-HSP70 antigens in immunized sheep showed that MSP4-HSP70 was only partially protective against pathogen infection. This limited protection may be associated with several factors, including the recognition of non-protective epitopes by IgG in immunized lambs. Nevertheless, these antigens may be combined with other candidate protective antigens for the development of vaccines for the control of human and animal granulocytic anaplasmosis. Focusing on the characterization of host protective immune mechanisms and protein-protein interactions at the host-pathogen interface may lead to the discovery and design of new effective protective antigens.Peer reviewedVeterinary Pathobiolog

    The 13th Data Release of the Sloan Digital Sky Survey: First Spectroscopic Data from the SDSS-IV Survey Mapping Nearby Galaxies at Apache Point Observatory

    The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) began observations in July 2014. It pursues three core programs: APOGEE-2,MaNGA, and eBOSS. In addition, eBOSS contains two major subprograms: TDSS and SPIDERS. This paper describes the first data release from SDSS-IV, Data Release 13 (DR13), which contains new data, reanalysis of existing data sets and, like all SDSS data releases, is inclusive of previously released data. DR13 makes publicly available 1390 spatially resolved integral field unit observations of nearby galaxies from MaNGA,the first data released from this survey. It includes new observations from eBOSS, completing SEQUELS. In addition to targeting galaxies and quasars, SEQUELS also targeted variability-selected objects from TDSS and X-ray selected objects from SPIDERS. DR13 includes new reductions ofthe SDSS-III BOSS data, improving the spectrophotometric calibration and redshift classification. DR13 releases new reductions of the APOGEE-1data from SDSS-III, with abundances of elements not previously included and improved stellar parameters for dwarf stars and cooler stars. For the SDSS imaging data, DR13 provides new, more robust and precise photometric calibrations. Several value-added catalogs are being released in tandem with DR13, in particular target catalogs relevant for eBOSS, TDSS, and SPIDERS, and an updated red-clump catalog for APOGEE.This paper describes the location and format of the data now publicly available, as well as providing references to the important technical papers that describe the targeting, observing, and data reduction. The SDSS website, http://www.sdss.org, provides links to the data, tutorials and examples of data access, and extensive documentation of the reduction and analysis procedures. DR13 is the first of a scheduled set that will contain new data and analyses from the planned ~6-year operations of SDSS-IV.PostprintPeer reviewe
