Histone deacetylase adaptation in single ventricle heart disease and a young animal model of right ventricular hypertrophy.
BackgroundHistone deacetylase (HDAC) inhibitors are promising therapeutics for various forms of cardiac diseases. The purpose of this study was to assess cardiac HDAC catalytic activity and expression in children with single ventricle (SV) heart disease of right ventricular morphology, as well as in a rodent model of right ventricular hypertrophy (RVH).MethodsHomogenates of right ventricle (RV) explants from non-failing controls and children born with a SV were assayed for HDAC catalytic activity and HDAC isoform expression. Postnatal 1-day-old rat pups were placed in hypoxic conditions, and echocardiographic analysis, gene expression, HDAC catalytic activity, and isoform expression studies of the RV were performed.ResultsClass I, IIa, and IIb HDAC catalytic activity and protein expression were elevated in the hearts of children born with a SV. Hypoxic neonatal rats demonstrated RVH, abnormal gene expression, elevated class I and class IIb HDAC catalytic activity, and protein expression in the RV compared with those in the control.ConclusionsThese data suggest that myocardial HDAC adaptations occur in the SV heart and could represent a novel therapeutic target. Although further characterization of the hypoxic neonatal rat is needed, this animal model may be suitable for preclinical investigations of pediatric RV disease and could serve as a useful model for future mechanistic studies
Bioaccumulation of heavy metals in water, sediment and periwinkle (Tympanotonus fuscatus var radula) from the Elechi Creek, Niger Delta
The accumulation of three heavy metals; chromium (Cr), cadmium (Cd) and lead (Pb) in periwinkle (Tympanotonus fuscatus var radula; shell and soft tissues), water and sediment collected from four stations along Elechi Creek course was studied. Elechi Creek receives effluents discharges from heavily industrialized and highly populated settlements. The water, sediment and periwinkles were processed and analyzed for heavy metals and the results showed that the sediment concentrated more heavy metals than the water while the periwinkles accumulated more of these metals than the sediment. Cr was the highest concentrated heavy metals in both the normal and depurated periwinkles. The biological concentration factor (BCF) revealed that these periwinkles have high potential to concentrate heavy metals in their shells and soft tissues, and it is directly proportionate to their sizes. However, the observed heavy metals concentrations in these animals are below the recommended limits for human consumption. This study therefore advocates environmental surveillance of this creek in order to achieve good sediment quality and contaminant-free periwinkles for safe human healt
Growth response and survival of Heterobranchus longifilis fingerlings fed at different feeding frequencies
In an effort to utilize feed efficiently, promote growth, increase survival and reduce labour costs associated with feeding, Heterobranchus longifilis ingerlings were placed on one of four feeding frequencies; once/day, twice/day; once every other day, and twice every other day for 56 days. Theywere fed with a commercial floating diet containing 40% crude protein level. They were reared in twelve 14 L plastic aquaria, with 3 aquaria/treatment under laboratory conditions. Each aquarium contained 10 fingerlings. The growth parameters except relative growth rate (RGR) were significantly different for fishes receiving the different treatments. Survival was not significantly different among treatments except for fingerlings fed once every other day. Induced growth of phytoplankton using two fertilizers (NPK and agrolyser) under laboratory conditions
An investigation was carried out on the ability of two types of fertilizer; agrolyser micronutrient and NPK (nitrogen, phosphorus, potassium)-macronutrient in inducing phytoplankton growth for sixteen days under laboratory conditions. The parameters studied were abundance, species diversity and occurrence of the phytoplankton. Twelve 14 L plastics tanks labeled A, B, C and D was used. Each letter represented a fertilizer treatment (three replicates): A, 0.27 g agrolyser; B, 200 g NPK; C, 200 g agrolyser + 0.27 g NPK; and D which is the control. Before the application of these fertilizers, water samples were taken for phytoplankton analyses using 250 ml wide mouth plastic containers. All the tanks were allowed to be naturally inoculated for 3 days. Samples were collected for phytoplankton and physicochemical parameters at an interval of 3 days except temperature which was measured twice daily (morning and evening). A total of 1244 phytoplankton made up of 20 species, belonging to three families: Bacillariophyceae (diatoms), Cyanophyceae (blue-green algae) and Euglenophyceae (euglenophytes) were observed. Out of these algae, tanks C had 51.7%, B 33.2%, A 10.6% and D 4.5% phytoplankters. In the various groups of tanks, the orders of abundance, species diversity and percentage distribution of species of algae were C>B>A>D, C>B>A=D and C>B>D>A, respectively. This paper intends to provide preliminary information to fish farmers that agrolyser should be combined with a macronutrient fertilizer for better fertilization of pond.Keywords: Induced growth, phytoplankton, fertilizer, NPK, agrolyse
A four-helix bundle stores copper for methane oxidation
Methane-oxidising bacteria (methanotrophs) require large quantities of copper for the membrane-bound (particulate) methane monooxygenase (pMMO). Certain methanotrophs are also able to switch to using the iron-containing soluble MMO (sMMO) to catalyse methane oxidation, with this switchover regulated by copper. MMOs are Nature’s primary biological mechanism for suppressing atmospheric levels of methane, a potent greenhouse gas. Furthermore, methanotrophs and MMOs have enormous potential in bioremediation and for biotransformations producing bulk and fine chemicals, and in bioenergy, particularly considering increased methane availability from renewable sources and hydraulic fracturing of shale rock. We have discovered and characterised a novel copper storage protein (Csp1) from the methanotroph Methylosinus trichosporium OB3b that is exported from the cytosol, and stores copper for pMMO. Csp1 is a tetramer of 4-helix bundles with each monomer binding up to 13 Cu(I) ions in a previously unseen manner via mainly Cys residues that point into the core of the bundle. Csp1 is the first example of a protein that stores a metal within an established protein-folding motif. This work provides a detailed insight into how methanotrophs accumulate copper for the oxidation of methane. Understanding this process is essential if the wide-ranging biotechnological applications of methanotrophs are to be realised. Cytosolic homologues of Csp1 are present in diverse bacteria thus challenging the dogma that such organisms do not use copper in this location
Altered brain gene expression but not steroid biochemistry in a genetic mouse model of neurodevelopmental disorder
Background
The 39,XY*O mouse, which lacks the orthologues of the ADHD and autism candidate genes STS (steroid sulphatase) and ASMT (acetylserotonin O-methyltransferase), exhibits behavioural phenotypes relevant to developmental disorders. The neurobiology underlying these phenotypes is unclear, although there is evidence for serotonergic abnormalities in the striatum and hippocampus.
