Anti-neoplastic drugs increase caveolin-1-dependent migration, invasion and metastasis of cancer cells
Indexación: Scopus.Expression of the scaffolding protein Caveolin-1 (CAV1) enhances migration and invasion of metastatic cancer cells. Yet, CAV1 also functions as a tumor suppressor in early stages of cancer, where expression is suppressed by epigenetic mechanisms. Thus, we sought to identify stimuli/mechanisms that revert epigenetic CAV1 silencing in cancer cells and evaluate how this affects their metastatic potential. We reasoned that restricted tissue availability of anti-neoplastic drugs during chemotherapy might expose cancer cells to sub-therapeutic concentrations, which activate signaling pathways and the expression of CAV1 to favor the acquisition of more aggressive traits. Here, we used in vitro [2D, invasion] and in vivo (metastasis) assays, as well as genetic and biochemical approaches to address this question. Colon and breast cancer cells were identified where CAV1 levels were low due to epigenetic suppression and could be reverted by treatment with the methyltransferase inhibitor 5'-azacytidine. Exposure of these cells to anti-neoplastic drugs for short periods of time (24-48 h) increased CAV1 expression through ROS production and MEK/ERK activation. In colon cancer cells, increased CAV1 expression enhanced migration and invasion in vitro via pathways requiring Src-family kinases, as well as Rac-1 activity. Finally, elevated CAV1 expression in colon cancer cells following exposure in vitro to sub-cytotoxic drug concentrations increased their metastatic potential in vivo. Therefore exposure of cancer cells to anti-neoplastic drugs at non-lethal drug concentrations induces signaling events and changes in transcription that favor CAV1-dependent migration, invasion and metastasis. Punto de corte de circunferencia braquial media: importancia obstétrica y neonatal
Introducción: La preeclampsia es un trastorno multisis- témico que se encuentra entre las principales causas de morbi-mortalidad materna durante la gestación, la obe- sidad es uno de los factores que interviene en la siopa- tología de esta alteración vascular. Por lo cual el objetivo de este estudio es evaluar el comportamiento del Índice de Masa Corporal (IMC) y la Circunferencia Braquial Me- dia (CBM) en pacientes preclámpticas ingresadas en la Maternidad Enrique C. Sotomayor de la ciudad de Guaya- quil, así como establecer un punto de corte del parámetro más relacionado con la preeclampsia. Materiales y Métodos: Se realizó un estudio caso-con- trol, aleatorio, en 30 mujeres sin preeclampsia y 30 con preeclampsia ingresadas en la Maternidad Enrique C. So- tomayor de la ciudad de Guayaquil, Ecuador, 2010. Se realizó una Curva COR para determinar el punto de corte de CBM para preeclampsia. Asimismo se realizó un mod- elo de regresión logística múltiple para factores asociados a complicación neonatal. Resultados: Las mujeres preeclámpticas presentaron va- lores más altos de CBM (30,3cm); p<0.0001. El punto de corte seleccionado para el diagnóstico de preeclampsia fue 27,5cm con un ABC=0,898 (sensibilidad: 90%; espe- ci cidad: 83,3%). La CBM alta fue el factor más asociado con la presencia de complicación neonatal (OR: 5,13; IC95%: 1,04-25,39; p=0,04). Conclusiones: Se sugiere medir la CBM en la paciente gestante para la valoración de obesidad, siendo 27,5cm el punto de corte a partir del cual hay una mayor relación con preeclampsia y complicaciones neonatales como bajo peso al nacer. 
