    Biological Evaluation of New Thienopyridinium and Thienopyrimidinium Derivatives as Human Choline Kinase Inhibitors

    Due to its role in lipid biosynthesis, choline kinase α1 (CKα1) is an interesting target for the development of new antitumor agents. In this work, we present a series of 41 compounds designed based on the well-known and successful strategy of introducing thienopyridine and pyrimidine as bioisosteres of other heterocycles in active antitumor compounds. Notwithstanding the fact that some of these compounds do not show significant enzymatic inhibition, others, in contrast, feature substantially improved enzymatic and antiproliferative inhibition values. This is also confirmed by docking analysis, whereby compounds with longer linkers and thienopyrimidine cationic head have been identified as the most compelling. Among the best compounds is Ff-35, which inhibits the growth of different tumor cells at submicromolar concentrations. Moreover, Ff-35 is more potent in inhibiting CKα1 than other previous biscationic derivatives. Treatment of A549, Hela, and MDA-MB-231 cells with Ff-35 results in their arrest at the G1 phase of the cell cycle. Furthermore, the compound induces cellular apoptosis in a concentration-dependent manner. Altogether, these findings indicate that Ff-35 is a promising new chemotherapeutic agent with encouraging preclinical potential

    Colleters, osmophores, and nectaries in the species Ceropegia lenewtonii: a sapromyiophilous stapeliad (Asclepiadoideae, Apocynaceae)

    Ceropegia lenewtonii (Plowes) Bruyns (=Huernia keniensis), currently belonging to the Huernia section of the genus Ceropegia, is a stapeliad species distributed in Africa and the Arabian Peninsula; but it is widely cultivated as ornamental in most parts of the world. This species of stapeliad presents “carrion flowers” associated with a sapromyophilous pollination syndrome since the flowers emit an unpleasant odor. In this work, we describe the floral morphology and anatomy of the calyx, corolla, and corona of this species based on bright-field and scanning electron microscope techniques. We detected the presence of diverse floral secretor tissues, and based on different histochemical tests, the principal component of the secreted substance was identified. We interpret the functions of the glands and compare with other related species of stapeliads. Our results indicate that flowers of C. lenewtonii present colleters in sepals, osmophores in corolla, and primary and secondary nectaries in corona. All these floral glands have specific functions that involve the processes of pollination and reproduction of this species, as well as protection and defense mechanisms.Fil: Aliscioni, Sandra Silvina.     Estudio citogenético en rumiantes de importancia económica mediante el cultivo de linfocitos

    La citogenética dispone de métodos que permiten evaluar, directa o indirectamente, anomalías cromosómicas causantes de importantes mejoras o pérdidas ganaderas en varios países que las han estudiado. Por ello, diferentes países de zootecnia avanzada dan importancia creciente a los estudios citogenéticos, planteándose en algunos la necesidad o exigencia de realizar estudios del cariotipo en reproductores que se usan en centros de inseminación artificial, como de sus ascendientes o descendientes en test de progenie o bancos de embriones. Existen numerosas anomalías cromosómicas causantes de trastornos del desarrollo, neoplasias e infertilidad en el bovino, ovino, caprino, porcino, equino y otros animales domésticos. Las pérdidas estudiadas son cuantiosas y algunas de estas anomalías se refieren a menor tasa de fertilidad y cantidad de terneros. Debido a que la evaluación citogenética involucra etapas que consumen mucho tiempo, se desarrollan procedimientos semiautomáticos que reducen los tiempos significativamente, sin afectar la seguridad y consistencia. Por tal motivo, debido a que se dispone de un analizador digital de imágenes con el software base y software para el análisis del cariotipo, se capacitarán recursos humanos, que permitan luego agilizar la obtención de resultados. En Argentina, los estudios citogenéticos en el bovino son recientes, por lo que es necesario continuarlos y extenderlos a otros animales de importancia económica, afectiva o en vías de extinción, con el objeto de protegerlos de alteraciones hereditarias que resulten negativas. En este sentido, se han dispuesto algunas acciones que propenden a reglamentar la necesidad del cariotipado en reproductores dadores de semen y los que se emplean en la transferencia de embriones. No conocemos de estudios citogenéticos a nivel nacional en camélidos andinos, ovinos, caprinos, equinos, caninos y/o aves (...) 1. Objetivos Generales: - Desarrollar recursos humanos y técnicos para realizar estudios cariotípicos (cultivo y técnicas de bandeo) en animales de importancia económica. - Capacitación en análisis digital de imágenes para realizar el análisis cromosómico. - Realizar estudios cariotípicos en reproductores con problemas de fertilidad. - Realizar estudios cariotípicos en bovinos con diagnóstico serológico de leucosis. - Realizar estudios de prevalencia de anomalías cromosómicas en toros usados en inseminación artificial. - Ofrecer servicios de registros cariotípicos para reproductores empleados en inseminación artificial, transplante de embriones, animales con tumores o trastornos del desarrollo

    Adding Omega fatty acids (ω) 3 to the diet of stud bucks, the quality of sperm and their resistance to cryopreservation could increase. The aim of this study is to determine the effect of supplementation with ω3 on the metabolic state, sperm quality and resistance to freezing, in bucks kept in confinement under natural photoperiod. The experiment will be conducted in the facilities of the Faculty of Agronomy and Veterinary, UNRC (National University of Río Cuarto). Ten Anglo Nubian adult bucks, trained for semen collection with artificial vagina will be used. Males will be randomly allocated into 2 groups (5 animals each): control (C) and treatment (T). During the breeding season, group C will be fed with a ration of alfalfa and ground corn, according to the requirements for each category and sex (NRC, 2007). Group T will receive the same diet with the addition of linseeds. Both will have free access to water. Every week, semen of each buck, will be collected, evaluated and frozen. Sperm quality “in vitro” after thawing will be studied with a digital image analyzer. To assess oxidative stress in fresh and cryopreserved semen, levels of thiobarbituric acid reactive substances (TBARS) and quantification of the activity of superoxide dismutase (SOD) and catalase (CAT) will be determined. To establish the metabolic state, blood samples will be collected every two weeks. The statistical analysis will include an exploratory data analysis, multivariate analysis of multiple correspondences on a completely randomized design, analysis of variance and Fisher post-test. The level of significance will be set at P <0.05 and all results will be expressed as means ± SEM
