Currently, limited data exist about the mutational profiles of grade 1 meningiomas and patient outcome. We investigated the genetic variants present in 32 WHO grade 1 meningiomas using whole exome sequencing, and correlated gene mutational profiles with tumor cytogenetics and patient outcome. Overall, WHO grade 1 meningiomas harbored numerous and heterogeneous genetic variants, which most frequently affected the NF2 (47%) gene and to a less extent the PNMA6A (22%), TIGD1 (16%), SMO (13%), PTEN (13%), CREG2 (9%), EEF1A1 (6%), POLR2A (6%), ARID1B (3%), and FAIM3 (3%) genes. Notably, non-synonymous genetic variants of SMO and POLR2A were restricted to diploid meningiomas, whereas NF2 mutations were only found among tumors that showed -22/22q─ (with or without a complex karyotype). Based on NF2 mutations and tumor cytogenetics, four genetic profiles were defined with an impact on patient recurrence-free survival (RFS). These included (1) two good-prognosis tumor subgroups—diploid meningiomas (n=9) and isolated -22/22q─ associated with NF2 mutation (n=7)—with RFS rates at 10 y of 100%; and (2) two subgroups of poor-prognosis meningiomas—isolated -22/22q─ without NF2 mutation (n=3) and tumors with complex karyotypes (n=11)—with a RFS rate at 10 y of 48% (p=0.003).     Somatic indices and nutritional composition of the roe of the native fish Dormitator latifrons: no aplica

    Objective: To evaluate some reproductive aspects of D. latifrons and the nutritional quality of its eggs. Design/methods/approach: Eighty-two fish were randomly collected (August 2021), and their sex, length, weight, somatic indices, Fulton condition index (K), proximate composition, and amino acid and lipid composition (fatty acids) of the gonads were determined. Results: Of the total specimens collected, 62% were females; length and weight values were higher in males, but their gonadosomatic index (GSI) was lower than in females. Somatic indices did not show differences between different weight ranges. In the roe of D. latifrons, the average proximate composition was 24.3% protein and 8.5% lipids. The most abundant essential amino acids were leucine and lysine. Linoleic acid (C18:2n6) was the fatty acid with the highest concentration. Limitations/implications: It is necessary to complement the analysis of the amino acid and fatty acid profile of the roe in wild organisms to relate the changes caused by balanced feed. Findings/conclusions: This study shows that the D. latifrons roe is a good source of amino acids and PUFA.     La extensión dando respuestas en plena pandemia

    En la convocatoria 2019 de programas y proyectos de la UNLP, nuestro equipo se presentó a la misma en la modalidad programas. Ya llevamos varios años trabajando en extensión, y el equipo viene desarrollando dicho trabajo en un par de líneas, lo que hace que la experiencia se haya ido concentrando en el tiempo en líneas tales como: instalaciones edilicias en general, complemento de áreas de comunicación, fortalecimiento de organizaciones territoriales, entre otras. También se viene trabajando, ya sea en las presentaciones de los años anteriores y en las distintas actividades de extensión, en el concepto del avance en forma colectiva, es decir, siempre presentándonos en forma conjunta con otras Facultades, y por sobre todas las cosas, en una labor codo a codo con organizaciones territoriales de la barriada platense. Nuestra presentación en éste caso, se centró en la línea de fortalecimiento institucional. Hemos podido corroborar que el trabajo con algunas organizaciones a través del tiempo, ha permitido tener un panorama muy amplio de su funcionamiento, de sus fortalezas y debilidades y también, ese trabajo en conjunto nos ha dado legitimidad a la hora de plantearles algún consejo, alguna propuesta de mejora, o cualquier cuestión que haga a transmitirles algo que nos parece que podía o puede aportar a su fortalecimiento o mejorar como organización. De ahí la línea de presentación que elegimos y la institución cabecera con la cual nos presentamos en el Club Corazones de El Retiro. El club está implantado en pleno barrio El Retiro, de la ciudad de La Plata, y en contacto directo con la zona más humilde y necesitada en todo aspecto del mismo.Facultad de Ingenierí

    Treatment with tocilizumab or corticosteroids for COVID-19 patients with hyperinflammatory state: a multicentre cohort study (SAM-COVID-19)

    Objectives: The objective of this study was to estimate the association between tocilizumab or corticosteroids and the risk of intubation or death in patients with coronavirus disease 19 (COVID-19) with a hyperinflammatory state according to clinical and laboratory parameters. Methods: A cohort study was performed in 60 Spanish hospitals including 778 patients with COVID-19 and clinical and laboratory data indicative of a hyperinflammatory state. Treatment was mainly with tocilizumab, an intermediate-high dose of corticosteroids (IHDC), a pulse dose of corticosteroids (PDC), combination therapy, or no treatment. Primary outcome was intubation or death; follow-up was 21 days. Propensity score-adjusted estimations using Cox regression (logistic regression if needed) were calculated. Propensity scores were used as confounders, matching variables and for the inverse probability of treatment weights (IPTWs). Results: In all, 88, 117, 78 and 151 patients treated with tocilizumab, IHDC, PDC, and combination therapy, respectively, were compared with 344 untreated patients. The primary endpoint occurred in 10 (11.4%), 27 (23.1%), 12 (15.4%), 40 (25.6%) and 69 (21.1%), respectively. The IPTW-based hazard ratios (odds ratio for combination therapy) for the primary endpoint were 0.32 (95%CI 0.22-0.47; p < 0.001) for tocilizumab, 0.82 (0.71-1.30; p 0.82) for IHDC, 0.61 (0.43-0.86; p 0.006) for PDC, and 1.17 (0.86-1.58; p 0.30) for combination therapy. Other applications of the propensity score provided similar results, but were not significant for PDC. Tocilizumab was also associated with lower hazard of death alone in IPTW analysis (0.07; 0.02-0.17; p < 0.001). Conclusions: Tocilizumab might be useful in COVID-19 patients with a hyperinflammatory state and should be prioritized for randomized trials in this situatio

