    Refining palaeoenvironmental analysis using integrated quantitative granulometry and palynology

    Accurate palaeoenvironmental analysis is at the heart of producing reliable interpretations and depositional models. This study demonstrates a multivariate statistical approach to facies analysis based on relationships between grain size and quantitative palynology. Our methodology has the advantage that it can be used on small amounts of sample, such as core or well cuttings, as the basis for facies analysis. Proof of concept studies involving the collection of grain-size and palynological datasets from well-exposed outcrops of the Middle Jurassic, Lajas Formation of the Neuquén Basin, Argentina, demonstrate that canonical correspondence analysis can be used to consistently recognize facies and aid in the determination of depositional environments. This study demonstrates the link between depositional facies, grain-size distribution, palynomorph hydrodynamics and assemblage taphonomy of palynomorphs. This knowledge can be transferred into a semi-automated statistical facies prediction technique for the subsurface in complex depositional settings, particularly when calibrated against conventional sedimentary facies analysis.The attached article is the accepted manuscript for this article.     Interventions to Promote More Effective Balance-Recovery Reactions in Industrial Settings: New Perspectives on Footwear and Handrails

    “Change-in-support” balance-recovery reactions that involve rapid stepping or reaching movements play a critical role in preventing falls. Recent geriatrics studies have led to new interventions to improve ability to execute these reactions effectively. Some of these interventions have the potential to reduce fall risk for younger persons working in industrial settings. In this paper, we review research pertaining to two such interventions: 1) balance-enhancing footwear insoles designed to improve stepping reactions, and 2) proximity-triggered handrail cueing systems designed to improve reach-to-grasp reactions. The insole has a raised ridge around the perimeter that is intended to improve balance control by providing increased stimulation of sensory receptors on the footsole in situations where loss of balance may be imminent. The cueing system uses flashing lights and/or verbal prompts to attract attention to the handrail and ensure that the brain registers its location, thereby facilitating more rapid and accurate grasping of the rail if and when sudden loss of balance occurs. Results to date support the efficacy of both interventions in geriatric populations. There is also some evidence that these interventions may improve balance control in younger persons; however, further research is needed to confirm their efficacy in preventing falls in industrial settings

    Induction of the suppressor of cytokine signalling 3 (SOCS-3) gene is vital to the normal control of inflammatory signalling. In order to understand these processes we investigated the role of the proto-oncogene component of the AP-1 transcription factor complex, c-Jun, in the regulation of SOCS-3 gene induction. We found that cyclic AMP stimulation of HUVECs promoted phosphorylation and activation of JNK MAP kinase and its substrate c-Jun. The JNK responsive element of the human SOCS-3 promoter mapped to a putative AP-1 site within 1000 bp of the transcription start site. The PKC inhibitors, GF-109203X, Gö-6983 and Ro-317549, were all found to inhibit AP-1 transcriptional activity, transcriptional activation of this minimal SOCS-3 promoter and SOCS-3 gene induction in HUVECs. Interestingly, Ro-317549 treatment was also found to promote PKC-dependent activation of ERK and JNK MAP kinases and promote JNK-dependent hyper-phosphorylation of c-Jun, whereas GF-109203X and Gö-6983 had little effect. Despite this, all three PKC inhibitors were found to be effective inhibitors of c-Jun DNA-binding activity. The JNK-dependent hyper-phosphorylation of c-Jun in response to Ro-317549 treatment of HUVECs does therefore not interfere with its ability to inhibit c-Jun activity and acts as an effective inhibitor of c-Jun-dependent SOCS-3 gene induction

    Decoupling seasonal fluctuations in fluvial discharge from the tidal signature in ancient deltaic deposits: an example from the Neuquén Basin, Argentina

    Fluvial discharge fluctuations are a fundamental characteristic of almost all modern rivers and can produce distinctive deposits that are rarely described from ancient fluvial or mixed-energy successions. Large-scale outcrops from the Middle Jurassic Lajas Formation (Argentina) expose a well-constrained stratigraphic succession of marginal-marine deposits with a strong fluvial influence and well-known tidal indicators. The studied deposits show decimetre-scale interbedding of coarser- and finer-grained facies with mixed fluvial and tidal affinities. The alternation of these two types of beds forms non-cyclic successions that are interpreted to be the result of seasonal variation in river discharge, rather than regular and predictable changes in current velocity caused by tides. Seasonal bedding is present in bar deposits that form within or at the mouth of minor and major channels. Seasonal bedding is not preserved in channel thalweg deposits, where river flood processes were too powerful, or in floodplain, muddy interdistributary-bay, prodelta and transgressive deposits, where the river signal was weak and sporadic. The identification of sedimentary facies characteristic of seasonal river discharge variations is important for accurate interpretation of ancient deltaic process regime

    Force-Controlled Balance Perturbations Associated with Falls in Older People: A Prospective Cohort Study

    Balance recovery from an unpredictable postural perturbation can be a challenging task for many older people and poor recovery could contribute to their risk of falls. This study examined associations between responses to unpredictable perturbations and fall risk in older people. 242 older adults (80.064.4 years) underwent assessments of stepping responses to multi-directional force-controlled waist-pull perturbations. Participants returned monthly falls calendars for the subsequent 12 months. Future falls were associated with lower force thresholds for stepping in the posterior and lateral but not anterior directions. Those with lower posterior force thresholds for stepping were 68% more likely to fall at home than those with higher force thresholds for stepping. These results suggest that amount of force that can be withstood following an unpredictable balance perturbation predicts future falls in community-dwelling older adults. Perturbations in the posterior direction best discriminated between future fallers and non-fallers

    Potential immunological consequences of pharmacological suppression of gastric acid production in patients with multiple sclerosis

    Corticosteroids are standard treatment for patients with multiple sclerosis experiencing acute relapse. Because dyspeptic pain is a common side effect of this intervention, patients can be given a histamine receptor-2 antagonist, proton pump inhibitor or antacid to prevent or ameliorate this disturbance. Additionally, patients with multiple sclerosis may be taking these medications independent of corticosteroid treatment. Interventions for gastric disturbances can influence the activation state of the immune system, a principal mediator of pathology in multiple sclerosis. Although histamine release promotes inflammation, activation of the histamine receptor-2 can suppress a proinflammatory immune response, and blocking histamine receptor-2 with an antagonist could shift the balance more towards immune stimulation. Studies utilizing an animal model of multiple sclerosis indicate that histamine receptor-2 antagonists potentially augment disease activity in patients with multiple sclerosis. In contrast, proton pump inhibitors appear to favor immune suppression, but have not been studied in models of multiple sclerosis. Antacids, histamine receptor-2 antagonists and proton pump inhibitors also could alter the intestinal microflora, which may indirectly lead to immune stimulation. Additionally, elevated gastric pH can promote the vitamin B12 deficiency that patients with multiple sclerosis are at risk of developing. Here, we review possible roles of gastric acid inhibitors on immunopathogenic mechanisms associated with multiple sclerosis

    Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018.

    Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field
