Sleep Problems in Children With Autism Spectrum Disorder
The purpose of the current study was to examine the effect of restricted and repetitive behaviors (RRBs) on the relationship between sleep problems and externalizing behaviors in young children with autism spectrum disorder (ASD). Sixty-six participants with ASD and a mean age of 4 years and their parents participated in the study. Parents reported on their child’s sleep, ASD symptoms, and externalizing behavior during a one-time lab visit. Bivariate correlations and a mediation analysis were conducted to assess the associations between sleep problems, externalizing behavior, and RRB. Results suggested that children with higher scores in measures of RRB had higher scores in sleep problems and externalizing behavior. Results also suggested that RRB partially mediated the relationship between sleep problems and externalizing behavior. Implications for the early identification of sleep and behavior problems, as well as ASD symptoms, are discussed

Concomitant medication use and clinical outcome of repetitive Transcranial Magnetic Stimulation (rTMS) treatment of Major Depressive Disorder.
BackgroundRepetitive Transcranial Magnetic Stimulation (rTMS) is commonly administered to Major Depressive Disorder (MDD) patients taking psychotropic medications, yet the effects on treatment outcomes remain unknown. We explored how concomitant medication use relates to clinical response to a standard course of rTMS.MethodsMedications were tabulated for 181 MDD patients who underwent a six-week rTMS treatment course. All patients received 10 Hz rTMS administered to left dorsolateral prefrontal cortex (DLPFC), with 1 Hz administered to right DLPFC in patients with inadequate response to and/or intolerance of left-sided stimulation. Primary outcomes were change in Inventory of Depressive Symptomatology Self Report (IDS-SR30) total score after 2, 4, and 6 weeks.ResultsUse of benzodiazepines was associated with less improvement at week 2, whereas use of psychostimulants was associated with greater improvement at week 2 and across 6 weeks. These effects were significant controlling for baseline variables including age, overall symptom severity, and severity of anxiety symptoms. Response rates at week 6 were lower in benzodiazepine users versus non-users (16.4% vs. 35.5%, p = 0.008), and higher in psychostimulant users versus non-users (39.2% vs. 22.0%, p = 0.02).ConclusionsConcomitant medication use may impact rTMS treatment outcome. While the differences reported here could be considered clinically significant, results were not corrected for multiple comparisons and findings should be replicated before clinicians incorporate the evidence into clinical practice. Prospective, hypothesis-based treatment studies will aid in determining causal relationships between medication treatments and outcome
Substantial regional variation in substitution rates in the human genome: importance of GC content, gene density and telomere-specific effects
This study presents the first global, 1 Mbp level analysis of patterns of
nucleotide substitutions along the human lineage. The study is based on the
analysis of a large amount of repetitive elements deposited into the human
genome since the mammalian radiation, yielding a number of results that would
have been difficult to obtain using the more conventional comparative method of
analysis. This analysis revealed substantial and consistent variability of
rates of substitution, with the variability ranging up to 2-fold among
different regions. The rates of substitutions of C or G nucleotides with A or T
nucleotides vary much more sharply than the reverse rates suggesting that much
of that variation is due to differences in mutation rates rather than in the
probabilities of fixation of C/G vs. A/T nucleotides across the genome. For all
types of substitution we observe substantially more hotspots than coldspots,
with hotspots showing substantial clustering over tens of Mbp's. Our analysis
revealed that GC-content of surrounding sequences is the best predictor of the
rates of substitution. The pattern of substitution appears very different near
telomeres compared to the rest of the genome and cannot be explained by the
genome-wide correlations of the substitution rates with GC content or exon
density. The telomere pattern of substitution is consistent with natural
selection or biased gene conversion acting to increase the GC-content of the
sequences that are within 10-15 Mbp away from the telomere.Comment: 35 pages, 6 figure
Tensile and Compressive Constitutive Response of 316 Stainless Steel at Elevated Temperatures
Creep rate in compression is lower by factors of 2 to 10 than in tension if the microstructure of the two specimens is the same and are tested at equal temperatures and equal but opposite stresses. Such behavior is characteristic for monotonic creep and conditions involving cyclic creep. In the latter case creep rate in both tension and compression progressively increases from cycle to cycle, rendering questionable the possibility of expressing a time stabilized constitutive relationship. The difference in creep rates in tension and compression is considerably reduced if the tension specimen is first subjected to cycles of tensile creep (reversed by compressive plasticity), while the compression specimen is first subjected to cycles of compressive creep (reversed by tensile plasticity). In both cases, the test temperature is the same and the stresses are equal and opposite. Such reduction is a reflection of differences in microstructure of the specimens resulting from different prior mechanical history
Specialization and Variety in Repetitive Tasks: Evidence from a Japanese Bank
Sustaining operational productivity in the completion of repetitive tasks is critical to many organizations' success. Yet research points to two different work-design related strategies for accomplishing this goal: specialization to capture the benefits of repetition or variety to keep workers motivated and allow them to learn. In this paper, we investigate how these two strategies may bring different benefits within the same day and across days. Additionally, we examine the impact of these strategies on both worker productivity and workers' likelihood of staying at a firm. For our empirical analyses, we use two and a half years of transaction data from a Japanese bank's home loan application processing line. We find that over the course of a single day, specialization, as compared to variety, is related to improved worker productivity. However, when we examine workers' experience across days we find that variety, or working on different tasks, helps improve worker productivity. We also find that workers with higher variety are