Ovogénesis y formación de la puesta de los peces invasores Pterois miles y Pterois volitans
The Indo-Pacific lionfish, Pterois miles and P. volitans, have recently invaded the U.S. east coast and the Caribbean and pose a significant threat to native reef fish communities. Few studies have documented reproduction in pteroines from the Indo-Pacific. This study provides a description of oogenesis and spawn formation in P. miles and P. volitans collected from offshore waters of North Carolina, U.S.A and the Bahamas. Using histological and laboratory observations, we found no differences in reproductive biology between P. miles and P. volitans. These lionfish spawn buoyant eggs that are encased in a hollow mass of mucus produced by specialized secretory cells of the ovarian wall complex. Oocytes develop on highly vascularized peduncles with all oocyte stages present in the ovary of spawning females and the most mature oocytes placed terminally, near the ovarian lumen. Given these ovarian characteristics, these lionfish are asynchronous, indeterminate batch spawners and are thus capable of sustained reproduction throughout the year when conditions are suitable. This mode of reproduction could have contributed to the recent and rapid establishment of these lionfish in the northwestern Atlantic and Caribbean.Los peces Indo-Pacíficos, Pterois miles y P. volitans, han invadido recientemente la costa este de los Estados Unidos y el Caribe y representan una significativa amenaza a las comunidades nativas de peces coralinos. Unos pocos estudios han documentado la reproducción en peces de la subfamilia Pteroinae del Indo-Pacífico. Este estudio presenta la descripción de la ovogénesis y la formación de puesta en P. miles y P. volitans recolectados desde aguas a mar abierto de Carolina del Norte, U.S.A, y las Bahamas. Mediante el uso de observaciones histológicas y de laboratorio, encontramos que no había diferencias en la biología reproductiva entre P. miles y P. volitans. Estas especies desovan huevos flotantes que están encerrados en una masa hueca de moco producida por células secretoras especializadas del complejo de la pared del ovario. Los ovocitos se desarrollan en pedúnculos altamente especializados, estando todos los estadios de los ovocitos presentes en el ovario de las hembras en puesta, y los ovocitos más maduros se localizan en la zona terminal, cerca del lumen del ovario. Dadas estas características del ovario, estas especies son asincrónicas, ponedores secuenciales indeterminados y son, por tanto, capaces de tener una reproducción sostenida a lo largo del año cuando las condiciones son adecuadas. An ovary transcriptome for all maturational stages of the striped bass (Morone saxatilis), a highly advanced perciform fish
<p>Abstract</p> <p>Background</p> <p>The striped bass and its relatives (genus <it>Morone</it>) are important fisheries and aquaculture species native to estuaries and rivers of the Atlantic coast and Gulf of Mexico in North America. To open avenues of gene expression research on reproduction and breeding of striped bass, we generated a collection of expressed sequence tags (ESTs) from a complementary DNA (cDNA) library representative of their ovarian transcriptome.</p> <p>Results</p> <p>Sequences of a total of 230,151 ESTs (51,259,448 bp) were acquired by Roche 454 pyrosequencing of cDNA pooled from ovarian tissues obtained at all stages of oocyte growth, at ovulation (eggs), and during preovulatory atresia. Quality filtering of ESTs allowed assembly of 11,208 high-quality contigs ≥ 100 bp, including 2,984 contigs 500 bp or longer (average length 895 bp). Blastx comparisons revealed 5,482 gene orthologues (E-value < 10<sup>-3</sup>), of which 4,120 (36.7% of total contigs) were annotated with Gene Ontology terms (E-value < 10<sup>-6</sup>). There were 5,726 remaining unknown unique sequences (51.1% of total contigs). All of the high-quality EST sequences are available in the National Center for Biotechnology Information (NCBI) Short Read Archive (GenBank: <ext-link ext-link-id="SRX007394" ext-link-type="gen">SRX007394</ext-link>). Informative contigs were considered to be abundant if they were assembled from groups of ESTs comprising ≥ 0.15% of the total short read sequences (≥ 345 reads/contig). Approximately 52.5% of these abundant contigs were predicted to have predominant ovary expression through digital differential display <it>in silico </it>comparisons to zebrafish (<it>Danio rerio</it>) UniGene orthologues. Over 1,300 Gene Ontology terms from Biological Process classes of Reproduction, Reproductive process, and Developmental process were assigned to this collection of annotated contigs.</p> <p>Conclusions</p> <p>This first large reference sequence database available for the ecologically and economically important temperate basses (genus <it>Morone</it>) provides a foundation for gene expression studies in these species. The predicted predominance of ovary gene expression and assignment of directly relevant Gene Ontology classes suggests a powerful utility of this dataset for analysis of ovarian gene expression related to fundamental questions of oogenesis. Additionally, a high definition Agilent 60-mer oligo ovary 'UniClone' microarray with 8 × 15,000 probe format has been designed based on this striped bass transcriptome (eArray Group: Striper Group, Design ID: 029004).</p

Enzyme-Linked Immunosorbent Assay (ELISA) of Vitellogenin in Temperate Basses (Genus Morone): Plasma and In Vitro Analyses
Blood levels of the egg yolk precursor vi-
tellogenin (VTG) can be used as a definitive marker for
the onset and progress of maturation in female teleosts.
