    Efecto de las condiciones de almacenamiento refrigerado sobre el estrés oxidativo en raquis de uva de mesa, cv. Red Globe

In the present report, physiological parameters of Red Globe rachises from fully elongated inflorescences (RFEI) and from mature clusters were studied after storage at 0 or 20 °C for different durations. To understand changes in rachis physiology as a result of changes in temperature conditions and storage time, the activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) were measured. In addition, hydrogen peroxide content, membrane lipoperoxidation (TBARS), total phenolic compounds and antioxidant capacity (FRAP) were assayed. TBARS was higher in mature rachises than in RFEI. This parameter remained constant throughout storage, indicating a change presumably associated with ontogeny or senescence processes. Short-term storage (096 h) increased SOD, CAT and APX activities in RFEI, while in mature rachises, no changes were observed in enzyme activities or in hydrogen peroxide content. Longer cold storage (25 or 53 days at 0 °C) of mature rachises reduced CAT activity, but SOD and APX activities did not change under these conditions. At 0 h, the FRAP and total phenolic contents of mature rachises were three and 20 times higher than in immature rachises, respectively.La calidad de uva de mesa (Vitis vinifera L.) involucra tanto la condición de bayas y el raquis. En el presente trabajo, los parámetros fisiológicos de raquis de 'Red Globe' de inflorescencias completamente elongadas (RFEI) y de racimos maduros fueron estudiados después de almacenamientos por diferentes tiempos a 0 ó 20 °C. Para entender los cambios en la fisiología del raquis debido a variaciones de temperatura y condiciones de almacenamiento, se midieron las actividades de superoxido dismutasa (SOD), catalasa (CAT), ascorbato peroxidasa (APX). Además fueron analizados el contenido de peróxido de hidrógeno, lipoperoxidación de membranas (TBARS), compuestos fenólicos totales y capacidad antioxidante (FRAP). TBARS de los raquis maduros a la cosecha fue mayor que el de inflorescencias completamente elongadas (RFEI). Este parámetro permaneció constante a través del almacenamiento, indicando cambios presuntamente asociados a ontogenia o procesos de senescencia. Almacenamiento cortos (0-96h) incrementaron la actividad de SOD, CAT y APX en RFEI, sin embargo no se observaron cambios en la actividad de estas enzimas o contenido de peróxido de hidrogeno en raquis maduros. Almacenamiento refrigerado prolongado (25 ó 30 días 0 °C) de raquis maduros redujo la actividad de CAT, pero SOD y APX no mostraron cambios bajo estas condiciones. A 0 h el contenido de FRAP y fenólicos totales de raquis maduros fueron tres y 20 veces mayores que en raquis inmaduro respectivamente.http://ref.scielo.org/m3mg4

    Comparative branching order and root anatomy of clonal and seed-grown avocado trees (Persea americana Mill.)

    Characterizing roots according to their branching order and anatomy is a useful approach for identifying functional differences within and among root systems. In this study, the root branching order and the anatomy of each root order (stele and cortex area) were examined in two-year-old “Duke 7” avocado (Persea americana Mill.) trees propagated by seed and by clonal techniques. The root systems were found to have three different root orders that exhibited differences in the occurrence of secondary xylem. Fine roots (first- and second-order roots) presented only primary growth, whereas the main roots (third-order roots) exhibited secondary growth. Transverse sections of roots from the different orders showed pentarch, hexarch or heptarch tracheal element distributions. Newly emerged, long, unbranched pioneer roots were observed only in the clonal trees and showed particular anatomical features, such as a larger diameter and a proportionally greater cortex area than other roots, as well as primary growth. Additionally, significant differences were found between clonal and seedling trees in the stele area of third-order roots; clonal propagation resulted in larger stele areas in this type of root. Our results suggest that propagation methods influence the presence of new pioneer roots and the anatomy of third-order roots; clonal root systems branch more extensively than seed-grown root systems and develop a vascular system with a larger transport capacity

    Proteomic and low-polar metabolite profiling reveal unique dynamics in fatty acid metabolism during flower and berry development of table grapes

    Grapevine development and ripening are complex processes that involve several biochemical pathways, including fatty acid and lipid metabolism. Fatty acids are essential components of lipids, which play crucial roles in fruit maturation and flavor development. However, the dynamics of fatty acid metabolism in grape flowers and berries are poorly understood. In this study, we present those dynamics and investigate the mechanisms of fatty acid homeostasis on ‘Thompson Seedless’ berries using metabolomic and proteomic analyses. Low-polar metabolite profiling indicated a higher abundance of fatty acids at the pre-flowering and pre-veraison stages. Proteomic analyses revealed that grape flowers and berries display unique profiles of proteins involved in fatty acid biosynthesis, triacylglycerol assembly, fatty acid β-oxidation, and lipid signaling. These findings show, for the first time, that fatty acid metabolism also plays an important role in the development of non-oil-rich tissues, opening new perspectives about lipid function and its relation to berry quality

    Abdominal aortic aneurysm screening program using hand-held ultrasound in primary healthcare

