    Study on Optimized Elman Neural Network Classification Algorithm Based on PLS and CA

    High-dimensional large sample data sets, between feature variables and between samples, may cause some correlative or repetitive factors, occupy lots of storage space, and consume much computing time. Using the Elman neural network to deal with them, too many inputs will influence the operating efficiency and recognition accuracy; too many simultaneous training samples, as well as being not able to get precise neural network model, also restrict the recognition accuracy. Aiming at these series of problems, we introduce the partial least squares (PLS) and cluster analysis (CA) into Elman neural network algorithm, by the PLS for dimension reduction which can eliminate the correlative and repetitive factors of the features. Using CA eliminates the correlative and repetitive factors of the sample. If some subclass becomes small sample, with high-dimensional feature and fewer numbers, PLS shows a unique advantage. Each subclass is regarded as one training sample to train the different precise neural network models. Then simulation samples are discriminated and classified into different subclasses, using the corresponding neural network to recognize it. An optimized Elman neural network classification algorithm based on PLS and CA (PLS-CA-Elman algorithm) is established. The new algorithm aims at improving the operating efficiency and recognition accuracy. By the case analysis, the new algorithm has unique superiority, worthy of further promotion

    A 3D network of nanochannels for possible ion and molecule transit in mineralizing bone and cartilage

    During crucial growth stages of vertebrate long bones, calcified cartilage beneath the growth plate is anchored to bone by a third mineralized component, the cement line. Proper skeletal development is contingent on the interplay of these three constituents, yet their mineralization processes and structural interactions are incompletely understood, in part from limited knowledge of their meso- and nanoscale features. Herein, focused ion beam-scanning electron microscopy (FIB-SEM) with serial surface imaging is applied to examine the cartilage–bone interface of mouse femoral heads at an unprecedented scale: FIB-SEM provides 3D, nanometer resolution of structural details for volumes encompassing metaphyseal calcified cartilage, bone, and the intervening cement line. A novel and complex structural network is revealed, comprising densely packed nanochannels smaller than bone canaliculi (≈10–50 nm diameter) within the calcified cartilage and bone extracellular matrices, but absent in the cement line. A structural correlation is demonstrated between the nanochannels and ellipsoidal mineral domains, which appear to coalesce during mineralization in a process analogous to powder sintering in metallurgy. A mineralization process is proposed, supported by energy-dispersive X-Ray spectroscopy of nanochannel contents, in which these unreported structures offer ion and molecule conduits to access the extracellular matrices of calcified cartilage and bone

    Breast cancer–secreted factors perturb murine bone growth in regions prone to metastasis

    Breast cancer frequently metastasizes to bone, causing osteolytic lesions. However, how factors secreted by primary tumors affect the bone microenvironment before the osteolytic phase of metastatic tumor growth remains unclear. Understanding these changes is critical as they may regulate metastatic dissemination and progression. To mimic premetastatic bone adaptation, immunocompromised mice were injected with MDA-MB-231–conditioned medium [tumor-conditioned media (TCM)]. Subsequently, the bones of these mice were subjected to multiscale, correlative analysis including RNA sequencing, histology, micro–computed tomography, x-ray scattering analysis, and Raman imaging. In contrast to overt metastasis causing osteolysis, TCM treatment induced new bone formation that was characterized by increased mineral apposition rate relative to control bones, altered bone quality with less matrix and more carbonate substitution, and the deposition of disoriented mineral near the growth plate. Our study suggests that breast cancer–secreted factors may promote perturbed bone growth before metastasis, which could affect initial seeding of tumor cells

    The effects of collagen peptides on muscle damage, inflammation and bone turnover following exercise:a randomized, controlled trial

    This study examined whether consuming collagen peptides (CP) before and after strenuous exercise alters markers of muscle damage, inflammation and bone turnover. Using a double-blind, independent group’s design, 24 recreationally active males consumed either 20 g day−1 of CP or a placebo control (CON) for 7 days before and 2 days after performing 150 drop jumps. Maximal isometric voluntary contractions, countermovement jumps (CMJ), muscle soreness (200 mm visual analogue scale), pressure pain threshold, Brief Assessment of Mood Adapted (BAM +) and a range of blood markers associated with muscle damage, inflammation and bone turnover C-terminal telopeptide of type 1 collagen (β-CTX) and N-terminal propeptides of type 1 pro-collagen (P1NP) were measured before supplementation (baseline; BL), pre, post, 1.5, 24 and 48 h post-exercise. Muscle soreness was not significantly different in CP and CON (P = 0.071) but a large effect size was evident at 48 h postexercise, indicative of lower soreness in the CP group (90.42 ± 45.33 mm vs. CON 125.67 ± 36.50 mm; ES = 2.64). CMJ height recovered quicker with CP than CON at 48 h (P = 0.050; CP 89.96 ± 12.85 vs. CON 78.67 ± 14.41% of baseline values; ES = 0.55). There were no statistically significant effects for the other dependent variables (P > 0.05). β-CTX and P1NP were unaffected by CP supplementation (P > 0.05). In conclusion, CP had moderate benefits for the recovery of CMJ and muscle soreness but had no influence on inflammation and bone collagen synthesis

