    Pilot trial of a type I - polarized autologous dendritic cell vaccine incorporating tumor blood vessel antigen-derived peptides in patients with metastatic breast cancer

    Cancer vaccines based on tumor-associated antigens are rarely curative in advanced cancer. This limitation relates to the heterogeneity of cancer due to defects in antigen presentation and altered immunophenotypes. Therefore, another method to promote anti-tumor immunity is to prime T cells against tumor-associated stromal cells. We have reported [1] that IL-12 gene therapy of established HLA-A2neg B16 melanomas in HLA-A2+ transgenic mice resulted in CD8+ T cell-mediated immunity against the host HLA-A2+ stromal cells within the tumor microenvironment (TME). We have also shown [2] that vaccines based on a subset of tumor blood vessel antigen (TBVA)-derived peptides (DLK1, EphA2, HBB, NRP1, PDGFRβ, RGS5 or TEM1) prevented HLA-A2neg MC38 tumor establishment and promoted the regression of tumors in HLA-A2+ mice by CD8+ T cell targeting of HLA-A2+ pericytes and vascular endothelial cells in the TME. Based on this pre-clinical data, we propose to undertake a Susan G. Komen -funded (IIR13261822) clinical trial of chemo-immunotherapy using the immunomodulatory drug gemcitabine with a dendritic cell vaccine pulsed with six HLA-A2-presented TBVA-derived peptides (DLK1310-318, EphA2883-891, HBB31-39, NRP1433-441, RGS55-13 and TEM1691-700) in 30 HLA-A2+ patients with metastatic breast cancer.     Silicate weathering and carbon cycle controls on the Oligocene-Miocene transition glaciation

    Changes in both silicate weathering rates and organic carbon burial have been proposed as drivers of the transient “Mi-1” glaciation event at the Oligocene-Miocene transition (OMT; ~23 Ma). However detailed geochemical proxy data are required to test these hypotheses. Here we present records of Li/Ca, Mg/Ca, Cd/Ca, U/Ca, δ18O, δ13C, and shell weight in planktonic foraminifera from marine sediments spanning the OMT in the equatorial Atlantic Ocean. Li/Ca values increase by 1 μmol/mol across this interval. We interpret this to indicate a ~20% increase in silicate weathering rates, which would have lowered atmospheric CO2, potentially forcing the Antarctic glaciation circa 23 Ma. δ13C of thermocline dwelling planktonic foraminifera track the global increase in seawater δ13C across the OMT and during the Mi-1 event, hence supporting a hypothesized global increase in organic carbon burial rates. High δ13C previously measured in epipelagic planktonic foraminifera and high Cd/Ca ratios during Mi-1 are interpreted to represent locally enhanced primary productivity, stimulated by increased nutrients supply to surface waters. The fingerprint of high export production and associated organic carbon burial at this site is found in reduced bottom water oxygenation (inferred from high foraminiferal U/Ca), and enhanced respiratory dissolution of carbonates, characterised by reduced foraminiferal shell weight. Replication of our results elsewhere would strengthen the case that weathering-induced CO2 sequestration preconditioned climate for Antarctic ice sheet growth across the OMT and increased burial of organic carbon acted as a feedback that intensified cooling at this time

    Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018.

    Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field

    Circular polarization of transmitted light by sapphirinidae copepods.

    Circularly polarized light, rare in the animal kingdom, has thus far been documented in only a handful of animals. Using a rotating circular polarization (CP) analyzer we detected CP in linearly polarized light transmitted through epipelagic free living Sapphirina metallina copepods. Both left and right handedness of CP was detected, generated from specific organs of the animal's body, especially on the dorsal cephalosome and prosome. Such CP transmittance may be generated by phase retardance either in the muscle fibers or in the multilayer membrane structure found underneath the cuticle. Although the role, if any, played by circularly polarized light in Sapphirinidae has yet to be clarified, in other animals it was suggested to take part in mate choice, species recognition, and other forms of communication.Planktonic Sapphirinidae copepods were found to circularly polarize the light passing through them. Circular polarization may be created by unique, multilayered features of the membrane structure found under their cuticle or by organized muscle fibers
