Evolution of sex-specific pace-of-life syndromes: genetic architecture and physiological mechanisms
Sex differences in life history, physiology, and behavior are nearly ubiquitous across taxa, owing to sex-specific selection that arises from different reproductive strategies of the sexes. The pace-of-life syndrome (POLS) hypothesis predicts that most variation in such traits among individuals, populations, and species falls along a slow-fast pace-of-life continuum. As a result of their different reproductive roles and environment, the sexes also commonly differ in pace-of-life, with important consequences for the evolution of POLS. Here, we outline mechanisms for how males and females can evolve differences in POLS traits and in how such traits can covary differently despite constraints resulting from a shared genome. We review the current knowledge of the genetic basis of POLS traits and suggest candidate genes and pathways for future studies. Pleiotropic effects may govern many of the genetic correlations, but little is still known about the mechanisms involved in trade-offs between current and future reproduction and their integration with behavioral variation. We highlight the importance of metabolic and hormonal pathways in mediating sex differences in POLS traits; however, there is still a shortage of studies that test for sex specificity in molecular effects and their evolutionary causes. Considering whether and how sexual dimorphism evolves in POLS traits provides a more holistic framework to understand how behavioral variation is integrated with life histories and physiology, and we call for studies that focus on examining the sex-specific genetic architecture of this integration
Rotavirusinfecties en vaccinatie bij kinderen
Rotavirus is de belangrijkste oorzaak van acute gastro-​enteritis bij kinderen; de ziektelast is hoog, vooral vanwege dehydratie, en het betreft vaak zuigelingen. In Nederland is het rotavirus verantwoordelijk voor 1900 tot 3400 ziekenhuisopnames per jaar en vijf tot zes doden. Kinderen met een medische risicofactor (prematuriteit, laag geboortegewicht of congenitale aandoeningen) hebben een verhoogd risico op ernstige complicaties als gevolg van rotavirusgastro-​enteritis. Sinds 2006 zijn er werkzame, levend verzwakte orale vaccins beschikbaar voor zuigelingen, RotaTeq en Rotarix, die het risico op ernstige rotavirusgastro-​enteritis tot meer dan 80 procent verminderen. De vaccins worden over het algemeen goed verdragen. Er is echter een associatie met een klein verhoogd risico op invaginatie, vooral in de eerste zeven dagen na de eerste dosis. De WHO adviseert desondanks universele vaccinatie tegen het rotavirus voor zuigelingen, vanwege de gunstige risico-​batenverhouding van vaccinatie. In Nederland is er tot op heden geen rotavirusvaccinatieprogramma. De Gezondheidsraad heeft in september 2017 wel een advies uitgebracht waarin zij positief zijn ten opzichte van universele vaccinatie en het rotavirusvaccin in ieder geval aanraden voor kinderen met een medische risicofactor. Op dit moment wordt deze risicogroepstrategie uitgetest in het Risk group Infant Vaccination Against Rotavirus (RIVAR)-​project. Zuigelingen met een dergelijke risicofactor in dertien deelnemende Nederlandse ziekenhuizen krijgen het rotavirusvaccin aangeboden

Impact analysis of rotavirus vaccination in various geographic regions in Western Europe.
BACKGROUND: Universal mass vaccination (UMV) against rotavirus has been implemented in many but not all European countries. This study investigated the impact of UMV on rotavirus incidence trends by comparing European countries with UMV: Belgium, England/Wales and Germany versus countries without UMV: Denmark and the Netherlands. METHODS: For this observational retrospective cohort study, time series data (2001-2016) on rotavirus detections, meteorological factors and population demographics were collected. For each country, several meteorological and population factors were investigated as possible predictors of rotavirus incidence. The final set of predictors were incorporated in negative binomial models accounting for seasonality and serial autocorrelation, and time-varying incidence rate ratios (IRR) were calculated for each age group and country separately. The overall vaccination impact two years after vaccine implementation was estimated by pooling the results using a random effects meta-analyses. Independent t-tests were used to compare annual epidemics in the pre-vaccination and post-vaccination era to explore any changes in the timing of rotavirus epidemics. RESULTS: The population size and several meteorological factors were predictors for the rotavirus epidemiology. Overall, we estimated a 42% (95%-CI 23;56%) reduction in rotavirus incidence attributable to UMV. Strongest reductions were observed for age-groups 0-, 1- and 2-years (IRR 0.47, 0.48 and 0.63, respectively). No herd effect induced by UMV in neighbouring countries was observed. In all UMV countries, the start and/or stop and corresponding peak of the rotavirus season was delayed by 4-7Â weeks. CONCLUSIONS: The introduction of rotavirus UMV resulted in an overall reduction of 42% in rotavirus incidence in Western European countries two years after vaccine introduction and caused a change in seasonal pattern. No herd effect induced by UMV neighbouring countries was observed for Denmark and the Netherlands
Plant aquaporins: multifunctional water and solute channels with expanding roles
There is strong evidence that aquaporins are central components in plant water relations. Plant species possess more aquaporin genes than species from other kingdoms. According to sequence similarities, four major groups have been identified, which can be further divided into subgroups that may correspond to localization and transport selectivity. They may be involved in compatible solute distribution, gas-transfer (CO₂, NH₃) as well as in micronutrient uptake (boric acid). Recent advances in determining the structure of some aquaporins gives further details on the mechanism of selectivity. Gating behaviour of aquaporins is poorly understood but evidence is mounting that phosphorylation, pH, pCa and osmotic gradients can affect water channel activity. Aquaporins are enriched in zones of fast cell division and expansion, or in areas where water flow or solute flux density would be expected to be high. This includes biotrophic interfaces between plants and parasites, between plants and symbiotic bacteria or fungi, and between germinating pollen and stigma. On a cellular level aquaporin clusters have been identified in some membranes. There is also a possibility that aquaporins in the endoplasmic reticulum may function in symplasmic transport if water can flow from cell to cell via the desmotubules in plasmodesmata. Functional characterization of aquaporins in the native membrane has raised doubt about the conclusiveness of expression patterns alone and need to be conducted in parallel. The challenge will be to elucidate gating on a molecular level and cellular level and to tie those findings into plant water relations on a macroscopic scale where various flow pathways need to be considered