Caveolin-1-Enhanced Motility and Focal Adhesion Turnover Require Tyrosine-14 but Not Accumulation to the Rear in Metastatic Cancer Cells
Caveolin-1 is known to promote cell migration, and increased caveolin-1 expression is associated with tumor progression and metastasis. In fibroblasts, caveolin-1 polarization and phosphorylation of tyrosine-14 are essential to promote migration. However, the role of caveolin-1 in migration of metastatic cells remains poorly defined. Here, caveolin-1 participation in metastatic cell migration was evaluated by shRNA targeting of endogenous caveolin-1 in MDA-MB-231 human breast cancer cells and ectopic expression in B16-F10 mouse melanoma cells. Depletion of caveolin-1 in MDA-MB-231 cells reduced, while expression in B16-F10 cells promoted migration, polarization and focal adhesion turnover in a sequence of events that involved phosphorylation of tyrosine-14 and Rac-1 activation. In B16-F10 cells, expression of a non-phosphorylatable tyrosine-14 to phenylalanine mutant failed to recapitulate the effects observed with wild-type caveolin-1. Alternatively, treatment of MDA-MB-231 cells with the Src family kinase inhibitor PP2 reduced caveolin-1 phosphorylation on tyrosine-14 and cell migration. Surprisingly, unlike for fibroblasts, caveolin-1 polarization and re-localization to the trailing edge were not observed in migrating metastatic cells. Thus, expression and phosphorylation, but not polarization of caveolin-1 favor the highly mobile phenotype of metastatic cells
Comparing algorithms for automated vessel segmentation in computed tomography scans of the lung: the VESSEL12 study
The VESSEL12 (VESsel SEgmentation in the Lung) challenge objectively compares the performance of different algorithms to identify vessels in thoracic computed tomography (CT) scans. Vessel segmentation is fundamental in computer aided processing of data generated by 3D imaging modalities. As manual vessel segmentation is prohibitively time consuming, any real world application requires some form of automation. Several approaches exist for automated vessel segmentation, but judging their relative merits is difficult due to a lack of standardized evaluation. We present an annotated reference dataset containing 20 CT scans and propose nine categories to perform a comprehensive evaluation of vessel segmentation algorithms from both academia and industry. Twenty algorithms participated in the VESSEL12 challenge, held at International Symposium on Biomedical Imaging (ISBI) 2012. All results have been published at the VESSEL12 website http://vessel12.grand-challenge.org. The challenge remains ongoing and open to new participants. Latitude gradient influences the age of onset of rheumatoid arthritis : a worldwide survey
The age of onset of rheumatoid arthritis (RA) is an important outcome predictor. Northern countries report an age of RA onset of around 50 years, but apparently, variability exists across different geographical regions. The objective of the present study is to assess whether the age of onset of RA varies across latitudes worldwide. In a proof-of-concept cross-sectional worldwide survey, rheumatologists from preselected cities interviewed 20 consecutive RA patients regarding the date of RA onset (RAO, when the patient first noted a swollen joint). Other studied variables included location of each city, rheumatologist settings, latitudes (10A degrees increments, south to north), longitudes (three regions), intracountry consistency, and countries' Inequality-adjusted Human Development Index (IHDI). Data from 2481 patients (82% females) were obtained from 126 rheumatologists in 77 cities of 41 countries. Worldwide mean age of RAO was 44 +/- 14 years (95% CI 44-45). In 28% of patients, RA began before age 36 years and before age 46 years in 50% of patients. RAO was 8 years earlier around the Tropic of Cancer when compared with northern latitudes (p <0.001, 95% CI 3.5-13). Multivariate analysis showed that females, western cities, and latitudes around the Tropic of Cancer are associated with younger age of RAO (R (2) 0.045, p <0.001). A positive correlation was found between the age of RAO and IHDI (r = 0.7, p <0.01, R (2) 0.5). RA often begins at an early age and onset varies across latitudes worldwide. We postulate that countries' developmental status and their geographical and geomagnetic location influence the age of RAO.Peer reviewe
Prevalencia de hipertensión arterial en individuos adultos de las parroquias urbanas de la ciudad de Cuenca, Ecuador
Introducción y Objetivo: La hipertensión es el másimportante factor de riesgo modificable, para la enfermedadcardiovascular, cerebrovascular y enfermedad renal,muy prevalente en el mundo. Sus características en nuestrapoblación han sido muy poco estudiadas por lo que elobjetivo fue determinar la prevalencia y comportamientoepidemiológico de la HTA en las parroquias urbanas de laciudad de Cuenca, Ecuador.Materiales y Métodos: Se realizó un estudio de campoy transversal en 318 individuos de ambos sexos seleccionadosaleatoriamente. Las variables cuantitativasse expresaron como media±DE, las comparaciones entreestas se realizaron con la prueba T de Student. Las variablescualitativas representadas en frecuencias absolutasy relativas, las comparaciones mediante la prueba Z deproporciones y asociaciones con la prueba del chi cuadrado.Resultados: La prevalencia de HTA conocida fue deun 14,78% (15,38% en mujeres y 13,97% en hombres);la prevalencia de nuevos diagnósticos de HTA fue de11,01% (9,34% en mujeres y 13,24% en hombres). Elporcentaje de HTA total fue de 25,79% (24,72% en mujeresy 27,21% en hombres). El nivel de control de PA enlos individuos hipertensos fue de un 51,1% según las metasde control de la JNC-7 y de un 61,7% según la JNC-8.Conclusión: Se observa una menor prevalencia de HTAen nuestra población y un mejor control de la PA con respectoa otros estudios realizados a nivel regional, nacionaly otros países.Palabras clave: hipertensión arterial, obesidad, enfermedadcardiovascular, diabetes, Cuenc
Associated risk factors for arterial hypertension in an adult population of the parish Quingeo, Cuenca-Ecuador.
