    Viscosity of R134a, R32, and R125 at saturation

    This paper reports the results of the measurement of the viscosity of R134a close to the saturation line in the vapor phase. The new measurements were carried out in a vibrating-wire viscometer specially constructed for the purpose, and the results have an accuracy of ±2%. In addition, the opportunity is taken to present a reevaluation of earlier measurements along the saturation line of the viscosity of R32 and R125. Improved equations of state for these fluids are now available and can be employed to generate improved values for the viscosity

    Vertical flux of fatty acids in the North Atlantic Ocean

    The quantitative and qualitative composition of fatty acids in particulate material collected in traps deployed during 98 days at 389, 988, 3755 and 5068 m depths in the equatorial North Atlantic was determined. The fatty acid composition indicates a predominantly marine source (14:0, 16:0, 16:1, 18:0, 18:1, 20:5, 20:4, 22:6, 22:5) with possibly a minor terrigenous component in the bathypelagic traps. The vertical fluxes of fatty acids and lipids decrease rapidly with depth. The rate of net loss of carboxylic acids increases with number of double bonds and decreases with number of carbon atoms. Iso- and anteiso- as well as some monoenoic fatty acids are more persistent, probably due to enhanced microbial synthesis during settling which counteracts degradation

    Viscosity measurements of liquid toluene at low temperatures using a dual vibrating-wire technique

    A recently developed dual vibrating-wire technique has been used to perform viscosity measurements of liquid toluene in the temperature range 213 K ≤ T ≤ 298 K, and at pressures up to approximately 20 MPa. The results were obtained by operating the vibrating-wire sensor in both forced and free decay modes. The estimated precision of the viscosity measurements, in either mode of operation, is ±0.5%, for temperatures above or equal to 273 K, increasing with decreasing temperature up to ±1% at 213 K. The corresponding overall uncertainty is estimated to be within ±1% and ±1.5%, respectively

    An industrial reference fluid for moderately high viscosity

    In industrial practice, there is a demand for a reference standard for viscosity that is established for a readily available fluid to simplify the calibration of industrial viscometers for moderately high viscosities [(50 to 125) mPa · s]. Diisodecyl phthalate (DIDP) has been suggested as that reference fluid, and a number of studies of its properties have been carried out in several laboratories throughout the world, within the auspices of a project coordinated by the International Association for Transport Properties. That project has now progressed to the point where it is possible to collate the results of studies of the viscosity of the fluid by a number of different techniques, so as to lead to a proposed standard reference value which will be included in the paper. To support this recommended value, the various measurements conducted have been critically reviewed, and the sample purity and other factors affecting the viscosity have been studied. Density and surface tension measurements have also been performed. This paper does not describe the individual viscosity determinations carried out in independent laboratories because these are the subject of individual publications, but it does describe the ancillary studies conducted and their relevance to the viscosity standard. In addition, the paper contains recommended values for the viscosity of liquid DIDP. The samples of DIDP to which the recommended values refer are isomeric mixtures available commercially from certain suppliers, with a minimum purity by gas chromatography of 99.8 %. The recommended values result from a critical examination of all the measurements conducted to date and are supported by careful arguments dealing with the likely effects of the isomeric content of the sample as well as of other impurities. The proposed reference standard is intended particularly to serve an industrial need for a readily available calibration material with a viscosity close to that required in practical situations. To that end, the recommended value has an overall relative uncertainty of approximately 1 %. It is therefore not intended to supersede for the reference value for the viscosity of water at 20 °C, which is known much more accurately, but rather to complement it

    Selective preservation of organic matter in marine environments; processes and impact on the sedimentary record

The definitive version was published in Biogeosciences 7 (2010): 483-511, doi: 10.5194/bg-7-483-2010The present paper is the result of a workshop sponsored by the DFG Research Center/Cluster of Excellence MARUM "The Ocean in the Earth System", the International Graduate College EUROPROX, and the Alfred Wegener Institute for Polar and Marine Research. The workshop brought together specialists on organic matter degradation and on proxy-based environmental reconstruction. The paper deals with the main theme of the workshop, understanding the impact of selective degradation/preservation of organic matter (OM) in marine sediments on the interpretation of the fossil record. Special attention is paid to (A) the influence of the molecular composition of OM in relation to the biological and physical depositional environment, including new methods for determining complex organic biomolecules, (B) the impact of selective OM preservation on the interpretation of proxies for marine palaeoceanographic and palaeoclimatic reconstruction, and (C) past marine productivity and selective preservation in sediments. It appears that most of the factors influencing OM preservation have been identified, but many of the mechanisms by which they operate are partly, or even fragmentarily, understood. Some factors have not even been taken carefully into consideration. This incomplete understanding of OM breakdown hampers proper assessment of the present and past carbon cycle as well as the interpretation of OM based proxies and proxies affected by OM breakdown. To arrive at better proxy-based reconstructions "deformation functions" are needed, taking into account the transport and diagenesis-related molecular and atomic modifications following proxy formation. Some emerging proxies for OM degradation may shed light on such deformation functions. The use of palynomorph concentrations and selective changes in assemblage composition as models for production and preservation of OM may correct for bias due to selective degradation. Such quantitative assessment of OM degradation may lead to more accurate reconstruction of past productivity and bottom water oxygenation. Given the cost and effort associated with programs to recover sediment cores for paleoclimatological studies, as well as with generating proxy records, it would seem wise to develop a detailed sedimentological and diagenetic context for interpretation of these records.     The Tumor Suppressor Gene Brca1 Is Required for Embryonic Cellular Proliferation in the Mouse

