    One case, four approaches: The application of psychotherapeutic approaches in sport psychology.

    Sport and exercise psychology practitioners tasked with service provision within any environment can decide which framework(s) they draw upon to inform their applied work. However, the similarities and differences between psychotherapeutic approaches are under represented in current literature. Therefore, this paper brings together practitioners from four dominant psychotherapeutic approaches to address one specific hypothetical case. Four different cognitive behavioral approaches are outlined, namely rational emotive behavior therapy (REBT), cognitive therapy (CT), schema therapy (ST), and acceptance and commitment therapy (ACT). Each practitioner outlines their approach and proceeds to address the case by covering assessment, intervention and evaluation strategies that are specific to their approach. Similarities and differences across the approaches are discussed and implications for practice are put forth. Finally, two further practitioners introduce motivational interviewing (MI) as an additional framework to foster the working alliance

    A Large Scale Survey of NGC1333

    We observed the clustered star forming complex NGC1333 with the BIMA and FCRAO telescopes in the transitions HCO+(1-0) and N2H+(1-0) over an 11'x11' area with resolution ~10" (0.015pc). The N2H+ emission follows very closely the submillimeter dust continuum emission, while HCO+ emission appears more spatially extended and also traces outflows. We have identified 93 N2H+ cores using the CLUMPFIND algorithm, and we derive N2H+ core masses between 0.05 and 2.5M_sun, with uncertainties of a factor of a few, dominated by the adopted N2H+ abundance. From a comparison with virial masses, we argue that most of these N2H+ cores are likely to be bound, even at the lowest masses, suggesting that the cores do not trace transient structures, and implies the entire mass distribution consists of objects that can potentially form stars. We find that the mass distribution of N2H+ cores resembles the field star IMF, which suggests that the IMF is locked in at the pre-stellar stage of evolution. We find that the N2H+ cores associated with stars identified from Spitzer infrared images have a flat mass distribution. This might be because lower mass cores lose a larger fraction of their mass when forming a star. Even in this clustered environment, we find no evidence for ballistic motions of the cores relative to their lower density surroundings traced by isotopic CO emission, though this conclusion must remain tentative until the surroundings are observed at the same high resolution as the N2H+.Comment: 35 pages, 13 figure

    The "Ca2+ switch" model with cultured Madin-Darby canine kidney (MDCK) cells is useful in studying the biogenesis of epithelial polarity and junction formation and provides insight into early steps in the morphogenesis of polarized epithelial tissues. When extracellular Ca2+ in the medium is changed from less than 5 microM to 1.8 mM, MDCK cells rapidly change from a nonpolarized state exhibiting little cell-cell contact (with the apical membrane and junctional proteins largely within the cell) to a polarized state with well-formed tight junctions and desmosomes. To examine the role of intracellular Ca2+ in the development of polarity and junctions, we made continuous spectrofluorimetric measurements of intracellular Ca2+ during the "switch," using the fluorescent indicator fura-2. Intracellular Ca2+ increased greater than 10-fold during the switch and gave a complex pattern of increase, decrease, and stabilization. In contrast, intracellular pH [monitored with 29,79-bis(2-carboxyethyl)-5(and 6)-carboxyfluorescein (BCECF)] did not change during the period studied. When intracellular Ca2+ curves in several cells were compared, considerable heterogeneity in the rate of increase of intracellular Ca2+ levels and in peak levels was evident, perhaps reflecting the heterogeneity among cells in establishing junctions and polarity. The heterogeneity of the process was confirmed by digital imaging of intracellular Ca2+ and was present even in a "clonal" line of MDCK cells, indicating the heterogeneity was intrinsic to the process and not simply a function of slight genetic variation within the population of MDCK cells. In pairs of cells that had barely established cell-cell contact, often one cell exhibited a much greater increase in intracellular Ca2+ than the other cell in the pair. At the site of cell-cell contact, an apparent localized change (an increase over the basal level) in intracellular Ca2+ was frequently present and occasionally appeared to extend beyond the point of cell-cell contact. Since the region of cell-cell contact is also the site where junctions form and where vesicles containing apical membranes fuse during the development of polarity, we postulate a role for global and local changes in intracellular Ca2+ in these events

    An investigation of lucky imaging techniques

DOI: 10.1111/j.1365-2966.2009.15249.xWe present an empirical analysis of the effectiveness of frame selection (also known as Lucky Imaging) techniques for high resolution imaging. A high-speed image recording system has been used to observe a number of bright stars. The observations were made over a wide range of values of D/r0 and exposure time. The improvement in Strehl ratio of the stellar images due to aligning frames and selecting the best frames was evaluated as a function of these parameters. We find that improvement in Strehl ratio by factors of 4 to 6 can be achieved over a range of D/r0 from 3 to 12, with a slight peak at D/r0 ~ 7. The best Strehl improvement is achieved with exposure times of 10 ms or less but significant improvement is still obtained at exposure times as long as 640 ms. Our results are consistent with previous investigations but cover a much wider range of parameter space. We show that Strehl ratios of >0.7 can be achieved in appropiate conditions whereas previous studies have generally shown maximum Strehl ratios of ~0.3. The results are in reasonable agreement with the simulations of Baldwin et al. (2008).Peer reviewe

    Building More Resilient Culture Collections: A Call for Increased Deposits of Plant-Associated Bacteria

    Biological collections preserve our past, while helping protect our future and increase future knowledge. Plant bacterial culture collections are our security for domestic and global biosecurity. This feature article will provide an introduction to the global position of plant bacterial collections. The role of collections in monitoring plant pathogenic bacteria will be explored through the presentation of five cases studies. These case studies demonstrate why culture collections were imperative for the outcome in each situation. We discuss what we believe should be the best practices to improve microbial preservation and accessioning rates, and why plant bacterial culture collections must increase deposits to be prepared for future emerging pathogens. This is not only the case for global culture collections, but on a much bigger scale, our future scientific successes, our biosecurity decisions and responses, and our knowledge are contingent upon preserving our valuable bacterial strains. It is hoped that once you read this article, you will see the need to deposit your strains in registered public collections and make a concerted effort to build better bacterial culture collections with us

    Phylogenomic analysis supports the transfer of 20 pathovars from Xanthomonas campestris into Xanthomonas euvesicatoria

    The Gram-negative bacterial genus Xanthomonas includes numerous infra-specific taxa known as pathovars, which are defined primarily on host range and disease symptoms. With the advent of molecular sequence data, many pathovars have been transferred from X. campestris into other Xanthomonas species to better harmonise taxonomy and phylogeny. We performed whole-genome shotgun sequencing on pathotype strains of the following X. campestris pathovars: blepharidis, carissae, clerodendri, convolvuli, coriandri, daturae, euphorbiae, fici, heliotropii, ionidii, lawsoniae, mirabilis, obscurae, paulliniae, pennamericanum, spermacoces, uppalii, vernoniae, viegasii and zingibericola. These genomes showed more than 98% average nucleotide identity with the type-strain of X. euvesicatoria and less than 88% with the type-strain of X. campestris. We propose the transfer of these pathovars into X. euvesicatoria and present an emended species description for X. euvesicatoria
