    Comparison of 25‐hydroxyvitamin D concentration in chimpanzee dried blood spots and serum

    BackgroundDried blood spots (DBS) are used in human medicine to measure total 25‐hydroxyvitamin D (25‐OHD) in the blood. However, this easy and affordable sampling technique has not been evaluated in primates to measure vitamin D concentrations.ObjectivesWe aimed to compare 25‐OHD measurements in chimpanzee serum at two different laboratories and determine the precision and accuracy of the DBS method by comparing DBS and serum results.MethodsBlood samples from 17 captive chimpanzees were collected, and 25‐OHD3 and 25‐OHD2 were measured in serum at two accredited laboratories using liquid chromatography‐tandem mass spectrometry. The same analytes were measured on DBS cards, and results were compared with that of serum. Data were assessed using the Spearman correlation, Deming regression, and Bland‐Altman analyses. ResultsThe correlation coefficient between the two measurements in serum was r s = .51 (P = .04), and the mean bias was −1.25 ± 14.83. When comparing 25‐OHD concentrations measured in DBS and serum at the same laboratory, the r s was 0.7 (P = .002), and the mean bias was 1.42 ± 14.58. Estimated intra‐assay and inter‐assay coefficients of variation for DBS results were 6% and 12.6%, respectively. ConclusionsAlthough substantial analytical variability was found in 25‐OHD measurements regardless of the sample type, the identification of both constant and proportional error and wider limits of agreement with the DBS technique makes the interpretation of DBS results challenging, especially for values close to clinical cut‐off points. The DBS and serum methods were not interchangeable, and further studies are needed to validate DBS samples for vitamin D measurements in chimpanzees

    BeppoSAX observations of the X-ray pulsar MAXI J1409-619 in low state: discovery of cyclotron resonance features

    The transient 500 s X-ray pulsar MAXI J1409-619 was discovered by the slit cameras aboard MAXI on October 17, 2010, and soon after accurately localized by Swift. We found that the source position was serendipitously observed in 2000 during BeppoSAX observations of the Galactic plane. Two sources are clearly detected in the MECS: one is consistent with the position of IGR J14043-6148 and the other one with that of MAXI J1409-619. We report on the analysis of this archival BeppoSAX/MECS observation integrated with newly analyzed observation from ASCA and a set of high-energy observations obtained from the offset fields of the BeppoSAX/PDS instrument. For the ON-source observation, the 1.8-100 keV spectrum is fit by an absorbed power law with a photon index Gamma = 0.87_{-0.19}^{+0.29}, corresponding to 2-10 and 15-100 keV unabsorbed fluxes of 2.7E-12 and 4E-11 erg/cm2/s, respectively, and a 2-10 keV luminosity of 7E+34 erg/s for a 15 kpc distance. For a PDS offset field observation, performed about one year later and showing a 15-100 keV flux of 7E-11 erg/cm2/s, we clearly pinpoint three spectral absorption features at 44, 73, and 128 keV, resolved both in the spectral fit and in the Crab ratio. We interpret these not harmonically spaced features as due to cyclotron resonances. The fundamental energy of 44 +/- 3 keV corresponds to a magnetic field strength at the neutron star surface of 3.8E12 (1+z) G, where z is the gravitational redshift.     Efficient exploration of availability models guided by failure distances

    Recently, a method to bound the steady-state availability using the failure distance concept has been proposed. In this paper we refine that method by introducing state space exploration techniques. In the methods proposed here, the state space is incrementally generated based on the contributions to the steady-state availability band of the states in the frontier of the currently generated state space. Several state space exploration algorithms are evaluated in terms of bounds quality and memory and CPU time requirements. The more efficient seems to be a waved algorithm which expands transition groups. We compare our new methods with the method based on the failure distance concept without state exploration and a method proposed by Souza e Silva and Ochoa which uses state space exploration but does not use the failure distance concept. Using typical examples we show that the methods proposed here can be significantly more efficient than any of the previous methods.Postprint (published version

