    CLU, CR1 and PICALM genes associate with Alzheimer's-related senile plaques

    Introduction APOE is the strongest risk gene for sporadic Alzheimer's disease (AD) so far. Recent genome wide association studies found links for sporadic AD with CLU and CR1 involved in Aβ clearance, and PICALM affecting intracellular trafficking. Methods We investigated the associations of senile plaques (SP) and neurofibrillary tangles (NFT) with the proposed risk genes and APOE, in the Tampere Autopsy Study (TASTY) series (603 cases), a sample of the general population (0 to 97 yrs), who died out-of-hospital. Results Age and the APOEε4 allele associated strongly with all phenotypes of SP, as expected. In age and APOEε4 adjusted analyses, compared to the most common homozygous genotype, burnt out SP were more common among carriers of the C-allele of CLU, whereas the T-allele of PICALM and C-allele of CR1 were linked with lower SP coverage. We found no significant associations between any of the genetic variants and NFT. Conclusions Marginal effects from CLU, CR1 and PICALM suggest that these genes have minimal effects on the development of AD lesions.BioMed Central Open acces

    Vigor, vitality and seed dormancy of Avena sativa cultivars in a long-term experiment

    Vigor, vitality and seed dormancy of 14 Finnish cultivars of Avena sativa in room temperature were studied in a 22-year laboratory experiment. These parameters were studied by measuring morphological and physical characteristics of seeds and by basic germination and enzymatic tests 1, 4, 6, 11, 16, 21 and 22 years after seed harvesting in 1989. Methylene blue, Congo red and 2,3,5-triphenyltetrazolium chloride (TZ) tests were used to estimate seed quality and changes in vitality over time. Seed vitality clearly decreased in all cultivars during the experiment. The mean vitality declined from 96.3% (one year after harvest) to zero at the end. Vitality according to the TZ test was higher than indicated by the basic germination test. The mean vitality loss was 4.6% per year, but there were clear differences between cultivars. The decrease in vitality correlated with loss in seed weight. Clear signs of deepening dormancy were observed. Seed age is an important factor infl uencing vitality and dormancy. Vitality loss of seeds led to deep dormancy. The appearance, water uptake and imbibition of the seeds remained normal until the end. Ageing, vitality loss and dormancy are concluded to be expressions of genes. It is possible that in the future electronic simulation methods will be developed that will enable accurate estimation of oat seed quality without laboratory tests

    Bunyaviruses Affect Growth, Sporulation, and Elicitin Production in Phytophthora cactorum

    Phytophthora cactorum is an important oomycetous plant pathogen with numerous host plant species, including garden strawberry (Fragaria × ananassa) and silver birch (Betula pendula). P. cactorum also hosts mycoviruses, but their phenotypic effects on the host oomycete have not been studied earlier. In the present study, we tested polyethylene glycol (PEG)-induced water stress for virus curing and created an isogenic virus-free isolate for testing viral effects in pair with the original isolate. Phytophthora cactorum bunya-like viruses 1 and 2 (PcBV1 & 2) significantly reduced hyphal growth of the P. cactorum host isolate, as well as sporangia production and size. Transcriptomic and proteomic analyses revealed an increase in the production of elicitins due to bunyavirus infection. However, the presence of bunyaviruses did not seem to alter the pathogenicity of P. cactorum. Virus transmission through anastomosis was unsuccessful in vitro

    Consciousness, Action Selection, Meaning and Phenomenic Anticipation

    Phenomenal states are generally considered the ultimate sources of intrinsic motivation for autonomous biological agents. In this article, we will address the issue of the necessity of exploiting these states for the design and implementation of robust goal-directed artificial systems. We will provide an analysis of consciousness in terms of a precise definition of how an agent "understands" the informational flows entering the agent and its very own action possibilities. This abstract model of consciousness and understanding will be based in the analysis and evaluation of phenomenal states along potential future trajectories in the state space of the agents. This implies that a potential strategy to follow in order to build autonomous but still customer-useful systems is to embed them with the particular, ad hoc phenomenality that captures the system-external requirements that define the system usefulness from a customer-based, requirements-strict engineering viewpoint

