    Avoiding Another Step in a Series of Unfortunate Legal Events: A Consideration of Black Life Under American Law from 1619 to 1972 and a Challenge to Prevailing Notions of Legally Based Reparations

    The growing body of literature on reparations consists primarily of articles showing that black reparations are consistent with various legal theories, promote racial justice, or further broader societal goals like eliminating poverty and promoting education. This article takes the distinct position of challenging reparations supporters to justify their confidence in the legal system to deliver meaningful reparations for slavery and segregation in light of the historic use of law as a means of instantiating white racial supremacy and the prospective individualistic approach to race adopted by contemporary judges and legislators. The article also challenges those who oppose reparations based on its supposed unfairness to contemporary citizens to explain how their position differs from that of past generations who opposed reparations and related legal efforts to redress racial injustices as unfair at that time. To support the challenge to reparations commentators, the article examines the historical framework of blacks’ relationship to the law through legislation and court rulings from 1619–1963. The article closes by presenting an alternative approach to reparations focused on building and strengthening black political, economic, and educational institutions

    Factors that Influence Mass Incarceration among African-Americans

    poster abstractMass incarceration refers to the high rate of imprisonment among a certain racial group. The problem of mass incarceration is common among African-Americans. The main objective of this study is to identify the major factors that lead to the high rate of incarceration among African-Americans. For years, the issue of mass incarceration has been a subject of controversy. While some believe mass incarceration is a subtle form of racism against African-Americans and a posteffect of slavery, some others view it as an adaptation to the demands of the society. Due to these different assertions on the issue of mass incarceration and also the impact of mass incarceration on the society, it is imperative to conduct research on the causes of mass incarceration prevalent among African-Americans. My primary method of research was through analyzing statistical information while referring to scholarly articles and literature. This study aims at examining the factors that have led to mass incarceration with the hope that it will help minimize the number of African-Americans in incarceration. The goal of this research is to influence subsequent decisions and policies of the key players in the American Criminal Justice System on issues concerning incarceration of African Americans

    Evaluation of an epigenetic assay for predicting repeat prostate biopsy outcome in African American men

    OBJECTIVE: To evaluate an epigenetic assay performed on tissue from negative prostate biopsies in a group of African American (AA) men undergoing repeat biopsy, and to compare accuracy for predicting repeat biopsy outcome to prior studies conducted in predominantly Caucasian populations. MATERIALS AND METHODS: The study population consisted of 211 AA men from 7 urology centers across the United States; all of whom were undergoing 12-core transrectal ultrasound-guided repeat biopsy within 30 months from a negative index biopsy. All biopsy cores from the negative index biopsy were profiled for the epigenetic biomarkers GSTP1, APC, and RASSF1 using ConfirmMDx for Prostate Cancer (MDxHealth, Irvine, CA). RESULTS: Upon repeat biopsy, 130 of 211 subjects (62%) had no prostate cancer (PCa) detected and 81 of 211 (38%) were diagnosed with PCa. Of the subjects with PCa, 54 (67%) were diagnosed with Gleason score (GS) = 7 disease. For detection of PCa at repeat biopsy, ConfirmMDx sensitivity was 74.1% and specificity was 60.0%, equivalent to prior studies (P = .235 and .697, respectively). For detection of GS >= 7 PCa, sensitivity was 78% and specificity was 53%. The negative predictive values for detection of all PCa and GS >= 7 PCa were 78.8% and 94.2%, respectively. CONCLUSION: In this group of AA men, we successfully validated an epigenetic assay to assess the need for repeat biopsy. Results were consistent with previous studies from predominantly Caucasian populations. Therefore, the ConfirmMDx assay is a useful tool for risk stratification of AA men who had an initial negative biopsy