    Fertilizer Adoption by Smallholders in the Brazilian Amazon:Farm-level Evidence

    Multiple constraints prevent smallholders from adopting fertilizers even with regional supply of agricultural inputs expanding and soils being weared-out. Using comprehensive farm-level data from the eastern Brazilian Amazon, we found that market proximity had a significant positive correlation with fertilizer adoption, even after controlling for liquidity, land tenure, education, experience and access to rural extension services. Nevertheless, few smallholders completely replaced nutrients from vegetation with fertilizers. Instead, we found that a hybrid system that combines nutrients from vegetation and fertilizers was approximately twice as common as exclusive fertilizer use. We suggest that the option for this diversified “nutrient portfolio” may result not only from a lack of capital or knowledge regarding return on fertilizer use, but also from the need to adapt to the economic constraints facing smallholders and minimize risk. Results indicate that a rural extension program aimed at supporting a rapid and complete replacement of ashes from vegetation by fertilizers could prove unsuccessful for Amazonian smallholders

    The practicalities of adapting UK maternity clinical information systems for observational research: Experiences of the POOL study

    Background Using routinely collected clinical data for observational research is an increasingly important method for data collection, especially when rare outcomes are being explored. The POOL study was commissioned to evaluate the safety of waterbirth in the UK using routine maternity and neonatal clinical data. This paper describes the design, rationale, set-up and pilot for this data linkage study using bespoke methods. Methods Clinical maternity information systems hold many data items of value for research purposes, but often lack specific data items required for individual studies. This study used the novel method of amending an existing clinical maternity database for the purpose of collecting additional research data fields. In combination with the extraction of existing data fields, this maximised the potential use of existing routinely collected clinical data for research purposes, whilst reducing NHS staff data collection burden. Wellbeing SoftwareÂŽ provider of the EurokingÂŽ Maternity Information System, added new study specific data fields to their information system, extracted data from participating NHS sites and transferred data for matching with the National Neonatal Research Database to ascertain outcomes for babies admitted to neonatal units. Study set-up processes were put in place for all sites. The data extraction, linkage and cleaning processes were piloted with one pre-selected NHS site. Results Twenty-six NHS sites were set-up over 27 months (January 2019 - April 2021). Twenty-four thousand maternity records were extracted from the one NHS site, pertaining to the period January 2015 to March 2019. Data field completeness for maternal and neonatal primary outcomes were mostly acceptable. Neonatal identifiers flowed to the National Neonatal Research Database for successful matching and linkage between maternity and neonatal unit records. Discussion Piloting the data extraction and linkage highlighted the need for additional governance arrangements, training at NHS sites and new processes for the study team to ensure data quality and confidentiality are upheld during the study. Amending existing NHS electronic information systems and accessing clinical data at scale, is possible, but continues to be a time consuming and a technically challenging exercise

    In this article, we highlight some critical matters in the way that an issue is framed as a problem in policymaking and the consequent means of taming that problem, in focussing on the use and implications of neuroscientific discourse of brain claims in early intervention policy and practice. We draw on three sets of analyses: of the contradictory set of motifs framing the state of ‘evidence’ of what works in intervention in the early years; of the (mis)use of neuroscientific discourse to frame deficient parenting as causing inequalities and support particular policy directions; and of the way that early years practitioners adopt brain claims to tame the problem of deficient parenting. We argue that using expedient brain claims as a framing and taming justification is entrenching gendered and classed understandings and inequalities

    Effect of a test-and-treat approach to vitamin D supplementation on risk of all cause acute respiratory tract infection and covid-19:phase 3 randomised controlled trial (CORONAVIT)

