    Relationship of DAG1 and SERPINA5 Sperm Proteins With Bull Fertility

    Study Description: Semen from 22 dairy bulls was used to evaluate the presence, localization, and quantification of DAG1 and SERPINA5 on sperm. Sperm motility parameters and viability was also evaluated for semen from each bull. Semen from 19 out of the 22 dairy bulls was used for in vitro embryo production (two Low-SCR and one High-SCR were not available for in vitro embryo production). Bulls were classified based on their sire conception rates (SCR) values as High-SCR (SCR \u3e 1.0) or Low-SCR fertility (SCR \u3c -4.0). Low fertility bulls were subdivided based on their blastocyst rate (BL) as High-BL (Low-SCR/High-BL BL ≥ 31%) or Low-BL (Low-SCR/Low-BL BL ≤ 26%), and High-SCR bulls were not subdivided. The GLM procedure in SAS was used with bull as a fixed effect to determine if variance was greater between bulls compared to within bull. Correlations were determined among DAG1 and SERPINA5 concentrations, percentage of tail labeled for SERPINA5, SCR, sperm total motility, progressive motility, and viability, and in vitro embryo produced cleavage rate (CL) and BL. The GLIMMIX procedure of SAS was used to evaluate the relationship of bull field fertility (High- and Low-SCR), and field and in vitro fertility (High-SCR, Low-SCR/High-BL, Low-SCR/Low-BL) classifications with sperm total (TMOT) and progressive (PROG) motility, viability, CL, BL, DAG1 and SERPINA5 relative concentration, and proportion of sperm tail labeled for SERPINA5. Both SERPINA5 and DAG1 were localized on the sperm head; however, SERPINA5 was also localized on the sperm tail. There was greater variance in concentration among bulls compared to within bull for both DAG1 (P \u3c 0.01; 69.4 vs 49.1, respectively) and SERPINA5 (P \u3c 0.01; 325.8 vs 285.4, respectively). There was a positive correlation between concentration of DAG1 and SERPINA5 (P = 0.01; r = 0.54). Concentrations of SERPINA5 were also correlated with CL (P = 0.04; r = 0.48), and percentage of sperm tail labeled for SERPINA5 was correlated with viability (P = 0.05; r = 0.44) and tended to be correlated with CL (P = 0.10; r = 0.39). There was no relationship between SCR or BL rate classifications and DAG1 (P ≥ 0.66), SERPINA5 (P ≥ 0.54), or percentage of sperm tail labeled for SERPINA5 (P ≥ 0.48)

    Trade-offs between the natural environment and recreational infrastructure:A case study about peatlands under different management scenarios

    The importance of peatlands for conservation and provision of public services has been well evidenced in the last years, especially in relation to their contribution to the net zero carbon emission agenda. However, little is known about the importance of recreation relative to conservation and their trade-offs. In this paper we address this knowledge gap by exploring the trade-offs between natural properties of peatlands and recreational infrastructures for different categories of recreationists (walkers, cyclists, anglers, and birdwatchers) of an open heather moors and peatlands landscape. We do so building on a series of management scenarios formulated through participatory methods and applying choice experiment related to an Area of Outstanding Natural Beauty and UNESCO Global Geopark in the UK. Results show a high degree of heterogeneity in landscape preferences across different user groups. Recreationists had a higher appreciation for semi-natural habitats compared to pristine or restored peatland (e.g., land rewetting). Walkers and cyclists were more sensitive to changes in the availability of recreational facilities than to environmental quality, while anglers’ and birdwatchers’ preferences were more aligned with values promoted by restoration policies. Overall, our results point to a potential value conflict between benefits generated by conservation and the benefits valued most by some groups of recreationists. To maximise success conflicts like the one revealed here need to be considered in strategies that provide a central role for peatlands in net zero climate mitigation strategies

    Relationship of field and in vitro fertility of dairy bulls with sperm parameters, including DAG1 and SERPINA5 proteins

    IntroductionSperm interacts with the female reproductive tract and oocyte through proteins, and these cell-to-cell interactions may play a role in sperm fertility. For consideration of a protein as a potential marker of fertility, there must be variability expressed among animals. The proteins dystroglycan (DAG1) and plasma serine protease inhibitor (SERPINA5) have been reported to play a role in cell-to-cell interactions. Thus, the objectives of this study were to characterize the localization and abundance variability of DAG1 and SERPINA5 in bovine sperm, and to investigate the relationship of DAG1 and SERPINA5 with field fertility (i.e., sire conception rate; SCR), in vitro embryo production (IVP), and sperm parameters.Material and methodsDairy bulls (n = 22) were classified as high-SCR (SCR > 1.0) or low-SCR (SCR < –4.0), and good [blastocyst (BL)-by-cleavage (CL) ratio (BL/CL) > 39%] or poor (BL/CL < 38%) BL/CL. Sperm was evaluated for DAG1 and SERPINA5 immunolocalization, and concentration in two separate ejaculates. Variance between bulls compared with within bulls was evaluated using a generalized linear model (GLM) procedure. The relationship of SCR and IVP classification on DAG1 and SERPINA5 concentrations, percentage of tail labeled for SERPINA5, SCR, sperm total and progressive motility, sperm plasma membrane integrity (PMI), CL, BL, and BL/CL were evaluated with the GLIMMIX procedure, and the correlations between these variables were evaluated.ResultsBoth proteins were localized on the sperm head; however, SERPINA5 was also localized on the sperm tail. Concentrations of DAG1 and SERPINA5 on the sperm head were correlated with each other. The percentage of tail labeled for SERPINA5 was correlated with sperm PMI; however, neither protein was associated with SCR or IVP. Thus, when evaluated by immunofluorescent microscopy, DAG1 and SERPINA5 concentrations are variable and are not good fertility markers for bull sperm

