    Methanol in W3(H2O) and Surrounding Regions

    We present the results of an interferometric study of 38 millimeter-wave lines of CH3OH in the region around the water maser source W3(H2O) and a region extending about 30" to the south and west of the hydroxyl maser source W3(OH). The methanol emitting region around W3(H2O) has an extent of 2.0" x 1.2" (4400 x 2600 AU). The density is of order 1.e7 cm-3, sufficient to thermalize most of the methanol lines. The kinetic temperature is approximately 140 K and the methanol fractional abundance greater than 1.e-6, indicative of a high degree of grain mantle evaporation. The W3(H2O) source contains sub-structure, with peaks corresponding to the TW source and Wyrowski's B/C, separated by 2500 AU in projection. The kinematics are consistent with these being distinct protostellar cores in a wide binary orbit and a dynamical mass for the region of a few tens of Mo. The extended methanol emission to the southwest of W3(OH) is seen strongly only from the lowest excitation lines and from lines known elsewhere to be class I methanol masers, namely the 84.5 GHz 5(-1)-4(0)E and 95.2 GHz 8(0)-7(1)A+ lines. Within this region there are two compact clumps, which we denote as swA and swB, each about 15" (0.16 pc projected distance) away from W3(OH). Excitation analysis of these clumps indicates the presence of lines with inverted populations but only weak amplification. The sources swA and swB appear to have kinetic temperatures of order 50-100 K and densities of order 1.e5 - 1.e6 cm-3. The methanol fractional abundance for the warmer clump is of order 1.e-7, suggestive of partial grain mantle evaporation.     Towards an approach for analysing external representations created during sensemaking using generative grammar

    During sensemaking, users often create external representations to help them make sense of what they know, and what they need to know. In doing so, they necessarily adopt or construct some form of representational language using the tools at hand. By describing such languages implicit in representations we believe that we are better able to describe and differentiate what users do and better able to describe and differentiate interfaces that might support them. Drawing on approaches to the analysis of language, and in particular, Mann and Thompson’s Rhetorical Structure Theory, we analyse the representations that users create to expose their underlying ‘visual grammar’. We do this in the context of a user study involving evidential reasoning. Participants were asked to address an adapted version of IEEE VAST 2011 mini challenge 3 (interpret a potential terrorist plot implicit in a set of news reports). We show how our approach enables the unpacking of the heterogeneous and embedded nature of user-generated representations and allows us to show how visual grammars evolve and become more complex over time in response to evolving sensemaking needs

    Pluralitas Agama dalam Keluarga Jawa

    Dalam masyarakat Jawa terdapat pemahaman dan pemaknaan sendiri terhadap agama yaitu â€agami ageming ajiâ€. Artinya apa pun agama yang dipeluk sama saja karena semua agama mengajarkan keselamatan. Oleh sebab itu menjadi sebuah fenomena menarik di kalangan masyarakat Jawa karena mereka cenderung lebih toleran dalam menyikapi perbedaaan dan keragaman beragama. Salah satu contoh masyarakat yang menghargai pluralitas agama adalah masyarakat Desa Getas Kaloran Temanggung. Tujuan penelitian ini adalah untuk menjelaskan tentang sejumlah keluarga yang dapat menerima pluralitas agama dan toleransi terhadap pluralitas agama dalam keluarga Jawa. Tulisan ini merupakan hasil penelitian yang menggunakan pendekatan deskriptif kualitatif. Subyek penelitian adalah masyarakat Desa Getas yang memiliki keragaman agama dalam keluarganya. Berdasarkan hasil penelitian dapat disimpulkan bahwa masyarakat Desa Getas dapat menerima pluralitas agama karena menurut mereka agama adalah urusan pribadi seseorang jadi tidak ada pihak yang dapat memaksakan suatu keyakinan kepada individu lain. Pluralitas agama tersebut tidak menimbulkan masalah berarti karena masyarakat memiliki derajat toleransi yang tinggi antar anggota keluarga, yang ditunjukkan melalui saling menghargai dan mengormati dan tidak mencampuri urusan keagamaan orang lain, serta saling membantu antar anggota keluarga untuk memperlancar kegiatan ibadah masing – masing. In Javanese community there is a specific principle on the meaning of religion, namely â€agami ageming ajiâ€. This pilosophy means whatever religion people believe, it doesn’t matter because they all teach salvation. This is an interesting phenomenon among the Javanese community because they tend to be tolerant in dealing with differences and diversity of religion that happen in one household. The objective of this article is to discuss the practices of religious tolerance found in a rural community of Getas, Kaloran, Temanggung Central Java. Techniques of data collection is done by interviews and observation. The study subjects were villagers of Getas, which has a diversity of religion in families. Based on the research results, it can be concluded that the villagers embrace a tradition of religious pluralism because they think religion is one’s personal affairs so that no party can impose a conviction for another individual. The plurality of religion does not cause significant problems because the public has a high degree of tolerance among family members, which is demonstrated through mutual respect and attitude not to interfere in religious affairs of others, and mutual help among family members to facilitate the worship activities of their relatives

