    Density-dependence and within-host competition in a semelparous parasite of leaf-cutting ants

    BACKGROUND: Parasite heterogeneity and within-host competition are thought to be important factors influencing the dynamics of host-parasite relationships. Yet, while there have been many theoretical investigations of how these factors may act, empirical data is more limited. We investigated the effects of parasite density and heterogeneity on parasite virulence and fitness using four strains of the entomopathogenic fungus, Metarhizium anisopliae var. anisopliae, and its leaf-cutting ant host Acromyrmex echinatior as the model system. RESULTS: The relationship between parasite density and infection was sigmoidal, with there being an invasion threshold for an infection to occur (an Allee effect). Although spore production was positively density-dependent, parasite fitness decreased with increasing parasite density, indicating within-host scramble competition. The dynamics differed little between the four strains tested. In mixed infections of three strains the infection-growth dynamics were unaffected by parasite heterogeneity. CONCLUSIONS: The strength of within-host competition makes dispersal the best strategy for the parasite. Parasite heterogeneity may not have effected virulence or the infection dynamics either because the most virulent strain outcompeted the others, or because the interaction involved scramble competition that was impervious to parasite heterogeneity. The dynamics observed may be common for virulent parasites, such as Metarhizium, that produce aggregated transmission stages. Such parasites make useful models for investigating infection dynamics and the impact of parasite competition

    A human embryonic kidney 293T cell line mutated at the Golgi -mannosidase II locus

    Disruption of Golgi -mannosidase II activity can result in type II congenital dyserythropoietic anemia and can induce lupus-like autoimmunity in mice. Here, we isolate a mutant human embryonic kidney (HEK) 293T cell line, called Lec36, that displays sensitivity to ricin that lies between the parental HEK 293T cells, whose secreted and membrane-expressed proteins are dominated by complex-type glycosylation, and 293S Lec1 cells, which only produce oligomannose-type N-linked glycans. The stem cell marker, 19A, was transiently expressed in the HEK 293T Lec36 cells, and in parental HEK 293T cells with and without the potent Golgi -mannosidase II inhibitor, swainsonine. Negative-ion nano-electrospray ionization mass spectra of the 19A N-linked glycans from HEK 293T Lec36 and swainsonine-treated HEK 293T cells were qualitatively indistinguishable and, as shown by collision-induced dissociation spectra, dominated by hybrid-type glycosylation. Nucleotide sequencing revealed mutations in each allele of MAN2A1, the gene encoding Golgi -mannosidase II: a point mutation in one allele mapping to the active site and an in-frame deletion of twelve-nucleotides in the other. Expression of wild-type but not the mutant MAN2A1 alleles in Lec36 cells restored processing of the 19A reporter glycoprotein to complex-type glycosylation. The Lec36 cell line will be useful for expressing therapeutic glycoproteins with hybrid-type glycans and provides a sensitive host for detecting mutations in human MAN2A1 causing type II congenital dyserythropoietic anemia

    Dust Formation in Very Massive Primordial Supernovae

    At redshift z>5 Type II supernovae (SNII) are the only known dust sources with evolutionary timescales shorter than the Hubble time. We extend the model of dust formation in the ejecta of SNII by Todini & Ferrara (2001) to investigate the same process in pair-instability supernovae (PISN), which are though to arise from the explosion of the first, metal free, very massive (140-260 Msun) cosmic stars. We find that 15%-30% of the PISN progenitor mass is converted into dust, a value >10 times higher than for SNII; PISN dust depletion factors (fraction of produced metals locked into dust grains) range between 0.3 and 0.7. These conclusions depend very weakly on the mass of the PISN stellar progenitor, which instead affects considerably the composition and size distribution. For the assumed temperature evolution, grain condensation starts 150-200 days after the explosion; the dominant compounds for all progenitor masses are SiO2 and Mg2SiO4 while the contribution of amorphous carbon and magnetite grains grows with progenitor mass; typical grain sizes range between 0.001 and a few 0.1 micron and are always smaller than 1 micron. We give a brief discussion of the implications of dust formation for the IMF evolution of the first stars, cosmic reionization and the intergalactic medium.Comment: 10 pages, 8 figures, accepted for publication in MNRA

    Accuracy of Orthodontic Soft Tissue Prediction Software between Different Ethnicities