    Oxidation mechanisms occurring in wines

    The present review aims to show the state of the art on the oxidation mechanisms occurring in wines, as well as the methods to monitor, classify and diagnose wine oxidation. Wine oxidation can be divided in enzymatic oxidation and non-enzymatic oxidation. Enzymatic oxidation almost entirely occurs in grape must and is largely correlated with the content of hydroxycinnamates, such as caffeoyltartaric acid and para-coumaroyltartaric acid, and flavan-3-ols. Non-enzymatic oxidation, also called chemical oxidation of wine, prevails in fermented wine and begin by the oxidation of polyphenols containing a catechol or a galloyl group. These phenolic. reactions, both enzymatic and non-enzymatic, result in by-products named quinones. However, in non-enzymatic oxidation, oxygen does not react directly with phenolic compounds. The limitation on the reactivity of triplet oxygen is overcome by the stepwise addition of a single electron, which can be provided by reduced transition metal ions, essentially iron(II) and copper(I). The sequential electron transfer leads to the formation of hydroperoxide radical (HOO center dot), hydrogen peroxide (H2O2), and hydroxyl radical (HO center dot). The later radical will oxidize almost any organic molecule found in wine and will react with the first species it encounters, depending on their concentration. Sulfur dioxide (SO2) and ascorbic acid, when added to wine, are able to reduce the quinones. Alternative options have been assessed for the prevention of oxidation during wine storage; nevertheless, these are not fully understood or commonly accepted. During aging, aldehydes are important intermediates in the chemical transformations occurring in wines, leading to color and flavor changes. In the same way, a range of off-flavors can be formed from wine oxidation. At low concentrations these flavors may add to the complexity of a wine, but as these increase they begin to detract from wine quality. In addition to the major chemical browning involving wine phenols, the main oxidation reactions occurring during grape juice heating or storage are caramelization and Maillard reaction, which are temperature dependent. Different methods have been proposed in the literature, addressing the complexity and multi-scale related with the oxidation process, to attempt the quantification of antioxidant activity in wines. These methods can be broadly divided in: i) methods based on chemical reactions and ii) methods based on the chemical-physical properties of antioxidants

    The minimal kinome of Giardia lamblia illuminates early kinase evolution and unique parasite biology

    Background: The major human intestinal pathogen Giardia lamblia is a very early branching eukaryote with a minimal genome of broad evolutionary and biological interest. Results: To explore early kinase evolution and regulation of Giardia biology, we cataloged the kinomes of three sequenced strains. Comparison with published kinomes and those of the excavates Trichomonas vaginalis and Leishmania major shows that Giardia's 80 core kinases constitute the smallest known core kinome of any eukaryote that can be grown in pure culture, reflecting both its early origin and secondary gene loss. Kinase losses in DNA repair, mitochondrial function, transcription, splicing, and stress response reflect this reduced genome, while the presence of other kinases helps define the kinome of the last common eukaryotic ancestor. Immunofluorescence analysis shows abundant phospho-staining in trophozoites, with phosphotyrosine abundant in the nuclei and phosphothreonine and phosphoserine in distinct cytoskeletal organelles. The Nek kinase family has been massively expanded, accounting for 198 of the 278 protein kinases in Giardia. Most Neks are catalytically inactive, have very divergent sequences and undergo extensive duplication and loss between strains. Many Neks are highly induced during development. We localized four catalytically active Neks to distinct parts of the cytoskeleton and one inactive Nek to the cytoplasm. Conclusions: The reduced kinome of Giardia sheds new light on early kinase evolution, and its highly divergent sequences add to the definition of individual kinase families as well as offering specific drug targets. Giardia's massive Nek expansion may reflect its distinctive lifestyle, biphasic life cycle and complex cytoskeleton

    Effects of acute tryptophan depletion on central processing of CT-targeted and discriminatory touch in humans.

    C-tactile afferents (CTs) are slowly conducting nerve fibres, present only in hairy skin. They are optimally activated by slow, gentle stroking touch, such as those experienced during a caress. CT-stimulation activates affective processing brain regions, alluding to their role in affective touch perception. We tested a theory that CT-activating touch engages the pro-social functions of serotonin, by determining whether reducing serotonin, through acute tryptophan depletion, diminishes subjective pleasantness and affective brain responses to gentle touch. A tryptophan depleting amino acid drink was administered to 16 healthy females, with a further 14 receiving a control drink. After 4 hours, participants underwent an fMRI scan, during which time CT-innervated forearm skin and CT non-innervated finger skin was stroked with 3 brushes of differing texture, at CT-optimal force and velocity. Pleasantness ratings were obtained post-scanning. The control group showed a greater response in ipsilateral orbitofrontal cortex to CT-activating forearm touch compared to touch to the finger where CTs are absent. This differential response was not present in the tryptophan depleted group. This interaction effect was significant. Additionally, control participants showed a differential primary somatosensory cortex response to brush texture applied to the finger, a purely discriminatory touch response, which was not observed in the tryptophan depleted group. This interaction effect was also significant. Pleasantness ratings were comparable across treatment groups. These results implicate serotonin in the differentiation between CT-activating and purely discriminatory touch responses. Such effects could contribute to some of the social abnormalities seen in psychiatric disorders associated with abnormal serotonin function. This article is protected by copyright.