    We determined the feasibility of abdominal aortic aneurysm (AAA) screening program led by family physicians in public primary healthcare setting using hand-held ultrasound device. The potential study population was 11,214 men aged ≥ 60 years attended by three urban, public primary healthcare centers. Participants were recruited by randomly-selected telephone calls. Ultrasound examinations were performed by four trained family physicians with a hand-held ultrasound device (Vscan®). AAA observed were verified by confirmatory imaging using standard ultrasound or computed tomography. Cardiovascular risk factors were determined. The prevalence of AAA was computed as the sum of previously-known aneurysms, aneurysms detected by the screening program and model-based estimated undiagnosed aneurysms. We screened 1,010 men, with mean age of 71.3 (SD 6.9) years; 995 (98.5%) men had normal aortas and 15 (1.5%) had AAA on Vscan®. Eleven out of 14 AAA-cases (78.6%) had AAA on confirmatory imaging (one patient died). The total prevalence of AAA was 2.49% (95%CI 2.20 to 2.78). The median aortic diameter at diagnosis was 3.5 cm in screened patients and 4.7 cm (p<0.001) in patients in whom AAA was diagnosed incidentally. Multivariate logistic regression analysis identified coronary heart disease (OR = 4.6, 95%CI 1.3 to 15.9) as the independent factor with the highest odds ratio. A screening program led by trained family physicians using hand-held ultrasound was a feasible, safe and reliable tool for the early detection of AAA

    Photography-based taxonomy is inadequate, unnecessary, and potentially harmful for biological sciences

    The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & Nemésio 2007; Donegan 2008, 2009; Nemésio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016

    Nitrate Transport Rate in the Xylem of Tomato Plants Grafted onto a Vigorous Rootstock

    Vigorous interspecific rootstocks increase nitrogen (N) uptake in tomato plants but limited information is available on xylem transport rate. Non-grafted and self-grafted tomato plants cv. Attiya and plants grafted onto an interspecific hybrid, Kaiser, were grown under growth chamber conditions and subjected to two light levels, 400 or 800 &micro;mol PAR m&minus;2 s&minus;1. Plant water uptake, xylem sap NO3&minus; content, and stem hydraulic conductance (ks) were measured after two weeks of growth. Xylem vessel number and diameter were evaluated in cross-sectional stem cuts and the theoretical xylem hydraulic conductance (kh) was calculated. Only the light level modified the xylem NO3&minus; content. Grafting reduced ks by 84% in comparison to non-grafted plants. The water uptake rate and xylem sap NO3&minus; content were 4.02 &plusmn; 0.66 g H2O kg&minus;1 DW h&minus;1 and 12.78 &plusmn; 1.16 mM, respectively, across all grafting treatments. The rootstock has a higher kh because the vessel diameter is 79.3 &plusmn; 14.4 &micro;m while in non-grafted plants it is 62.0 &plusmn; 10.1 &micro;m.     Exogenous Applications of Brassinosteroids Improve Color of Red Table Grape (Vitis vinifera L. Cv. “Redglobe”) Berries

    Color and other quality parameters of “Redglobe” grape (Vitis vinifera L.) berries were evaluated after treatment with brassinosteroid (BR) analogs. Three BRs analogs (24-epibrassinolide, Triol, or Lactone) were applied at three concentrations (0.0, 0.4, or 0.8 mg⋅L-1), at the onset of veraison. A commercial formulation (B-2000®) was also applied, at a recommended rate of 0.06 mg⋅L-1. The tested BR analogs were effective improving berry color (evaluated as color index for red grapes, CIRG), increasing the levels of soluble solids and anthocyanins, and changing the types of anthocyanins present without altering other quality and yield parameters. The effects of BR analogs on color enhancement could be explained by an increase in soluble solids content and/or anthocyanin content. Treatment with 24-epibrassinolide (at 0.4 mg⋅L-1) or the commercial formulation tended to favor the production of dihydroxylated anthocyanins, which are responsible for the red and pink colors of grape berries. Results indicate that the use of BRs constitutes a potential tool in the production of table grapes. This is the first report of this enhancement effect in a productive context

    Xylella fastidiosa Infection and Ethylene Exposure Result in Xylem and Water Movement Disruption in Grapevine Shoots

    It is conventionally thought that multiplication of the xylem-limited bacterium Xylella fastidiosa (Xf) within xylem vessels is the sole factor responsible for the blockage of water movement in grapevines (Vitis vinifera) affected by Pierce's disease. However, results from our studies have provided substantial support for the idea that vessel obstructions, and likely other aspects of the Pierce's disease syndrome, result from the grapevine's active responses to the presence of Xf, rather than to the direct action of the bacterium. The use of magnetic resonance imaging (MRI) to observe the distribution of water within the xylem has allowed us to follow nondestructively the development of vascular system obstructions subsequent to inoculation of grapevines with Xf. Because we have hypothesized a role for ethylene produced in vines following infection, the impact of vine ethylene exposure on obstruction development was also followed using MRI. In both infected and ethylene-exposed plants, MRI shows that an important proportion of the xylem vessels become progressively air embolized after the treatments. The loss of xylem water-transporting function, assessed by MRI, has been also correlated with a decrease in stem-specific hydraulic conductivity (K(S)) and the presence of tyloses in the lumens of obstructed water conduits. We have observed that the ethylene production of leaves from infected grapevines is greater than that from healthy vines and, therefore, propose that ethylene may be involved in a series of cellular events that coordinates the vine's response to the pathogen