Methods
Using microarray and quantitative gene expression analyses, and gas chromatography–mass spectrometry, we compared brain gene expression and steroid biochemistry in wildtype (40,XY) and 39,XY*O adult mice to identify non-obvious genetic and endocrine candidates for between-group differences in behaviour and neurochemistry. We also tested whether acute STS inhibition by COUMATE in wildtype (40,XY) adult male mice recapitulated any significant gene expression or biochemical findings from the genetic comparison. Data were analysed by unpaired t-test or Mann Whitney U-test depending on normality, with a single factor of KARYOTYPE.
Results
Microarray analysis indicated seven robust gene expression differences between the two groups (Vmn2r86, Sfi1, Pisd-ps1, Tagap1, C1qc, Metap1d, Erdr1); Erdr1 and C1qc expression was significantly reduced in the 39,XY*O striatum and hippocampus, whilst the expression of Dhcr7 (encoding 7-dehydrocholesterol reductase, a modulator of serotonin system development), was only reduced in the 39,XY*O hippocampus. None of the confirmed gene expression changes could be recapitulated by COUMATE administration. We detected ten free, and two sulphated steroids in 40,XY and 39,XY*O brain; surprisingly, the concentrations of all of these were equivalent between groups.
Conclusions
Our data demonstrate that the mutation in 39,XY*O mice: i) directly disrupts expression of the adjacent Erdr1 gene, ii) induces a remarkably limited suite of downstream gene expression changes developmentally, with several of relevance to associated neurobehavioural phenotypes and iii) does not elicit large changes in brain steroid biochemistry. It is possible that individuals with STS/ASMT deficiency exhibit a similarly specific pattern of gene expression changes to the 39,XY*O mouse, and that these contribute towards their abnormal neurobiology. Future work may focus on whether complement pathway function, mitochondrial metabolism and cholesterol biosynthesis pathways are perturbed in such subjects
Search for direct pair production of the top squark in all-hadronic final states in proton-proton collisions at s√=8 TeV with the ATLAS detector
The results of a search for direct pair production of the scalar partner to the top quark using an integrated luminosity of 20.1fb−1 of proton–proton collision data at √s = 8 TeV recorded with the ATLAS detector at the LHC are reported. The top squark is assumed to decay via t˜→tχ˜01 or t˜→ bχ˜±1 →bW(∗)χ˜01 , where χ˜01 (χ˜±1 ) denotes the lightest neutralino (chargino) in supersymmetric models. The search targets a fully-hadronic final state in events with four or more jets and large missing transverse momentum. No significant excess over the Standard Model background prediction is observed, and exclusion limits are reported in terms of the top squark and neutralino masses and as a function of the branching fraction of t˜ → tχ˜01 . For a branching fraction of 100%, top squark masses in the range 270–645 GeV are excluded for χ˜01 masses below 30 GeV. For a branching fraction of 50% to either t˜ → tχ˜01 or t˜ → bχ˜±1 , and assuming the χ˜±1 mass to be twice the χ˜01 mass, top squark masses in the range 250–550 GeV are excluded for χ˜01 masses below 60 GeV
A systematic review of the effectiveness and cost-effectiveness of peer education and peer support in prisons.
BACKGROUND: Prisoners experience significantly worse health than the general population. This review examines the effectiveness and cost-effectiveness of peer interventions in prison settings. METHODS: A mixed methods systematic review of effectiveness and cost-effectiveness studies, including qualitative and quantitative synthesis was conducted. In addition to grey literature identified and searches of websites, nineteen electronic databases were searched from 1985 to 2012. Study selection criteria were: Population: Prisoners resident in adult prisons and children resident in Young Offender Institutions (YOIs). INTERVENTION: Peer-based interventions Comparators: Review questions 3 and 4 compared peer and professionally led approaches. OUTCOMES: Prisoner health or determinants of health; organisational/ process outcomes; views of prison populations. STUDY DESIGNS: Quantitative, qualitative and mixed method evaluations. RESULTS: Fifty-seven studies were included in the effectiveness review and one study in the cost-effectiveness review; most were of poor methodological quality. Evidence suggested that peer education interventions are effective at reducing risky behaviours, and that peer support services are acceptable within the prison environment and have a positive effect on recipients, practically or emotionally. Consistent evidence from many, predominantly qualitative, studies, suggested that being a peer deliverer was associated with positive effects. There was little evidence on cost-effectiveness of peer-based interventions. CONCLUSIONS: There is consistent evidence from a large number of studies that being a peer worker is associated with positive health; peer support services are also an acceptable source of help within the prison environment and can have a positive effect on recipients.
