    Effect of epithelial debridement on human cornea proteoglycans

    Corneal transparency is attributed to the regular spacing and diameter of collagen fibrils, and proteoglycans may play a role in fibrillogenesis and matrix assembly. Corneal scar tissue is opaque and this opacity is explained by decreased ultrastructural order that may be related to proteoglycan composition. Thus, the objectives of the present study were to characterize the proteoglycans synthesized by human corneal explants and to investigate the effect of mechanical epithelial debridement. Human corneas unsuitable for transplants were immersed in F-12 culture medium and maintained under tissue culture conditions. The proteoglycans synthesized in 24 h were labeled metabolically by the addition of 35S-sulfate to the medium. These compounds were extracted by 4 M GuHCl and identified by a combination of agarose gel electrophoresis, enzymatic degradation with protease and mucopolysaccharidases, and immunoblotting. Decorin was identified as the main dermatan sulfate proteoglycan and keratan sulfate proteoglycans were also prominent components. When the glycosaminoglycan side chains were analyzed, only keratan sulfate and dermatan sulfate were detected (~50% each). Nevertheless, when these compounds were 35S-labeled metabolically, the label in dermatan sulfate was greater than in keratan sulfate, suggesting a lower synthesis rate for keratan sulfate. 35S-Heparan sulfate also appeared. The removal of the epithelial layer caused a decrease in heparan sulfate labeling and induced the synthesis of dermatan sulfate by the stroma. Despite the relevance of this species, the genetic research on the goat traits is limited compared to other domestic species. Thanks to the latest goat reference genomic sequence (ARS1), which is considered to be one of the most continuous assemblies in livestock, we deduced the genomic structure of the T cell receptor beta (TRB) and gamma (TRG) loci in this ruminant species

    Concentrations of available heavy metals in mediterranean agricultural soils and their relation with some soil selected properties: a case study in typical mediterranean soils

    The characterization of the content of trace metals in soils is an instrument in many programs of environmental protection, including the establishment of regional-level standards to detect sites affected by contamination. The objectives of the present study were to study the available levels of Cd, Cr, Cu, Ni, Pb, and Zn in surface horizons of agricultural soils in a typical European Mediterranean region, to establish the geochemical baseline concentration (GBC), background level (BL), and reference value (RV) of each of these available metals, and to investigate their possible correlations with soil properties. To establish the GBC and RV values, we used the " standard threshold method". Topsoil samples (0-20 cm) were collected from 630 sites, and extracted with Diethylene Triamine Pentaacetic Acid (DTPA) to determine their available heavy metal concentrations. The GBC values established were: 0.04 to 0.90 mg kg(-1), 0.70 to 2.50 mg kg(-1), 0.10 to 6.30 mg kg(-1), 0.30 to 7.90 mg kg(-1), 0.29 to 4.50 mg kg(-1), and 0.18 to 2.50 mg kg(-1) for Cd, Cr, Cu, Ni, Pb, and Zn, respectively.     Coordinación de asignaturas de Física en titulaciones de Grado y Master para Ingenierías y Arquitectura

    En este trabajo de investigación docente realizamos una puesta en común y recopilación de materiales docentes relacionados con asignaturas de Física de titulaciones de Master y Grado orientadas a la ingeniería y la arquitectura impartidas en la Escuela Politécnica Superior de la Universidad de Alicante. Para ello se fomenta la comunicación e intercambio de experiencias entre los docentes de nuestro departamento que imparten estas asignaturas con la finalidad de optimizar los recursos humanos, técnicos y económicos disponibles. Todo el material recopilado se revisa previamente por cada docente y se publica en servidores conectados a una red interna del departamento lo que permite el acceso de todo el personal docente relacionado con estas asignaturas a dichos recursos docentes fomentando la colaboración, mejora y enriquecimiento profesional entre los miembros involucrados de nuestro departamento.     Safety and Efficacy of Dolutegravir in Treatment-Experienced Subjects With Raltegravir-Resistant HIV Type 1 Infection: 24-Week Results of the VIKING Study

    Background. Dolutegravir (DTG; S/GSK1349572), a human immunodeficiency virus type 1 (HIV-1) integrase inhibitor, has limited cross-resistance to raltegravir (RAL) and elvitegravir in vitro. This phase IIb study assessed the activity of DTG in HIV-1–infected subjects with genotypic evidence of RAL resistance.Methods. Subjects received DTG 50 mg once daily (cohort I) or 50 mg twice daily (cohort II) while continuing a failing regimen (without RAL) through day 10, after which the background regimen was optimized, when feasible, for cohort I, and at least 1 fully active drug was mandated for cohort II. The primary efficacy end point was the proportion of subjects on day 11 in whom the plasma HIV-1 RNA load decreased by ≥0.7 log10 copies/mL from baseline or was <400 copies/mL.Results. A rapid antiviral response was observed. More subjects achieved the primary end point in cohort II (23 of 24 [96%]), compared with cohort I (21 of 27 [78%]) at day 11. At week 24, 41% and 75% of subjects had an HIV-1 RNA load of <50 copies/mL in cohorts I and II, respectively. Further integrase genotypic evolution was uncommon. Dolutegravir had a good, similar safety profile with each dosing regimen.Conclusion. Dolutegravir 50 mg twice daily with an optimized background provided greater and more durable benefit than the once-daily regimen. These data are the first clinical demonstration of the activity of any integrase inhibitor in subjects with HIV-1 resistant to RAL

    The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & Nemésio 2007; Donegan 2008, 2009; Nemésio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016

    As câmeras cinematográficas nos anos 1950/1960 e o cinema brasileiro

    The motion picture cameras have changed through the years. However, they have kept their main design which has not changed during this period. One of the goals of this text is to understand how that design was created, and also the changes it has suffered without losing its essence. Besides that, this text aims to understand how different cameras connected with the Brazilian cinema production in the fifties and sixties. A change on camera s use is noticeable in that period in Brazil. There is a change in appreciation from the more traditional cameras linked to the studio system to the European cameras developed in the thirties and forties, which were lighter and more portable. This issue coincides with a specific characteristic in Brazilian cinema at that time, when people started to believe in the independent cinema production as an answer to the studio system, which was the main thought until then. In order to show this entire context, it has been chosen the Vera Cruz and Cinema Novo, their movies and shootings, to confront and connect them to camera models and their relation to the mode of productionAs câmeras cinematográficas passaram por mudanças ao longo dos anos. Porém, manteve um design que se perpetuou durante este período. Um dos objetivos do presente trabalho é entender como este design foi criado, e as variações pelas quais ele passou, sem perder a sua essência. Em paralelo a isso, entender como estas diferentes câmeras travaram relação com a produção do cinema brasileiro dos anos 1950 e 1960. É nesta época que se observa no país uma troca de postura em relação às câmeras. Passa-se de uma valorização das câmeras mais tradicionais, ligadas ao sistema de estúdio, à valorização das câmeras européias criadas no anos 1930 e 1940, que eram mais leves e portáteis. Isso coincide com um momento específico do cinema brasileiro, aquele em que se passa a crer numa solução de cinema independente como resposta ao cinema de estúdio, que era o pensamento majoritário até então. Para ilustrar todo este contexto, escolhemos a Vera Cruz e o Cinema Novo, seus filmes e filmagens, para relacioná-los e confrontá-los em relação aos tipos de câmeras e sua relação ao modo de produção