Introducción: La hipertensión arterial (HTA) es un factor
de riesgo modificable para la presencia de enfermedades
vasculares, cardiacas y renales, con importantes repercusiones
en la salud pública mundial. Los datos epidemiológicos
de esta patología en poblaciones rurales de Ecuador
son limitados, por lo que el objetivo de este estudio fue
evaluar la prevalencia y factores asociados a HTA en la
población rural de Quingeo, Ecuador.
Materiales y Métodos: Se realizó un estudio analítico,
transversal, con muestreo aleatorio multietápico que incluyó
530 individuos mayores de 18 años, de ambos sexos,
a quienes se les aplicó una historia clínica completa.
La presión arterial se clasificó según los criterios de la
JNC7. Se construyó un modelo de regresión logística para
determinar los factores asociados a HTA.
Resultados: La prevalencia de HTA fue de 16,2% (n=86),
siendo para las mujeres de 16,3% (n=51) y para los hombres
de 16,1% (n=35). A su vez, la edad mostró asociación
con la prevalencia de HTA (χ2=4,276; p<0,001), con
porcentajes más altos a mayor grupo etario. Los principales
factores asociados a la HTA fueron ser adulto medio
(OR=3,41; IC95%:1,73-6,09; p<0,001), adulto mayor
(OR=4,98; IC 95%:2,24-10,09; p<0,001), consumo de
alcohol (OR=2,56; IC95%:1,02-6,44; p=0,044), estado
civil divorciado (OR=5,16; IC95%:1,53-17,44; p=0,008)
y la actividad física alta en la esfera de ocio (OR=0,20;
IC95%:0,08-0,94; p=0,040).
Conclusión: Existe una baja prevalencia de HTA en la población
rural de Quingeo en comparación a otras latitudes.
La edad, el estado civil divorciado, el consumo de
alcohol y la actividad física alta en la esfera de ocio fueron
los principales factores asociados a HTA, siendo necesario
políticas en salud que disminuya su impacto en la población
y la promoción de estilo de vida beneficioso.Introduction: Hypertension (HT) is a modifiable risk factor
for the presence of vascular, cardiac and renal diseases,
with important repercussions in the global public
health. Epidemiological data of this pathology on rural
populations in Ecuador are limited, so the objective of this
study was to evaluate the prevalence and associated factors
with HT in the rural population of Quingeo, Ecuador.
Materials and Methods: A cross-sectional analytical
study with multi-stage random sampling was conducted
in 530 individuals over 18 years old of both sexes, who
were applied a complete medical history. Blood pressure
was classified according to JNC7 criteria. A logistic regression
model was constructed to determine associated factors
with HT.
Results: The prevalence of HT was 16.2% (n=86), 16.3%
(n= 51) in women and 16.1% (n= 35) in men. At the same
time, age was associated with the prevalence of HT (χ2=
4.276; p<0.001), with higher percentages to older adults.