    AbstractMutations of the BRCA1 gene in humans are associated with predisposition to breast and ovarian cancers. We show here that Brca1+/− mice are normal and fertile and lack tumors by age eleven months. Homozygous Brca15-6 mutant mice die before day 7.5 of embryogenesis. Mutant embryos are poorly developed, with no evidence of mesoderm formation. The extraembryonic region is abnormal, but aggregation with wild-type tetraploid embryos does not rescue the lethality. In vivo, mutant embryos do not exhibit increased apoptosis but show reduced cell proliferation accompanied by decreased expression of cyclin E and mdm-2, a regulator of p53 activity. The expression of cyclin-dependent kinase inhibitor p21 is dramatically increased in the mutant embryos. Buttressing these in vivo observations is the fact that mutant blastocyst growth is grossly impaired in vitro. Thus, the death of Brca15-6 mutant embryos prior to gastrulation may be due to a failure of the proliferative burst required for the development of the different germ layers

    In response to ionizing radiation (IR), the tumor suppressor p53 is stabilized and promotes either cell cycle arrest or apoptosis. Chk2 activated by IR contributes to this stabilization, possibly by direct phosphorylation. Like p53, Chk2 is mutated in patients with Li-Fraumeni syndrome. Since the ataxia telangiectasia mutated (ATM) gene is required for IR-induced activation of Chk2, it has been assumed that ATM and Chk2 act in a linear pathway leading to p53 activation. To clarify the role of Chk2 in tumorigenesis, we generated gene-targeted Chk2-deficient mice. Unlike ATM-/- and p53-/- mice, Chk2-/- mice do not spontaneously develop tumors, although Chk2 does suppress 7,12-dimethylbenzanthracene-induced skin tumors. Tissues from Chk2-/- mice, including those from the thymus, central nervous system, fibroblasts, epidermis, and hair follicles, show significant defects in IR-induced apoptosis or impaired G1/S arrest. Quantitative comparison of the G1/S checkpoint, apoptosis, and expression of p53 proteins in Chk2-/- versus ATM-/- thymocytes suggested that Chk2 can regulate p53-dependent apoptosis in an ATM-independent manner. IR-induced apoptosis was restored in Chk2-/- thymocytes by reintroduction of the wild-type Chk2 gene but not by a Chk2 gene in which the sites phosphorylated by ATM and ataxia telangiectasia and rad3+ related (ATR) were mutated to alanine. ATR may thus selectively contribute to p53-mediated apoptosis. These data indicate that distinct pathways regulate the activation of p53 leading to cell cycle arrest or apoptosis

    Diagnostic Tests and their Application in the Management of Soil- and Water-borne Oomycete Pathogen Species

    Oomycete diseases cause significant losses across a broad range of crop and aquaculture commodities worldwide. These losses can be greatly reduced by disease management practices steered by accurate and early diagnoses of pathogen presence. Determinations of disease potential can help guide optimal crop rotation regimes, varietal selections, targeted control measures, harvest timings and crop post-harvest handling. Pathogen detection prior to infection can also reduce the incidence of disease epidemics. Classical methods for the isolation of oomycete pathogens are normally deployed only after disease symptom appearance. These processes are often-time consuming, relying on culturing the putative pathogen(s) and the availability of expert taxonomic skills for accurate identification; a situation that frequently results in either delayed application, or routine ‘blanket’ over-application of control measures. Increasing concerns about pesticides in the environment and the food chain, removal or restriction of their usage combined with rising costs have focussed interest in the development and improvement of disease management systems. To be effective, these require timely, accurate and preferably quantitatve diagnoses. A wide range of rapid diagnostic tools, from point of care immunodiagnostic kits to next generation nucleotide sequencing have potential application in oomycete disease management. Here we review currently-available as well as promising new technologies in the context of commercial agricultural production systems, considering the impacts of specific biotic and abiotic and other important factors such as speed and ease of access to information and cost effectivenes

    Methanotropic microbial communities associated with bubble plumes above gas seeps in the Black Sea

    Bubbles evolving from active gas seeps can be traced by hydroacoustic imaging up to 1000 m high in the Black Sea water column. Although methane concentrations are not distinguishable between the water column above the deep seep and reference sites, atmospheric noble gas measurements clearly show the constant input of gases (mainly methane) via seepage into the Black Sea. Archaea (ANME-1, ANME-2) and methanotrophic bacteria detected with specific 16S rRNA-targeted oligonucleotide probes are related to active gas seeps in the oxic and anoxic water column. It is suggested that methane seeps have a much greater influence on the Black Sea methane budget than previously acknowledged and that ANME-1 and ANME-2 are injected via gas bubbles from the sediment into the anoxic water column mediating methane oxidation. Our results show further that only minor amounts of methane evolving from Black Sea gas seeps reach the atmosphere due to the very effective microbial barrier. Hence only major thermodynamically and/or tectonically triggered gas hydrate dissociation has the potential to induce rapid climate changes as suggested by the “clathrate gun hypothesis.