    CRP gene variation affects early development of Alzheimer's disease-related plaques

    Introduction We used the Tampere Autopsy Study (TASTY) series (n = 603, age 0-97 yrs), representing an unselected population outside institutions, to investigate the pathogenic involvement of inflammation in Alzheimer's disease-related lesions. Methods We studied senile plaque (SP), neurofibrillary tangles (NFT) and SP phenotype associations with 6 reported haplotype tagging single nucleotide polymorphisms (SNPs) in the CRP gene. CRP and Aβ immunohistochemistry was assessed using brain tissue microarrays. Results In multivariate analyses (age- and APOE-adjusted), non-neuritic SP were associated with the high-CRP TA-genotype (3.0% prevalence) of rs3091244 and CA-genotype (10.8%) of rs3093075 compared to common genotypes. Conversely, the low-CRP C allele (39.3%) of rs2794521 reduced the risk of harbouring early non-neuritic SP, compared to the TT genotype. CRP haplotype TAGCC (high) associated with non-neuritic SP, whereas haplotype CCGCC offered protection. TT genotypes (high) of rs3091244 and rs1130864 were associated with CRP staining. There were no associations between SNPs or haplotypes and NFT. CRP staining of the hippocampal CA1/2 region correlated with Aβ staining. Conclusions CRP gene variation affects early SP development in prodromal Alzheimer's disease, independent of APOE genotype.BioMed Central Open acces

    Molecular evolution of viral multifunctional proteins: the case of Potyvirus HC-Pro

    [EN] Our knowledge on the mode of evolution of the multifunctional viral proteins remains incomplete. To tackle this problem, here, we have investigated the evolutionary dynamics of the potyvirus multifunctional protein HC-Pro, with particular focus on its functional domains. The protein was partitioned into the three previously described functional domains, and each domain was analyzed separately and assembled. We searched for signatures of adaptive evolution and evolutionary dependencies of amino acid sites within and between the three domains using the entire set of available potyvirus sequences in GenBank. Interestingly, we identified strongly significant patterns of co-occurrence of adaptive events along the phylogenetic tree in the three domains. These patterns suggest that Domain I, whose main function is to mediate aphid transmission, has likely been coevolving with the other two domains, which are involved in different functions but all requiring the capacity to bind RNA. By contrast, episodes of positive selection on Domains II and III did not correlate, reflecting a trade-off between their evolvability and their evolutionary dependency likely resulting from their functional overlap.     What do we know about the α/β for prostate cancer?

    Since last decade, the debate on the parameter which reflects prostate cancer sensitivity to fractionation in a radiotherapy treatment, the α/β, has become extensive. Unlike most tumors, the low labeling indices (LI) and large potential doubling time that characterize the prostate tumor led some authors to consider that it may behave as a late responding tissue. So far, the existing studies with regard to this subject point to a low value of α/β, around 2.7 Gy, which may be considered as a therapeutic gain in relation to surrounding normal tissues by using fewer and larger fractions. The aim of this paper is to review several estimates that have been made in the last few years regarding the prostate cancer α/β both from clinical and experimental data, as well as the set of factors that have potentially influenced these evaluations

    Toward Self-Referential Autonomous Learning of Object and Situation Models

    Most current approaches to scene understanding lack the capability to adapt object and situation models to behavioral needs not anticipated by the human system designer. Here, we give a detailed description of a system architecture for self-referential autonomous learning which enables the refinement of object and situation models during operation in order to optimize behavior. This includes structural learning of hierarchical models for situations and behaviors that is triggered by a mismatch between expected and actual action outcome. Besides proposing architectural concepts, we also describe a first implementation of our system within a simulated traffic scenario to demonstrate the feasibility of our approach