The main associated factors with HT were the middle-age
adults (OR= 3.41; 95%CI= 1.73-6.09, p<0.001), older
adult (OR= 4.98; 95%CI: 2.24-10.09; p<0.001), alcohol
consumption (OR= 2.56; 95%CI: 1.02-6.44, p=0.044),
divorced civil status (OR=5.16; IC95%:1.53-17.44,
p=0.008) and high level of leisure time physical activity
(OR= 0.20; 95%CI:0.08-0.94, p=0.040).
Conclusion: There is a low prevalence of HT in the rural
population of Quingeo compared to other latitudes.
Aging, divorced marital status, alcohol consumption and
high physical activity in leisure-time were the main associated
factors with HT, being necessary policies in health
that diminish their impact on the population and promotion
a beneficial lifestyle

Social, educational and economic inequities in the rural population of Quingeo, Ecuador
Introducción: Ecuador es un país latinoamericano que ha
sufrido diversos cambios epidemiológicos en las últimas
décadas, acorde a la transición demográfica, caracterizada
por un aumento de ingresos monetarios, educación, empleo
y mejoras en los servicio de salud. Esto ha llevado a una
adopción de hábitos perjudiciales que han concordado con
el aumento de la mortalidad por enfermedades crónicas no
transmisibles. A su vez, estas enfermedades se han relacionado
con diversos factores socioeconómicos especialmente
en poblaciones en vías de desarrollo, por lo que el objetivo
de este estudio fue evaluar las características sociodemográficas
de la población de Quingeo, Ecuador.
Metodología: Se realizó un estudio de campo, descriptivo
y transversal que incluyó a 530 individuos adultos de la parroquia
rural de Quingeo, perteneciente al Cantón de Cuenca,
Ecuador. Se les aplicó una historia clínica completa indagándose
en los factores sociodemográficos como: edad, sexo,
estado civil, estatus socioeconómico, nivel de educación, estatus
laboral y ocupación.
Resultados: Del total de individuos evaluados, el 59,06%
(n=313) perteneció al sexo femenino. La edad media de la
muestra fue de 43,21±18,20 años, siendo el grupo etario más
frecuente los adultos jóvenes (57,9%; n=307). Los sujetos
fueron principalmente casados (55,7%; n=295), desempleados
(67,9%; n=360), con estudios de educación primaria (47,7%,
n=253), y con un nivel socioeconómico de clase obrera (49,1%,
n=260). A su vez, los individuos en extrema pobreza tenían
mayores proporciones de desempleo y de analfabetismo.
Conclusión: Este estudio constituye un reporte sobre la situación
socioeconómica y demográfica actual de la población
rural de Quingeo, en donde los individuos con estratos socioeconómicos
bajos presentaron un alto porcentaje de desempleo
y bajo nivel educativo, por lo que deben abordarse estas
situaciones para mejorar la promoción de la salud.Introduction: Ecuador is a Latin American country that has
undergone several epidemiological changes in the last decades,
according to the demographic transition, characterized
by an increase in monetary income, education, employment
and improvements in health service. This has led to an
adoption of harmful habits that have related to increase mortality
from chronic non-communicable diseases. In turn, these
diseases have been related to various socioeconomic factors
especially in developing country, so the purpose of this study
was to evaluate the sociodemographic characteristics of the
population of Quingeo, Ecuador.
Methodology: A field, descriptive and cross-sectional study
was carried out involving 530 individuals from the rural parish
of Quingeo, in the Canton of Cuenca, Ecuador. It was
applied a complete medical history including sociodemographic
factors such as: age, sex, marital status, socioeconomic
status, educational level, work status and occupation.
Results: Of the overall of individuals, 59.06% (n = 313) belonged
to the female sex. The mean age of the sample was
43.21 ± 18.20 years, with the youngest being the most frequent
age group (57.9%; n = 307). The subjects were mainly
married, with primary education (47.7%, n = 253), and belonging
to working class (49.1%, n = 260). At the same time,
individuals in extreme poverty had higher unemployment and
illiteracy rates.
Conclusion: This study presents a report of the actual socioeconomic
and demographic situation of the rural population
of Quingeo, where individuals with low socioeconomic strata
presented a high percentage of unemployment and low educational
leve
