    High porosity metallic glass foam: A powder metallurgy route

    A powder metallurgy route to the fabrication of metallic glass foam is introduced. The method involves consolidating metallic glass powder blended with blowing agent particulates to produce expandable precursors, capable of yielding foams with porosities as high as 86%. The foams are found to inherit the strength of the parent metallic glass and to be able to deform heavily toward full densification absorbing high amounts of energy

    Novel Methods in Disease Biogeography: A Case Study with Heterosporosis

    Disease biogeography is currently a promising field to complement epidemiology, and ecological niche modeling theory and methods are a key component. Therefore, applying the concepts and tools from ecological niche modeling to disease biogeography and epidemiology will provide biologically sound and analytically robust descriptive and predictive analyses of disease distributions. As a case study, we explored the ecologically important fish disease Heterosporosis, a relatively poorly understood disease caused by the intracellular microsporidian parasite Heterosporis sutherlandae. We explored two novel ecological niche modeling methods, the minimum-volume ellipsoid (MVE) and the Marble algorithm, which were used to reconstruct the fundamental and the realized ecological niche of H. sutherlandae, respectively. Additionally, we assessed how the management of occurrence reports can impact the output of the models. Ecological niche models were able to reconstruct a proxy of the fundamental and realized niche for this aquatic parasite, identifying specific areas suitable for Heterosporosis. We found that the conceptual and methodological advances in ecological niche modeling provide accessible tools to update the current practices of spatial epidemiology. However, careful data curation and a detailed understanding of the algorithm employed are critical for a clear definition of the assumptions implicit in the modeling process and to ensure biologically sound forecasts. In this paper, we show how sensitive MVE is to the input data, while Marble algorithm may provide detailed forecasts with a minimum of parameters. We showed that exploring algorithms of different natures such as environmental clusters, climatic envelopes, and logistic regressions (e.g., Marble, MVE, and Maxent) provide different scenarios of potential distribution. Thus, no single algorithm should be used for disease mapping. Instead, different algorithms should be employed for a more informed and complete understanding of the pathogen or parasite in question

    Network metrics can guide nearly-optimal management of invasive species at large scales

    Invasive species harm biodiversity and ecosystem services, with global economic costs of invasions exceeding $40 billion annually. Widespread invasions are a particular challenge because they involve large spatial scales with many interacting components. In these contexts, typical optimization-based approaches to management may fail due to computational or data constraints. Here we evaluate an alternative solution that leverages network science, representing the invasion as occurring across a network of connected sites and using network metrics to prioritize sites for intervention. Such heuristic network-guided methods require less data and are less computationally intensive than optimization methods, yet network-guided approaches have not been bench-marked against optimal solutions for real-world invasive species management problems. We provide the first comparison of the performance of network-guided management relative to optimal solutions for invasive species, examining the placement of watercraft inspection stations for preventing spread of invasive zebra mussels through recreational boat movement within 58 Minnesota counties in the United States. To additionally test the promise of network-based approaches in limited data contexts, we evaluate their performance when using only partial data on network structure and invaded status. Metric-based approaches can achieve a median of 100% of optimal performance with full data. Even with partial data, 80% of optimal performance is achievable. Finally, we show that performance of metric-guided management improves for counties with denser and larger networks, suggesting this approach is viable for large-scale invasions. Together, our results suggest network metrics are a promising approach to guiding management actions for large-scale invasions.Comment: 29 pages, 8 figures, 3 table