more likely to stay at the firm. This is an open-access article distributed under the terms of the Creative Commons Attribution License (CC BY).Background: The approximate 5:1 male to female ratio in clinical detection of Autism Spectrum Disorder (ASD) prevents research from characterizing the female phenotype. Current open access repositories [such as those in the Autism Brain Imaging Data Exchange (ABIDE I-II)] contain large numbers of females to help begin providing a new characterization of females on the autistic spectrum. Here we introduce new methods to integrate data in a scale-free manner from continuous biophysical rhythms of the nervous systems and discrete (ordinal) observational scores. Methods: New data-types derived from image-based involuntary head motions and personalized statistical platform were combined with a data-driven approach to unveil sub-groups within the female cohort. Further, to help refine the clinical DSM-based ASD vs. Asperger's Syndrome (AS) criteria, distributional analyses of ordinal score data from Autism Diagnostic Observation Schedule (ADOS)-based criteria were used on both the female and male phenotypes. Results: Separate clusters were automatically uncovered in the female cohort corresponding to differential levels of severity. Specifically, the AS-subgroup emerged as the most severely affected with an excess level of noise and randomness in the involuntary head micro-movements. Extending the methods to characterize males of ABIDE revealed ASD-males to be more affected than AS-males. A thorough study of ADOS-2 and ADOS-G scores provided confounding results regarding the ASD vs. AS male comparison, whereby the ADOS-2 rendered the AS-phenotype worse off than the ASD-phenotype, while ADOS-G flipped the results. Females with AS scored higher on severity than ASD-females in all ADOS test versions and their scores provided evidence for significantly higher severity than males. However, the statistical landscapes underlying female and male scores appeared disparate. As such, further interpretation of the ADOS data seems problematic, rather suggesting the critical need to develop an entirely new metric to measure social behavior in females. Conclusions: According to the outcome of objective, data-driven analyses and subjective clinical observation, these results support the proposition that the female phenotype is different. Consequently the “social behavioral male ruler” will continue to mask the female autistic phenotype. It is our proposition that new observational behavioral tests ought to contain normative scales, be statistically sound and combined with objective data-driven approaches to better characterize the females across the human lifespan.Peer reviewe
Optimization of the leak conductance in the squid giant axon
We report on a theoretical study showing that the leak conductance density,
\GL, in the squid giant axon appears to be optimal for the action potential
firing frequency. More precisely, the standard assumption that the leak current
is composed of chloride ions leads to the result that the experimental value
for \GL is very close to the optimal value in the Hodgkin-Huxley model which
minimizes the absolute refractory period of the action potential, thereby
maximizing the maximum firing frequency under stimulation by sharp, brief input
current spikes to one end of the axon. The measured value of \GL also appears
to be close to optimal for the frequency of repetitive firing caused by a
constant current input to one end of the axon, especially when temperature
variations are taken into account. If, by contrast, the leak current is assumed
to be composed of separate voltage-independent sodium and potassium currents,

Regional gray matter volumetric changes in autism associated with social and repetitive behavior symptoms.
BackgroundAlthough differences in brain anatomy in autism have been difficult to replicate using manual tracing methods, automated whole brain analyses have begun to find consistent differences in regions of the brain associated with the social cognitive processes that are often impaired in autism. We attempted to replicate these whole brain studies and to correlate regional volume changes with several autism symptom measures.MethodsWe performed MRI scans on 24 individuals diagnosed with DSM-IV autistic disorder and compared those to scans from 23 healthy comparison subjects matched on age. All participants were male. Whole brain, voxel-wise analyses of regional gray matter volume were conducted using voxel-based morphometry (VBM).ResultsControlling for age and total gray matter volume, the volumes of the medial frontal gyri, left pre-central gyrus, right post-central gyrus, right fusiform gyrus, caudate nuclei and the left hippocampus were larger in the autism group relative to controls. Regions exhibiting smaller volumes in the autism group were observed exclusively in the cerebellum. Significant partial correlations were found between the volumes of the caudate nuclei, multiple frontal and temporal regions, the cerebellum and a measure of repetitive behaviors, controlling for total gray matter volume. Social and communication deficits in autism were also associated with caudate, cerebellar, and precuneus volumes, as well as with frontal and temporal lobe regional volumes.ConclusionGray matter enlargement was observed in areas that have been functionally identified as important in social-cognitive processes, such as the medial frontal gyri, sensorimotor cortex and middle temporal gyrus. Additionally, we have shown that VBM is sensitive to associations between social and repetitive behaviors and regional brain volumes in autism
The prodrome of autism: early behavioral and biological signs, regression, peri- and post-natal development and genetics
Autism is one of the most heritable neurodevelopmental conditions and has an early onset, with symptoms being required to be present in the first 3 years of life in order to meet criteria for the ‘core’ disorder in the classification systems. As such, the focus on identifying a prodrome over the past 20 years has been on pre-clinical signs or indicators that will be present very early in life, certainly in infancy. A number of novel lines of investigation have been used to this end, including retrospective coding of home videos, prospective population screening and ‘high risk’ sibling studies; as well as the investigation of pre- and peri-natal, brain developmental and other biological factors. Whilst no single prodromal sign is expected to be present in all cases, a picture is emerging of indicative prodromal signs in infancy and initial studies are being undertaken to attempt to ameliorate the early presentation and even ‘prevent’ emergence of the full syndrome