    OBJECTIVE: To determine the effect of population level implementation of a test-and-treat approach to correction of suboptimal vitamin D status (25-hydroxyvitamin D (25(OH)D) <75 nmol/L) on risk of all cause acute respiratory tract infection and covid 19. DESIGN: Phase 3 open label randomised controlled trial. SETTING: United Kingdom. PARTICIPANTS: 6200 people aged ≥16 years who were not taking vitamin D supplements at baseline. INTERVENTIONS: Offer of a postal finger prick test of blood 25(OH)D concentration with provision of a six month supply of lower dose vitamin D (800 IU/day, n=1550) or higher dose vitamin D (3200 IU/day, n=1550) to those with blood 25(OH)D concentration <75 nmol/L, compared with no offer of testing or supplementation (n=3100). Follow-up was for six months. MAIN OUTCOME MEASURES: The primary outcome was the proportion of participants with at least one swab test or doctor confirmed acute respiratory tract infection of any cause. A secondary outcome was the proportion of participants with swab test confirmed covid-19. Logistic regression was used to calculate odds ratios and associated 95% confidence intervals. The primary analysis was conducted by intention to treat. RESULTS: Of 3100 participants offered a vitamin D test, 2958 (95.4%) accepted and 2674 (86.3%) had 25(OH)D concentrations <75 nmol/L and received vitamin D supplements (n=1328 lower dose, n=1346 higher dose). Compared with 136/2949 (4.6%) participants in the no offer group, at least one acute respiratory tract infection of any cause occurred in 87/1515 (5.7%) in the lower dose group (odds ratio 1.26, 95% confidence interval 0.96 to 1.66) and 76/1515 (5.0%) in the higher dose group (1.09, 0.82 to 1.46). Compared with 78/2949 (2.6%) participants in the no offer group, 55/1515 (3.6%) developed covid-19 in the lower dose group (1.39, 0.98 to 1.97) and 45/1515 (3.0%) in the higher dose group (1.13, 0.78 to 1.63). CONCLUSIONS: Among people aged 16 years and older with a high baseline prevalence of suboptimal vitamin D status, implementation of a population level test-and-treat approach to vitamin D supplementation was not associated with a reduction in risk of all cause acute respiratory tract infection or covid-19.     Tomato: a crop species amenable to improvement by cellular and molecular methods

    Tomato is a crop plant with a relatively small DNA content per haploid genome and a well developed genetics. Plant regeneration from explants and protoplasts is feasable which led to the development of efficient transformation procedures. In view of the current data, the isolation of useful mutants at the cellular level probably will be of limited value in the genetic improvement of tomato. Protoplast fusion may lead to novel combinations of organelle and nuclear DNA (cybrids), whereas this technique also provides a means of introducing genetic information from alien species into tomato. Important developments have come from molecular approaches. Following the construction of an RFLP map, these RFLP markers can be used in tomato to tag quantitative traits bred in from related species. Both RFLP's and transposons are in the process of being used to clone desired genes for which no gene products are known. Cloned genes can be introduced and potentially improve specific properties of tomato especially those controlled by single genes. Recent results suggest that, in principle, phenotypic mutants can be created for cloned and characterized genes and will prove their value in further improving the cultivated tomato.

    A systematic review of the effectiveness of self-management interventions in people with multiple sclerosis at improving depression, anxiety and quality of life.

    BACKGROUND: Self-management interventions have become increasingly popular in the management of long-term health conditions; however, little is known about their impact on psychological well-being in people with Multiple Sclerosis (MS). PURPOSE: To examine the effectiveness of self-management interventions on improving depression, anxiety and health related quality of life in people with MS. METHOD: A structured literature search was conducted for the years 2000 to 2016. The review process followed the PRISMA guidelines, and is registered with PROSPERO (no. CRD42016033925). RESULTS: The review identified 10 RCT trials that fulfilled selection criteria and quality appraisal. Self-management interventions improved health-related quality of life in 6 out of 7 studies, with some evidence of improvement in depression and anxiety symptoms. CONCLUSION: Although the results are promising more robust evaluation is required in order to determine the effectiveness of self-management interventions on depression, anxiety and quality of life in people with MS. Evaluation of the data was impeded by a number of methodological issues including incomplete content and delivery information for the intervention and the exclusion of participants representing the disease spectrum. Recommendations are made for service development and research quality improvement

    Standalone vertex nding in the ATLAS muon spectrometer

    A dedicated reconstruction algorithm to find decay vertices in the ATLAS muon spectrometer is presented. The algorithm searches the region just upstream of or inside the muon spectrometer volume for multi-particle vertices that originate from the decay of particles with long decay paths. The performance of the algorithm is evaluated using both a sample of simulated Higgs boson events, in which the Higgs boson decays to long-lived neutral particles that in turn decay to bbar b final states, and pp collision data at √s = 7 TeV collected with the ATLAS detector at the LHC during 2011