    Objective: The objective of this study was to assess the accuracy of the soft tissue prediction module of Dolphin Imaging Software (DIS) in patients requiring extractions as part of the orthodontic treatment plan and compare its accuracy between different ethnicities. Materials and Methods: Initial and final records of 57 patients from three ethnic groups (African Americans, Caucasians, and Hispanics) who completed orthodontic treatment were included for assessment. The identified cases were managed non-surgically with dental extractions. A predictive profile was generated using DIS and compared to post-treatment lateral photographs. Actual and predictive profile photographs were compared using five designated parameters. The assessment parameters were evaluated using a manual protractor. ANOVA was used to compare differences between actual and predicted parameters between the specified groups and ICC was used to assess correlations between the data. Results: Neither ethnicity nor gender had a significant effect on the difference between predicted and final values. No significant difference was noted between the predicted and final images for the nasolabial angle. Significant differences were observed for the mentolabial fold, upper lip to E-line, and lower lip to E-line between predicted and actual images. Additionally, soft tissue convexity was significantly different (p=0.019). Additionally, a clinically significant difference was found for the mentolabial fold. Conclusion: Ethnicity and gender had no impact on the accuracy of predicted and actual image parameters. Overall, DIS demonstrated acceptable accuracy when simulating soft tissue changes after extraction therapy. Additional research on the accuracy of the software is warranted

    Fatal Pancreatic Panniculitis Associated with Acute Pancreatitis: A Case Report

    Pancreatic panniculitis is a rare disease in which necrosis of fat in the panniculus and other distant foci occurs in the setting of pancreatic diseases; these diseases include acute and chronic pancreatitis, pancreatic carcinoma, pseudocyst, and other pancreatic diseases. This malady is manifested as tender erythematous nodules on the legs, buttock, or trunk. Histopathologically, it shows the pathognomonic findings of focal subcutaneous fat necrosis and ghost-like anucleated cells with a thick shadowy wall. We herein report a case of fatal pancreatic panniculitis that was associated with acute pancreatitis in a 50-yr-old man. He presented with a 3-week history of multiple tender skin nodules, abdominal pain and distension. Laboratory and radiologic findings revealed acute pancreatitis, and skin biopsy showed pancreatic panniculitis. Despite intensive medical care, he died of multi-organ failure 3 weeks after presentation

    Finite Element Analysis of Free Vibration of the Delaminated Composite Plate with Variable Kinematic Multilayered Plate Elements

    Composite laminates are prone to delamination. Implementation of delamination in the Carrera Unified Formulation frame work using nine noded quadrilateral MITC9 element is discussed in this article. MITC9 element is devoid of shear locking and membrane locking. Delaminated as well as healthy structure is analyzed for free mode vibration. The results from the present work are compared with the available experimental or/and research article or/and the three dimensional finite element simulations. The effect of different kinds and different percentages of area of delamination on the first three natural frequencies of the structure is discussed. The presence of open-mode delamination mode shape for large delaminations within the first three natural frequencies is discussed. Also, the switching of places between the second bending mode, with that of the first torsional mode frequency is discussed. Results obtained from different ordered theories are compared in the presence of delamination. Advantage of layerwise theories as compared to equivalent single layer theories for very large delaminations is stated.     Nanogels of poly(acrylic acid): Uptake and release behavior with fluorescent oligothiophene-labeled bovine serum albumin

    Nanometer-sized poly(acrylic acid) (PAA) hydrogels were synthesized by emulsion polymerization of methyl acrylate and subsequent acidic hydrolysis. The nanohydrogel was characterized by spectroscopic methods (FTIR and H-1-NMR) and scanning probe techniques, and their pH-dependent swelling behavior was studied by dynamic light scattering. To determine the suitability of PAA nanogels as pH-sensitive carriers for biomedical applications, uptake and release of an oligothiophene fluorophore and its albumin conjugated from PAA nanogels were investigated as a function of pH by absorption and photoluminescence measurements.     Deletion of mtorc1 activity in CD4+ T cells is associated with lung fibrosis and increased γδ T cells

    Pulmonary fibrosis is a devastating, incurable disease in which chronic inflammation and dysregulated, excessive wound healing lead to progressive fibrosis, lung dysfunction, and ultimately death. Prior studies have implicated the cytokine IL-17A and Th17 cells in promoting the development of fibrosis. We hypothesized that loss of Th17 cells via CD4-specific deletion of mTORC1 activity would abrogate the development of bleomycin-induced pulmonary fibrosis. However, in actuality loss of Th17 cells led to increased mortality and fibrosis in response to bleomycin. We found that in the absence of Th17 cells, there was continued production of IL-17A by γδT cells. These IL-17A+γδT cells were associated with increased lung neutrophils and M2 macrophages, accelerated development of fibrosis, and increased mortality. These data elucidate the critical role of IL-17A+ γδT cells in promoting chronic inflammation and fibrosis, and reveal a novel therapeutic target for treatment of pulmonary fibrosis