    Biodiversity recovery of Neotropical secondary forests

    Old-growth tropical forests harbor an immense diversity of tree species but are rapidly being cleared, while secondary forests that regrow on abandoned agricultural lands increase in extent. We assess how tree species richness and composition recover during secondary succession across gradients in environmental conditions and anthropogenic disturbance in an unprecedented multisite analysis for the Neotropics. Secondary forests recover remarkably fast in species richness but slowly in species composition. Secondary forests take a median time of five decades to recover the species richness of old-growth forest (80% recovery after 20 years) based on rarefaction analysis. Full recovery of species composition takes centuries (only 34% recovery after 20 years). A dual strategy that maintains both old-growth forests and species-rich secondary forests is therefore crucial for biodiversity conservation in human-modified tropical landscapes.     Labor productivity assessment of three different mechanized harvest systems in Colombian oil palm crops

    Labor shortages are increasingly problematic in rural areas worldwide and, in particular, in the oil palm sector in Colombia. Therefore, alternative methods and systems that increase labor productivity, such as using machines for collecting and lifting fresh fruit bunches (FFB), are needed. In oil palm cultivation, the most labor-intensive process is harvesting. We aimed to assess the labor productivity (in t · FFB/person/day) of three harvesting systems using mechanized lifting systems (harvest A: manual cutting, manual harvesting using meshes, carts, and livestock and lifting using a hydraulic arm; harvest B: manual cutting, manual harvest using a tractor, and lifting using a side-turning container; and harvest C: manual cutting, mechanized harvest using a tractor-grabber, and lifting using a side-turning container). Two bunch density scenarios were considered. Data were collected from adult palm crops in three oil palm plantations in the Eastern Plains of Colombia, and similar crop yields were obtained. Labor productivity was 1.20–2.53 t · FFB/person/day depending on the FFB density and the harvest system. Finally, the cost per ton harvested was 8.9 8.9– 16.6 per ton, being lower in the systems that demanded fewer personnel. The highest labor productivity and the lowest cost per ton harvested were obtained for harvest C

    EMFi-Based ultrasonic sensory array for 3D localization of reflectors using positioning algorithms

    This paper describes a first prototype of an airborne ultrasonic array that combines pulse-compression techniques with positioning algorithms in order to achieve accurate determination of the position of the reflectors placed in front of the array. The new sensory system is based on an array with four independently controlled emitters and a receiver. The emitters are made of ferroelectrets as active material, leveraging their features to produce broadband air-coupled ultrasonic transducers at low cost. The ultrasonic emissions are encoded with an orthogonal Kasami sequence for each transducer, making possible to transmit simultaneously from each one. Furthermore, one of the main novelties of the proposal is that reflectors are located by means of a spherical trilateration algorithm that uses the time-of-flight of the ultrasonic signals from each transmitter to the receiver, bounced back by the reflector.Fil: Jiménez Martín, Ana. El mecanismo de transmisión de SARS-CoV-2 por vía aérea en distancias cortas y medias (hasta varios metros) ha de considerarse, sin que se pueda decir que tenga un impacto superior o inferior a otras vías como la del contacto con boca, nariz y ojos. Por tanto, se requieren medidas preventivas para mitigar la transmisión aérea de corto a mediano alcance.N

    Na+ controls hypoxic signalling by the mitochondrial respiratory chain

    All metazoans depend on the consumption of O2 by the mitochondrial oxidative phosphorylation system (OXPHOS) to produce energy. In addition, the OXPHOS uses O2 to produce reactive oxygen species that can drive cell adaptations1-4, a phenomenon that occurs in hypoxia4-8 and whose precise mechanism remains unknown. Ca2+ is the best known ion that acts as a second messenger9, yet the role ascribed to Na+ is to serve as a mere mediator of membrane potential10. Here we show that Na+ acts as a second messenger that regulates OXPHOS function and the production of reactive oxygen species by modulating the fluidity of the inner mitochondrial membrane. A conformational shift in mitochondrial complex I during acute hypoxia11 drives acidification of the matrix and the release of free Ca2+ from calcium phosphate (CaP) precipitates. The concomitant activation of the mitochondrial Na+/Ca2+ exchanger promotes the import of Na+ into the matrix. Na+ interacts with phospholipids, reducing inner mitochondrial membrane fluidity and the mobility of free ubiquinone between complex II and complex III, but not inside supercomplexes. As a consequence, superoxide is produced at complex III. The inhibition of Na+ import through the Na+/Ca2+ exchanger is sufficient to block this pathway, preventing adaptation to hypoxia. These results reveal that Na+ controls OXPHOS function and redox signalling through an unexpected interaction with phospholipids, with profound consequences for cellular metabolism
