    Modelling cell cycle synchronisation in networks of coupled radial glial cells

    Radial glial cells play a crucial role in the embryonic mammalian brain. Their proliferation is thought to be controlled, in part, by ATP mediated calcium signals. It has been hypothesised that these signals act to locally synchronise cell cycles, so that clusters of cells proliferate together, shedding daughter cells in uniform sheets. In this paper we investigate this cell cycle synchronisation by taking an ordinary differential equation model that couples the dynamics of intracellular calcium and the cell cycle and extend it to populations of cells coupled via extracellular ATP signals. Through bifurcation analysis we show that although ATP mediated calcium release can lead to cell cycle synchronisation, a number of other asynchronous oscillatory solutions including torus solutions dominate the parameter space and cell cycle synchronisation is far from guaranteed. Despite this, numerical results indicate that the transient and not the asymptotic behaviour of the system is important in accounting for cell cycle synchronisation. In particular, quiescent cells can be entrained on to the cell cycle via ATP mediated calcium signals initiated by a driving cell and crucially will cycle in near synchrony with the driving cell for the duration of neurogenesis. This behaviour is highly sensitive to the timing of ATP release, with release at the G1/S phase transition of the cell cycle far more likely to lead to near synchrony than release during mid G1 phase. This result, which suggests that ATP release timing is critical to radial glia cell cycle synchronisation may help to understand normal and pathological brain development

    A One Health Framework for the Evaluation of Rabies Control Programmes: A Case Study from Colombo City, Sri Lanka

    <div><p>Background</p><p>One Health addresses complex challenges to promote the health of all species and the environment by integrating relevant sciences at systems level. Its application to zoonotic diseases is recommended, but few coherent frameworks exist that combine approaches from multiple disciplines. Rabies requires an interdisciplinary approach for effective and efficient management.</p><p>Methodology/Principal Findings</p><p>A framework is proposed to assess the value of rabies interventions holistically. The economic assessment compares additional monetary and non-monetary costs and benefits of an intervention taking into account epidemiological, animal welfare, societal impact and cost data. It is complemented by an ethical assessment. The framework is applied to Colombo City, Sri Lanka, where modified dog rabies intervention measures were implemented in 2007. The two options included for analysis were the control measures in place until 2006 (“baseline scenario”) and the new comprehensive intervention measures (“intervention”) for a four-year duration. Differences in control cost; monetary human health costs after exposure; Disability-Adjusted Life Years (DALYs) lost due to human rabies deaths and the psychological burden following a bite; negative impact on animal welfare; epidemiological indicators; social acceptance of dogs; and ethical considerations were estimated using a mixed method approach including primary and secondary data. Over the four years analysed, the intervention cost US $1.03 million more than the baseline scenario in 2011 prices (adjusted for inflation) and caused a reduction in dog rabies cases; 738 DALYs averted; an increase in acceptability among non-dog owners; a perception of positive changes in society including a decrease in the number of roaming dogs; and a net reduction in the impact on animal welfare from intermediate-high to low-intermediate.</p><p>Conclusions</p><p>The findings illustrate the multiple outcomes relevant to stakeholders and allow greater understanding of the value of the implemented rabies control measures, thereby providing a solid foundation for informed decision-making and sustainable control.</p></div

    Is mindfulness Buddhist? (and why it matters).

    Modern exponents of mindfulness meditation promote the therapeutic effects of "bare attention"--a sort of non-judgmental, non-discursive attending to the moment-to-moment flow of consciousness. This approach to Buddhist meditation can be traced to Burmese Buddhist reform movements of the first half of the 20th century, and is arguably at odds with more traditional Theravāda Buddhist doctrine and meditative practices. But the cultivation of present-centered awareness is not without precedent in Buddhist history; similar innovations arose in medieval Chinese Zen (Chan) and Tibetan Dzogchen. These movements have several things in common. In each case the reforms were, in part, attempts to render Buddhist practice and insight accessible to laypersons unfamiliar with Buddhist philosophy and/or unwilling to adopt a renunciatory lifestyle. In addition, these movements all promised astonishingly quick results. And finally, the innovations in practice were met with suspicion and criticism from traditional Buddhist quarters. Those interested in the therapeutic effects of mindfulness and bare attention are often not aware of the existence, much less the content, of the controversies surrounding these practices in Asian Buddhist history