In the present study, an enzyme-linked immunosorbent
assay (ELISA) was developed to measure VTG in blood
plasma from three species of temperate basses. The an-
tigen capture, competitive ELISA is based on a rabbit
antiserum raised against striped bass Morone saxatilis
VTG and uses purified striped bass VTG as standard and
in the final antigen capture step. The assay was validated
for detecting VTG in the plasma of maturing female
striped bass, white perch M. americana, and white bass
M. chrysops. Serial dilutions of blood plasma from vi-
tellogenic females of all three species yielded VTG
curves that paralleled the standard curve in the ELISA,
whereas no cross reactivity was observed for plasma
obtained from males of any Morone species. The work-
ing range of the ELISA was 33–1,118 ng/mL (90–10%
of binding), and the intra- and interassay coefficients of
variation (100 3 SD/mean) at 50% binding were 3.8%
(N 5 20) and 5.94% (N 5 4), respectively. Complete
recovery (detection) in the ELISA was verified for a
known quantity of VTG added to male striped bass plas-
ma. Changes in plasma VTG concentrations during the
annual reproductive cycle of female striped bass were
measured both by ELISA and an established radial im-
munodiffusion assay (RIDA) based on the same anti-
serum and standard. Vitellogenin was detected in ma-
turing females 7–8 months prior to spawning and the
correlation between individual VTG values measured by
ELISA and the RIDA was very high (r2 5 0.95). The
highly sensitive and precise VTG ELISA should allow
aquaculture and fisheries biologists to evaluate the gen-
der and maturational status of individual fish of any
Morone species during most of the year. Finally, VTG
was detected by ELISA in incubation medium following
culture of white perch liver fragments with 1 3 1026 M
estradiol-17b, providing the basis for an in vitro method
to study the physiology and toxicology of vitellogenesis

Variable Sodium Absorption in a Low-Extinction Type Ia Supernova
Recent observations have revealed that some Type Ia supernovae exhibit
narrow, time-variable Na I D absorption features. The origin of the absorbing
material is controversial, but it may suggest the presence of circumstellar gas
in the progenitor system prior to the explosion, with significant implications
for the nature of the supernova progenitors. We present the third detection of
such variable absorption, based on six epochs of high-resolution spectroscopy
of the Type Ia supernova SN 2007le from Keck and the HET. The data span ~3
months, from 5 days before maximum light to 90 days after maximum. We find that
one component of the Na D absorption lines strengthened significantly with
time, indicating a total column density increase of ~2.5 x 10^12 cm^-2. The
changes are most prominent after maximum light rather than at earlier times
when the UV flux from the SN peaks. As with SN 2006X, we detect no change in
the Ca II H&K lines over the same time period, rendering line-of-sight effects
improbable and suggesting a circumstellar origin for the absorbing material.
Unlike the previous two SNe exhibiting variable absorption, SN 2007le is not
highly reddened (E_B-V = 0.27 mag), also pointing toward circumstellar rather
than interstellar absorption. Photoionization models show that the data are
consistent with a dense (10^7 cm^-3) cloud or clouds of gas located ~0.1 pc
from the explosion. These results broadly support the single-degenerate
scenario previously proposed to explain the variable absorption, with mass loss
from a nondegenerate companion star responsible for providing the circumstellar
gas. We also present tentative evidence for narrow Halpha emission associated
with the SN, which will require followup observations at late times to confirm.

TFOS European Ambassador meeting: Unmet needs and future scientific and clinical solutions for ocular surface diseases
The mission of the Tear Film & Ocular Surface Society (TFOS) is to advance the research, literacy, and educational aspects of the scientific field of the tear film and ocular surface. Fundamental to fulfilling this mission is the TFOS Global Ambassador program. TFOS Ambassadors are dynamic and proactive experts, who help promote TFOS initiatives, such as presenting the conclusions and recommendations of the recent TFOS DEWS II™, throughout the world. They also identify unmet needs, and propose future clinical and scientific solutions, for management of ocular surface diseases in their countries. This meeting report addresses such needs and solutions for 25 European countries, as detailed in the TFOS European Ambassador meeting in Rome, Italy, in September 2019
Repeatability and validity of a standardised maximal step-up test for leg function-a diagnostic accuracy study
<p>Abstract</p> <p>Background</p> <p>Objectively assessed physical performance is a strong predictor for morbidity and premature death and there is an increasing interest in the role of sarcopenia in many chronic diseases. There is a need for robust and valid functional tests in clinical practice. Therefore, the repeatability and validity of a newly developed maximal step up test (MST) was assessed.</p> <p>Methods</p> <p>The MST, assessing maximal step-up height (MSH) in 3-cm increments, was evaluated in 60 healthy middle-aged subjects, 30 women and 30 men. The repeatability of MSH and the correlation between MSH and isokinetic knee extension peak torque (IKEPT), self-reported physical function (SF-36, PF), patient demographics and self-reported physical activity were investigated.</p> <p>Results</p> <p>The repeatability between occasions and between testers was 6 cm. MSH (range 12-45 cm) was significantly correlated to IKEPT, (<it>r </it>= 0.68, <it>P </it>< 0.001), SF-36 PF score, (<it>r </it>= 0.29, <it>P </it>= 0.03), sex, age, weight and BMI. The results also show that MSH above 32 cm discriminates subjects in our study with no limitation in self-reported physical function.</p> <p>Conclusions</p> <p>The standardised MST is considered a reliable leg function test for clinical practice. The MSH was related to knee extension strength and self-reported physical function. The precision of the MST for identification of limitations in physical function needs further investigation.</p
Snow Control - An RCT protocol for a web-based self-help therapy to reduce cocaine consumption in problematic cocaine users
Background: Cocaine use has increased in most European countries, including Switzerland, and many states worldwide. The international literature has described treatment models that target the general population. In addition to supplying informative measures at the level of primary and secondary prevention, the literature also offers web-based self-help tools for problematic substance users, which is in line with tertiary prevention. Such programs, however, have been primarily tested on individuals with problematic alcohol and cannabis consumption, but not on cocaine-dependent individuals.
