    Towards the Industrialization of New MDO Methodologies and Tools for Aircraft Design

    An overall summary of the Institute of Technology IRT Saint Exupery MDA-MDO project (Multi-Disciplinary Analysis - Multidisciplinary Design Optimization) is presented. The aim of the project is to develop efficient capabilities (methods, tools and a software platform) to enable industrial deployment of MDO methods in industry. At IRT Saint Exupery, industrial and academic partners collaborate in a single place to the development of MDO methodologies; the advantage provided by this mixed organization is to directly benefit from both advanced methods at the cutting edge of research and deep knowledge of industrial needs and constraints. This paper presents the three main goals of the project: the elaboration of innovative MDO methodologies and formulations (also referred to as architectures in the literature 1) adapted to the resolution of industrial aircraft optimization design problems, the development of a MDO platform featuring scalable MDO capabilities for transfer to industry and the achievement of a simulation-based optimization of an aircraft engine pylon with industrial Computational Fluid Dynamics (CFD) and Computational Structural Mechanics (CSM) tools

    International audienceThere are two competing theories about the interrelationships of craniates: the cyclostome theory assumes that lampreys and hagïŹshes are a clade, the cyclostomes, whose sister group is the jawed vertebrates (gnathostomes); the vertebrate theory assumes that lampreys and gnathostomes are a clade, the vertebrates, whose sister group is hagïŹshes. The vertebrate theory is best supported by a number of unique anatomical and physiological characters. Molecular sequence data from 18S and 28S rRNA genes rather support the cyclostome theory, but mtDNA sequence of Myxine glutinosa rather supports the vertebrate theory. Additional molecular data are thus needed to elucidate this three-taxon problem. We determined the complete nucleotide sequence of the mtDNA of the lamprey Lampetra ïŹ‚uviatilis. The mtDNA of L. ïŹ‚uviatilis possesses the same genomic organization as Petromyzon marinus, which validates this gene order as a synapomorphy of lampreys. The mtDNA sequence of L. ïŹ‚uviatilis was used in combination with relevant mtDNA sequences for an approach to the hagïŹsh/lamprey relationships using the max- imum-parsimony, neighbor-joining, and maximum-likelihood methods. Although trees compatible with our present knowledge of the phylogeny of craniates can be reconstructed by using the three methods, the data collected do not support the vertebrate or the cyclostome hypothesis. The present data set does not allow the resolution of this three-taxon problem, and new kinds of data, such as nuclear DNA sequences, need to be collected

    Rapid Improvement after Starting Elexacaftor–Tezacaftor–Ivacaftor in Patients with Cystic Fibrosis and Advanced Pulmonary Disease

    International audienceRationale: Elexacaftor-tezacaftor-ivacaftor is a CFTR (cystic fibrosis [CF] transmembrane conductance regulator) modulator combination, developed for patients with CF with at least one Phe508del mutation. Objectives: To evaluate the effects of elexacaftor-tezacaftor- ivacaftor in patients with CF and advanced respiratory disease. Methods: A prospective observational study, including all patients aged â©Ÿ12 years and with a percent-predicted FEV1 (ppFEV1) <40 who initiated elexacaftor-tezacaftor-ivacaftor from December 2019 to August 2020 in France was conducted. Clinical characteristics were collected at initiation and at 1 and 3 months. Safety and effectiveness were evaluated by September 2020. National-level transplantation and mortality figures for 2020 were obtained from the French CF and transplant centers and registries. Measurements and Main Results: Elexacaftor-tezacaftor- ivacaftor was initiated in 245 patients with a median (interquartile range) ppFEV1 = 29 (24-34). The mean (95% confidence interval) absolute increase in the ppFEV1 was +15.1 (+13.8 to +16.4; P < 0.0001), and the mean (95% confidence interval) in weight was +4.2 kg (+3.9 to +4.6; P < 0.0001). The number of patients requiring long-term oxygen, noninvasive ventilation, and/or enteral tube feeding decreased by 50%, 30%, and 50%, respectively (P < 0.01). Although 16 patients were on the transplant waiting list and 37 were undergoing transplantation evaluation at treatment initiation, only 2 received a transplant, and 1 died. By September 2020, only five patients were still on the transplantation path. Compared with the previous 2 years, a twofold decrease in the number of lung transplantations in patients with CF was observed in 2020, whereas the number of deaths without transplantation remained stable. Conclusions: In patients with advanced disease, elexacaftor-tezacaftor-ivacaftor is associated with rapid clinical improvement, often leading to the indication for lung transplantation being suspended