    Defective Molecular Timer in the Absence of Nucleotides Leads to Inefficient Caspase Activation

    In the intrinsic death pathway, cytochrome C (CC) released from mitochondria to the cytosol triggers Apaf-1 apoptosome formation and subsequent caspase activation. This process can be recapitulated using recombinant Apaf-1 and CC in the presence of nucleotides ATP or dATP [(d)ATP] or using fresh cytosol and CC without the need of exogenous nucleotides. Surprisingly, we found that stored cytosols failed to support CC-initiated caspase activation. Storage of cytosols at different temperatures led to the loss of all (deoxy)nucleotides including (d)ATP. Addition of (d)ATP to such stored cytosols partially restored CC-initiated caspase activation. Nevertheless, CC could not induce complete caspase-9/3 activation in stored cytosols, even with the addition of (d)ATP, despite robust Apaf-1 oligomerization. The Apaf-1 apoptosome, which functions as a proteolytic-based molecular timer appeared to be defective as auto-processing of recruited procaspase-9 was inhibited. Far Western analysis revealed that procaspase-9 directly interacted with Apaf-1 and this interaction was reduced in the presence of physiological levels of ATP. Co-incubation of recombinant Apaf-1 and procaspase-9 prior to CC and ATP addition inhibited CC-induced caspase activity. These findings suggest that in the absence of nucleotide such as ATP, direct association of procaspase-9 with Apaf-1 leads to defective molecular timer, and thus, inhibits apoptosome-mediated caspase activation. Altogether, our results provide novel insight on nucleotide regulation of apoptosome

    High orientation of long chain branched poly (lactic acid) with enhanced blood compatibility and bionic structure

    YesHighly-oriented poly (lactic acid) (PLA) with bionic micro-grooves was fabricated through solid hot drawing technology for further improving the mechanical properties and blood biocompatibility of PLA. In order to enhance the melt strength and thus obtain high orientation degree, long chain branched PLA (LCB-PLA) was prepared at first through a two-step ring-opening reaction during processing. Linear viscoelasticity combined with branch-on-branch (BOB) model was used to predict probable compositions and chain topologies of the products, and it was found that the molecular weight of PLA increased and topological structures with star like chain with three arms and tree-like chain with two generations formed during reactive processing, and consequently draw ratio as high as1200% can be achieved during the subsequent hot stretching. With the increase of draw ratio, the tensile strength and orientation degree of PLA increased dramatically. Long chain branching and orientation could significantly enhance the blood compatibility of PLA by prolonging clotting time and decreasing platelet activation. Micro-grooves can be observed on the surface of the oriented PLA which were similar to the intimal layer of blood vessel, and such bionic structure resulted from the formation of the oriented shish kebab-like crystals along the draw direction

    An Optimized Classification Algorithm by Neural Network Ensemble Based on PLS and OLS

    Using the neural network to classify the data which has higher dimension and fewer samples means overmuch feature inputs influence the structure design of neural network and fewer samples will generate incomplete or overfitting phenomenon during the neural network training. All of the above will restrict the recognition precision obviously. It is even better to use neural network to classify and, therefore, propose a neural network ensemble optimized classification algorithm based on PLS and OLS in this paper. The new algorithm takes some advantages of partial least squares (PLS) algorithm to reduce the feature dimension of small sample data, which obtains the low-dimensional and stronger illustrative data; using ordinary least squares (OLS) theory determines the weights of each neural network in ensemble learning system. Feature dimension reduction is applied to simplify the neural network’s structure and improve the operation efficiency; ensemble learning can compensate for the information loss caused by the dimension reduction; on the other hand, it improves the recognition precision of classification system. Finally, through the case analysis, the experiment results suggest that the operating efficiency and recognition precision of new algorithm are greatly improved, which is worthy of further promotion