    Functional protein rich extracts from bovine and porcine hearts using acid or alkali solubilisation and isoelectric precipitation

    Alkali solubilisation (ALS) was compared with acid solubilisation (ACS) for preparation of protein rich extracts from bovine and porcine hearts. ACS and ALS recovered 51.53%–55.74% of the total protein from bovine and porcine hearts. All extracts were rich in myofibrillar proteins with both treatments resulting in reductions in fat, collagen and cholesterol contents compared with starting materials. At 0% NaCl, ACS and ALS extracts had good gelling properties with the ALS gels having lower % cook loss. While treatments did not affect gel hardness, ACS extracts formed gel networks with higher storage modulus after heating and cooling. At 2% NaCl gel hardness, % cook loss and storage modulus values increased, with greater increases occurring for ACS extracts. The results show that ALS‐ and ACS‐based processes have potential to produce functional ingredients for processed meat products

    Antimicrobial resistance among migrants in Europe: a systematic review and meta-analysis

    BACKGROUND: Rates of antimicrobial resistance (AMR) are rising globally and there is concern that increased migration is contributing to the burden of antibiotic resistance in Europe. However, the effect of migration on the burden of AMR in Europe has not yet been comprehensively examined. Therefore, we did a systematic review and meta-analysis to identify and synthesise data for AMR carriage or infection in migrants to Europe to examine differences in patterns of AMR across migrant groups and in different settings. METHODS: For this systematic review and meta-analysis, we searched MEDLINE, Embase, PubMed, and Scopus with no language restrictions from Jan 1, 2000, to Jan 18, 2017, for primary data from observational studies reporting antibacterial resistance in common bacterial pathogens among migrants to 21 European Union-15 and European Economic Area countries. To be eligible for inclusion, studies had to report data on carriage or infection with laboratory-confirmed antibiotic-resistant organisms in migrant populations. We extracted data from eligible studies and assessed quality using piloted, standardised forms. We did not examine drug resistance in tuberculosis and excluded articles solely reporting on this parameter. We also excluded articles in which migrant status was determined by ethnicity, country of birth of participants' parents, or was not defined, and articles in which data were not disaggregated by migrant status. Outcomes were carriage of or infection with antibiotic-resistant organisms. We used random-effects models to calculate the pooled prevalence of each outcome. The study protocol is registered with PROSPERO, number CRD42016043681. FINDINGS: We identified 2274 articles, of which 23 observational studies reporting on antibiotic resistance in 2319 migrants were included. The pooled prevalence of any AMR carriage or AMR infection in migrants was 25·4% (95% CI 19·1-31·8; I2 =98%), including meticillin-resistant Staphylococcus aureus (7·8%, 4·8-10·7; I2 =92%) and antibiotic-resistant Gram-negative bacteria (27·2%, 17·6-36·8; I2 =94%). The pooled prevalence of any AMR carriage or infection was higher in refugees and asylum seekers (33·0%, 18·3-47·6; I2 =98%) than in other migrant groups (6·6%, 1·8-11·3; I2 =92%). The pooled prevalence of antibiotic-resistant organisms was slightly higher in high-migrant community settings (33·1%, 11·1-55·1; I2 =96%) than in migrants in hospitals (24·3%, 16·1-32·6; I2 =98%). We did not find evidence of high rates of transmission of AMR from migrant to host populations. INTERPRETATION: Migrants are exposed to conditions favouring the emergence of drug resistance during transit and in host countries in Europe. Increased antibiotic resistance among refugees and asylum seekers and in high-migrant community settings (such as refugee camps and detention facilities) highlights the need for improved living conditions, access to health care, and initiatives to facilitate detection of and appropriate high-quality treatment for antibiotic-resistant infections during transit and in host countries. Protocols for the prevention and control of infection and for antibiotic surveillance need to be integrated in all aspects of health care, which should be accessible for all migrant groups, and should target determinants of AMR before, during, and after migration.     IgE allergy diagnostics and other relevant tests in allergy, a World Allergy Organization position paper

    Currently, testing for immunoglobulin E (IgE) sensitization is the cornerstone of diagnostic evaluation in suspected allergic conditions. This review provides a thorough and updated critical appraisal of the most frequently used diagnostic tests, both in vivo and in vitro. It discusses skin tests, challenges, and serological and cellular in vitro tests, and provides an overview of indications, advantages and disadvantages of each in conditions such as respiratory, food, venom, drug, and occupational allergy. Skin prick testing remains the first line approach in most instances; the added value of serum specific IgE to whole allergen extracts or components, as well as the role of basophil activation tests, is evaluated. Unproven, non-validated, diagnostic tests are also discussed. Throughout the review, the reader must bear in mind the relevance of differentiating between sensitization and allergy; the latter entails not only allergic sensitization, but also clinically relevant symptoms triggered by the culprit allergen