    Beta-HPV E6 Contributes To Skin Cancer by Hindering DNA Repair

    <div><p>Recent work has explored a putative role for the E6 protein from some β-human papillomavirus genus (β-HPVs) in the development of non-melanoma skin cancers, specifically β-HPV 5 and 8 E6. Because these viruses are not required for tumor maintenance, they are hypothesized to act as co-factors that enhance the mutagenic capacity of UV-exposure by disrupting the repair of the resulting DNA damage. Supporting this proposal, we have previously demonstrated that UV damage signaling is hindered by β-HPV 5 and 8 E6 resulting in an increase in both thymine dimers and UV-induced double strand breaks (DSBs). Here we show that β-HPV 5 and 8 E6 further disrupt the repair of these DSBs and provide a mechanism for this attenuation. By binding and destabilizing a histone acetyltransferase, p300, β-HPV 5 and 8 E6 reduce the enrichment of the transcription factor at the promoter of two genes critical to the homology dependent repair of DSBs (BRCA1 and BRCA2). The resulting diminished BRCA1/2 transcription not only leads to lower protein levels but also curtails the ability of these proteins to form repair foci at DSBs. Using a GFP-based reporter, we confirm that this reduced foci formation leads to significantly diminished homology dependent repair of DSBs. By deleting the p300 binding domain of β-HPV 8 E6, we demonstrate that the loss of robust repair is dependent on viral-mediated degradation of p300 and confirm this observation using a combination of p300 mutants that are β-HPV 8 E6 destabilization resistant and p300 knock-out cells. In conclusion, this work establishes an expanded ability of β-HPV 5 and 8 E6 to attenuate UV damage repair, thus adding further support to the hypothesis that β-HPV infections play a role in skin cancer development by increasing the oncogenic potential of UV exposure.</p></div

    The Most Slowly Declining Type Ia Supernova 2001ay

    We present optical and near-infrared photometry, as well as ground-based optical spectra and Hubble Space Telescope ultraviolet spectra, of the Type Ia supernova (SN) 2001ay. At maximum light the Si II and Mg II lines indicated expansion velocities of 14,000 km/sec, while Si III and S II showed velocities of 9,000 km/sec There is also evidence for some unburned carbon at 12,000 km/sec. SN 2001ay exhibited a decline-rate parameter Delta m_15(B) = 0.68 \pm 0.05 mag; this and the B-band photometry at t > +25 d past maximum make it the most slowly declining Type Ia SN yet discovered. Three of four super-Chandrasekhar-mass candidates have decline rates almost as slow as this. After correction for Galactic and host-galaxy extinction, SN 2001ay had M_B = -19.19 and M_V = -19.17 mag at maximum light; thus, it was not overluminous in optical bands. In near-infrared bands it was overluminous only at the 2-sigma level at most. For a rise time of 18 d (explosion to bolometric maximum) the implied Ni-56 yield was (0.58 \pm 0.15)/alpha M_Sun, with alpha = L_max/E_Ni probably in the range 1.0 to 1.2. The Ni-56 yield is comparable to that of many Type Ia supernovae. The "normal" Ni-56 yield and the typical peak optical brightness suggest that the very broad optical light curve is explained by the trapping of the gamma rays in the inner regions.Comment: 57 pages, 22 figures.     Naked1 Antagonizes Wnt Signaling by Preventing Nuclear Accumulation of β-Catenin

    Cyto-nuclear shuttling of β-catenin is at the epicenter of the canonical Wnt pathway and mutations in genes that result in excessive nuclear accumulation of β-catenin are the driving force behind the initiation of many cancers. Recently, Naked Cuticle homolog 1 (Nkd1) has been identified as a Wnt-induced intracellular negative regulator of canonical Wnt signaling. The current model suggests that Nkd1 acts between Disheveled (Dvl) and β-catenin. Here, we employ the zebrafish embryo to characterize the cellular and biochemical role of Nkd1 in vivo. We demonstrate that Nkd1 binds to β-catenin and prevents its nuclear accumulation. We also show that this interaction is conserved in mammalian cultured cells. Further, we demonstrate that Nkd1 function is dependent on its interaction with the cell membrane. Given the conserved nature of Nkd1, our results shed light on the negative feedback regulation of Wnt signaling through the Nkd1-mediated negative control of nuclear accumulation of β-catenin

    Mind the gaps! A research agenda for urban interstices

    Processes of urbanisation can hardly be considered without reference to the spaces that lie between developments. However, the literature on such interstitial spaces is fragmentary. In this paper we draw together insights from the extant literature into a research agenda on urban interstices. We propose a research agenda centred on four themes: the multiple geographic scales at which the interstitial spaces of urban sprawl might be analysed; the pending nature of such spaces; their planned or unplanned character and their relational properties. We develop these themes, briefly illustrating them with reference to the case of metropolitan area of Santiago de Chile. In conclusion, we emphasise some of the implications of interstitial spaces for theories of urban politics and their value in forcing inter-disciplinarity in urban studies
