    Phytochemical bioprospecting, antioxidant, antimicrobial and cytotoxicity activities of saline extract from Tithonia diversifolia (Hemsl) A. Gray leaves

    Objective: To evaluate antimicrobial and antioxidant properties of saline extract from Tithonia diversifolia leaves by phytochemical bioprospecting, and investigate its safety against animal cells. Methods: The saline extract was prepared, with NaCl (0.15 M), by constant stirring of the dried and pulverized leaves, followed by volume reduction by lyophilization. The extract was phytochemical characterized using ultra-performance liquid chromatography, and total phenol and flavonoid analysis also was performed. The antioxidant capacity was determined through DPPH· radical, the antimicrobial property was evaluated against standard bacteria and fungi, and the viability assays were performed against mice splenocytes. Results: Fifteen compounds were identified belonging to two main classes terpenoids and phenolics. The extract showed 22.185 mg GAE/g of total phenolic compounds and 3.220 mg QE/g of flavonoid. Moreover, extract showed higher antioxidant ability similar to butylated hydroxytoluene a standard molecule [(3.042±0.019) mg AAE/g and (4.12±0.10) mg AAE/g to saline extract and butylated hydroxytoluene, respectively]. The antimicrobial assays demonstrated that the extract had a significant antifungal potential against Candida species and could be used with safety against mice splenocytes, in concentrations lower than 50 μg/mL, promoting higher proliferation in these cells. Conclusions: Saline extract from Tithonia diversifolia leaves presents potential antioxidant, antifungal properties and induces immunostimulation in mice splenocytes

    Avaliação da organização e funcionamento das salas de vacina na Atenção Primária à Saúde em Montes Claros, Minas Gerais, 2015

    Resumo OBJETIVO: avaliar a qualidade da organização e do funcionamento das salas de vacina em Montes Claros, Minas Gerais, Brasil, em 2015. MÉTODOS: estudo de avaliação da qualidade das salas de vacina, tendo como base as orientações e normas técnicas do Programa Nacional de Imunizações (PNI), utilizando-se questionário adaptado do Programa de Avaliação do Instrumento de Supervisão para Sala de Vacinação (PAISSV 2.0). RESULTADOS: foram avaliadas todas as 18 salas de vacina do município; quanto ao conceito de qualidade das salas de vacina, seis foram consideradas ideais, cinco boas, quatro regulares e três insuficientes. CONCLUSÃO: a maioria das salas de vacina foi classificada como boa/ideal; no entanto, há necessidade de sistematização de educação permanente para os profissionais e de melhorias estruturais, a fim de atender às normas preconizadas pelo PNI

    Considerando o cenário complexo, heterogêneo e caótico das cidades na contemporaneidade busca-se aproximar a coexistência da estrutura urbana associada às vivências sensíveis. O grupo de pesquisa propõe o acirramento das situações de ruptura em diversas áreas, seja no enunciar das cidades pequenas; na atenção a corporalidade da criança que ocupa o espaço público; na interferência do som urbano; no processo efêmero das feiras públicas-urbanas ou no entendimento da dinâmica da linha de fronteira que separa e une países. O fio que conecta cada situação é a experiência da cartografia sensível como metodologia, o modo de acompanhar os processos e não o de quem busca respostas ou motivos pré-estabelecidos, a possibilidade de apoderar de fontes variadas além das escritas-teóricas-conceituais.Deste modo, a imersão corporal do pesquisador no território reúne as percepções mais próximas da realidade, o que viabiliza aos arquitetos, urbanistas, artistas, uma nova perspectiva de sentido e criação das cidades.Considering the complex, heterogeneous and chaotic scenario of cities in the contemporary world, we seek to approximate the coexistence of the urban structure associated with sensitive experiences. The research group proposes the intensification of situations of rupture in several areas, be it in the enunciation of small cities; attention to the corporality of the child that occupies the public space; in the interference of urban sound; in the ephemeral process of the public-urban fairs or in the understanding of the dynamics of the border line that separates and unites countries. The thread that connects each situation is the experience of sensitive mapping as a methodology, the way to follow the processes and not the one of those who seek answers or preestablished motives, the possibility of taking over varied sources besides theoreticalconceptual-writing. In this way, the researcher’s immersion in the territory brings together the closest perceptions of reality, which enables architects, urbanists, artists, a new perspective of meaning and creation of cities.     Immunological response against breast lineage cells transfected with human papillomavirus (HPV)

    Breast cancer is the most common neoplasm worldwide. Viral infections are involved with carcinogenesis, especially those caused by oncogenic Human Papillomavirus (HPV) genotypes. Despite the detection of HPV in breast carcinomas, the virus’s activity against this type of cancer remains controversial. HPV infection promotes remodeling of the host’s immune response, resulting in an immunosuppressive profile. This study assessed the individual role of HPV oncogenes in the cell line MDA-MB-231 transfected with the E5, E6, and E7 oncogenes and co-cultured with peripheral blood mononuclear cells. Immunophenotyping was conducted to evaluate immune system modulation. There was an increase in CD4+ T cell numbers when compared with non-transfected and transfected MDA-MB-231, especially in the Treg profile. Pro-inflammatory intracellular cytokines, such as IFN-γ, TNF-α, and IL-17, were impaired by transfected cells, and a decrease in the cytolytic activity of the CD8+ and CD56+ lymphocytes was observed in the presence of HPV oncogenes, mainly with E6 and E7. The E6 and E7 oncogenes decrease monocyte expression, activating the expected M1 profile. In the monocytes found, a pro-inflammatory role was observed according to the cytokines released in the supernatant. In conclusion, the MDA-MB-231 cell lineage transfected with HPV oncogenes can downregulate the number and function of lymphocytes and monocytes

    Brazilian Flora 2020: Leveraging the power of a collaborative scientific network

    International audienceThe shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiversity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxonomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world's known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world's most biodiverse countries. We further identify collection gaps and summarize future goals that extend beyond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still unequally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the country. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora

    Characterisation of microbial attack on archaeological bone

    As part of an EU funded project to investigate the factors influencing bone preservation in the archaeological record, more than 250 bones from 41 archaeological sites in five countries spanning four climatic regions were studied for diagenetic alteration. Sites were selected to cover a range of environmental conditions and archaeological contexts. Microscopic and physical (mercury intrusion porosimetry) analyses of these bones revealed that the majority (68%) had suffered microbial attack. Furthermore, significant differences were found between animal and human bone in both the state of preservation and the type of microbial attack present. These differences in preservation might result from differences in early taphonomy of the bones. © 2003 Elsevier Science Ltd. All rights reserved