Methods/Design: This paper presents the protocol of a randomised clinical trial to test the effectiveness of a web-based self-help therapy to reduce cocaine use in problematic cocaine users. The primary outcome is severity of cocaine dependence. Secondary outcome measures include cocaine craving, consumption of cocaine and other substances of abuse in the past month, and changes in depression characteristics. The therapy group will receive a 6-week self-help therapy to reduce cocaine consumption based on methods of Cognitive Behavioural Therapy, principles of Motivational Interviewing and self-control practices. The control group will be presented weekly psycho-educative information with a quiz. The predictive validity of participant characteristics on treatment retention and outcome will be explored.
Discussion: To the best of our knowledge, this will be the first randomised clinical trial to test the effectiveness of online self-help therapy to reduce or abstain from cocaine use. It will also investigate predictors of outcome and retention. This trial is registered at Current Controlled Trials and is traceable as NTR-ISRCTN93702927
Genome-wide association and HLA fine-mapping studies identify risk loci and genetic pathways underlying allergic rhinitis
Allergic rhinitis is the most common clinical presentation of allergy, affecting 400 million people worldwide, with increasing incidence in westernized countries1,2. To elucidate the genetic architecture and understand the underlying disease mechanisms, we carried out a meta-analysis of allergic rhinitis in 59,762 cases and 152,358 controls of European ancestry and identified a total of 41 risk loci for allergic rhinitis, including 20 loci not previously associated with allergic rhinitis, which were confirmed in a replication phase of 60,720 cases and 618,527 controls. Functional annotation implicated genes involved in various immune pathways, and fine mapping of the HLA region suggested amino acid variants important for antigen binding. We further performed genome-wide association study (GWAS) analyses of allergic sensitization against inhalant allergens and nonallergic rhinitis, which suggested shared genetic mechanisms across rhinitis-related traits. Future studies of the identified loci and genes might identify novel targets for treatment and prevention of allergic rhinitis
Sensitivity to heat in MS patients: a factor strongly influencing symptomology - an explorative survey
<p>Abstract</p> <p>Background</p> <p>Many individuals diagnosed with Multiple Sclerosis (MS) are sensitive to increased body temperature, which has been recognized as correlating with the symptom of fatigue. The need to explore this association has been highlighted. The aim of this study was to investigate the occurrence of heat sensitivity and its relations to disease course, disability, common MS-related symptoms and ongoing immunosuppressive treatments among individuals 65 years of age or younger diagnosed with MS.</p> <p>Methods</p> <p>A cross-sectional designed survey was undertaken. A questionnaire was sent to MS-patients with an Expanded Disability Status Score (EDSS) in the interval of 0-6.5 and who were between 20 and 65 years of age, living in an eastern region of Sweden (n = 334). Besides occurrence of heat sensitivity (Yes/No) and corresponding questions, the Fatigue Severity Scale (FSS), the MS-related symptom checklist and the Perceived Deficit Questionnaire (PDQ) were included. Data were analysed in relation to data level using Chi-square, Mann Whitney U-test, and Student's t-test. Pearson's and Spearman's correlations were calculated. In the logistic regression analyses (enter) dichotomized MS-symptoms were used as dependent variables, and EDSS, disease-course, time since onset, heat-sensitivity, age and sex (female/male) were independent variables. In the linear regression analyses, enter, mean FSS and summarized PDQ were entered as dependent variables and EDSS, disease-course, time since onset, heat sensitivity, age and sex (female/male) were independent variables.</p> <p>Results</p> <p>Of the responding patients (n = 256), 58% reported heat sensitivity. The regression analyses revealed heat sensitivity as a significant factor relating not only to fatigue (p < 0.001), but also to several other common MS symptoms such as pain (p < 0.001), concentration difficulties (p < 0.001), and urination urgency (p = 0.009).</p> <p>Conclusions</p> <p>Heat sensitivity in MS patients is a key symptom that is highly correlated with disabling symptoms such as fatigue, pain, concentration difficulty and urination urgency.</p