    A Novel RNAi Lethality Rescue Screen to Identify Regulators of Adipogenesis

    Adipogenesis, the differentiation of fibroblast-like mesenchymal stem cells into mature adipocytes, is tightly regulated by a complex cascade of transcription factors, including the nuclear receptor Peroxisome proliferator activator receptor γ (PPARγ). RNAi-mediated knock down libraries may present an atractive method for the identification of additional adipogenic factors. However, using in vitro adipogenesis model systems for high-throughput screening with siRNA libraries is limited since (i) differentiation is not homogeneous, but results in mixed cell populations, and (ii) the expression levels (and activity) of adipogenic regulators is highly dynamic during differentiation, indicating that the timing of RNAi-mediated knock down during differentiation may be extremely critical. Here we report a proof-of-principle for a novel RNAi screening method to identify regulators of adipogenesis that is based on lethality rescue rather than differentiation, using microRNA expression driven by a PPARγ responsive RNA polymerase II promoter. We validated this novel method through screening of a dedicated deubiquitinase knock down library, resulting in the identification of UCHL3 as an essential deubiquitinase in adipogenesis. This system therefore enables the identification of novel genes regulating PPARγ-mediated adipogenesis in a high-throughput setting

    Assessing the carcinogenic potential of low-dose exposures to chemical mixtures in the environment: the challenge ahead.

    Lifestyle factors are responsible for a considerable portion of cancer incidence worldwide, but credible estimates from the World Health Organization and the International Agency for Research on Cancer (IARC) suggest that the fraction of cancers attributable to toxic environmental exposures is between 7% and 19%. To explore the hypothesis that low-dose exposures to mixtures of chemicals in the environment may be combining to contribute to environmental carcinogenesis, we reviewed 11 hallmark phenotypes of cancer, multiple priority target sites for disruption in each area and prototypical chemical disruptors for all targets, this included dose-response characterizations, evidence of low-dose effects and cross-hallmark effects for all targets and chemicals. In total, 85 examples of chemicals were reviewed for actions on key pathways/mechanisms related to carcinogenesis. Only 15% (13/85) were found to have evidence of a dose-response threshold, whereas 59% (50/85) exerted low-dose effects. No dose-response information was found for the remaining 26% (22/85). Our analysis suggests that the cumulative effects of individual (non-carcinogenic) chemicals acting on different pathways, and a variety of related systems, organs, tissues and cells could plausibly conspire to produce carcinogenic synergies. Additional basic research on carcinogenesis and research focused on low-dose effects of chemical mixtures needs to be rigorously pursued before the merits of this hypothesis can be further advanced. However, the structure of the World Health Organization International Programme on Chemical Safety 'Mode of Action' framework should be revisited as it has inherent weaknesses that are not fully aligned with our current understanding of cancer biology

    The microphytobenthos of Königshafen — spatial and seasonal distribution on a sandy tidal flat

    A microphytobenthic species composition of a tidal flat in the northern Wadden Sea was analysed regarding cell numbers and biomass (in carbon units). The three sampling sites differed in tidal inundation from 15 cm to about 90 cm water depth at high tide. The sediment was sandy at all three stations. A cluster analysis revealed a separation of the benthic diatoms into three areas: aNereis-Corophium-belt, a seagrass-bed and theArenicola-flat. Small epipsammic diatoms were most abundant and dominated the microalgal biomass. A microphytobenthic “spring bloom” even started beneath the ice cover of the flat in January. Lowest values of cell numbers and biomass of benthic microalgae were found in summer. Highest values were measured in the uppermost area (Nereis-Corophium-belt), and only here was an autumnal increase of benthic microalgae found. Further cluster analysis within each of the three areas revealed seasonal differences although the majority of species were present all year round.