    I examine the post-Independence role of Sri Lanka’s origin story, revealing the ways in which the foundational myth of the Mahavamsa functions as a conflicted site of cultural ‘encompassment’ (Kapferer) in literary and political discourse. Through an analysis of the fiction of Tissa Abeysekara, Carl Muller and the assassinated president Ranasinghe Premadasa, I show how the scripting of this myth in fiction reveals a shift from the celebratory drives of nationalism to a critique of patriotism in a way that both reflects and anticipates a broader paradigmatic shift in the construction of belonging and the outsider found in post-war Sri Lanka

    The spirit and the gifts: Dako, Benjamin Morrell and cargo in the Vitiaz trading area, New Guinea

    In 1830 an American trader, Benjamin Morrell, abducted Dako, the son of a prominent leader from Uneapa Island in the Bismarck Sea, took him to New York and, four years later, returned him to Uneapa. Dako's encounter with America and his return provides insight into the region half a century before colonization, and in particular into local mytho-practical knowledge at that time. This enables us to discern subsequent transformations. Myths concerning an origin spirit and guardian of the dead, Pango, which then dominated Uneapa cosmology have since ‘disappeared’. This, we argue, is not because Pango has been superseded or suppressed, but because the parallel ‘white’ world over which the mytho-practical Pango presided has become ever more manifest as Uneapa has been drawn into a colonial, post-colonial and globalised world. Today, Pango refers predominantly to white people. Islander's experience of American ‘Pango’ was a shocking event at the time, but we show how trading with Pango established transformatory possibilities for reciprocal trading relations with the dead which remain the concern of today's Cult movement on the island

    Modelling the coupling between intracellular calcium release and the cell cycle during cortical brain development

    Most neocortical neurons formed during embryonic brain development arise from radial glial cells which communicate, in part, via ATP mediated calcium signals. Although the intercellular signalling mechanisms that regulate radial glia proliferation are not well understood, it has recently been demonstrated that ATP dependent intracellular calcium release leads to an increase of nearly 100% in overall cellular proliferation. It has been hypothesised that cytoplasmic calcium accelerates entry into S phase of the cell cycle and/or acts to recruit otherwise quiescent cells onto the cell cycle. In this paper we study this cell cycle acceleration and recruitment by forming a differential equation model for ATP mediated calcium-cell cycle coupling via Cyclin D in a single radial glial cell. Bifurcation analysis and numerical simulations suggest that the cell cycle period depends only weakly on cytoplasmic calcium. Therefore the accelerative impact of calcium on the cell cycle can only account for a small fraction of the large increase in proliferation observed experimentally. Crucially however, our bifurcation analysis reveals that stable fixed point and stable limit cycle solutions can coexist, and that calcium dependent Cyclin D dynamics extend the oscillatory region to lower Cyclin D synthesis rates, thus rendering cells more susceptible to cycling. This supports the hypothesis that cycling glial cells recruit quiescent cells (in G0 phase) onto the cell cycle, via a calcium signalling mechanism, and that this may be the primary means by which calcium augments proliferation rates at the population scale.     Karma, morality, and evil

    The doctrine of karma has been praised as a rational and morally edifying explanatory response to the existence of evil and apparent injustice in the world. Critics have attacked it as a morally misguided dogma that distorts one's vision of reality. This essay, after outlining the traditional doctrine, examines three criticisms that have been central to recent debates: firstly, that the doctrine offers no practical guidance; second, that it faces a dilemma between free will and fatalism; and third, that it involves a morally repugnant form of blaming victims for their own misfortunes. Possible responses are considered, the depth of the disagreement is highlighted, and a morally significant difference between alternative ways of articulating the belief in karma is analyzed
