    SGDB: a database of synthetic genes re-designed for optimizing protein over-expression

    Here we present the Synthetic Gene Database (SGDB): a relational database that houses sequences and associated experimental information on synthetic (artificially engineered) genes from all peer-reviewed studies published to date. At present, the database comprises information from more than 200 published experiments. This resource not only provides reference material to guide experimentalists in designing new genes that improve protein expression, but also offers a dataset for analysis by bioinformaticians who seek to test ideas regarding the underlying factors that influence gene expression. The SGDB was built under MySQL database management system. We also offer an XML schema for standardized data description of synthetic genes. Users can access the database at , or batch downloads all information through XML files. Moreover, users may visually compare the coding sequences of a synthetic gene and its natural counterpart with an integrated web tool at , and discuss questions, findings and related information on an associated e-forum at

    Rottlerin Inhibits Lonicera japonica-Induced Photokilling in Human Lung Cancer Cells through Cytoskeleton-Related Signaling Cascade

    This study demonstrated that many apoptotic signaling pathways, such as Rho family, PKC family, MAP kinase family, and mitochondria-mediated apoptotic pathway, were triggered by Lonicera japonica extracts and irradiation in CH27 cells. Rottlerin, a PKCδ -selective inhibitor, reversed the photoactivated Lonicera japonica extract-induced decrease in PKCδ protein expression and change in cell morphology in this study. In addition, rottlerin inhibited the photoactivated Lonicera japonica-induced decrease in protein expression of Ras, ERK, p38, PKCα, and PKCε, which are the kinases of prosurvival signaling pathway. We also demonstrated that pretreatment with rottlerin prevented actin microfilaments and microtubules from damage during the photoactivated Lonicera japonica-induced CH27 cell death. Furthermore, the promotion of the cytoskeleton-related signaling cascade following rottlerin by upregulation of cytoskeleton-related mediators (p38, HSP27, FAK, paxillin, and tubulin) and molecules of downstream of F-actin (mitochondria-mediated apoptosis pathway) reduces CH27 cell death, indicating that cytoskeleton is the potential target in the photoactivated Lonicera japonicaextract-induced photokilling of CH27 cells

    Investigation of Ni/SiO2 catalysts prepared at different conditions for hydrogen production from ethanol steam reforming

    Ni/SiO2 catalysts prepared by a sol–gel method have been investigated for hydrogen production via steam reforming of ethanol using a continuous flow, fixed bed reactor system. Chemical equilibrium calculations were also performed to determine the effects of temperature and molar steam to carbon ratio on hydrogen production. The acidity of the preparation solution (modified by nitric acid and ammonia) and calcination atmosphere (air and N2) were investigated in the preparation of the catalysts. BET surface area and porosity, temperature-programmed oxidation (TPO), X-ray diffraction (XRD), transmission electron microscopy (TEM) and scanning electron microscopy (SEM) were used to characterise the prepared catalysts. The BET surface area was reduced when the solution acidity was lowered during the sol–gel preparation process. A pH value less than 2.0 was necessary to achieve high metal dispersion in the catalyst. Smaller NiO particles were obtained when the catalyst was calcined in N2. Material balances on ethanol steam reforming at 600 °C using the prepared Ni/SiO2 catalysts were determined, and higher hydrogen production with lower coke deposition on the reacted catalysts were also obtained from the catalysts calcined in N2 atmosphere

The proposed control strategies are based on Lyapunov stability theory and include back-stepping control (BSC) and super-twisting sliding mode control. The overall WECS model and control scheme are developed in MATLAB/Simulink and the simulation results have shown that the BSC leads to superior performance and improved transient response as compared to the STSMC controller.Peer reviewe

    Enhancement of Mechanical and Thermal Properties of Polycaprolactone/Chitosan Blend by Calcium Carbonate Nanoparticles

    This study investigates the effects of calcium carbonate (CaCO3) nanoparticles on the mechanical and thermal properties and surface morphology of polycaprolactone (PCL)/chitosan nanocomposites. The nanocomposites of PCL/chitosan/CaCO3 were prepared using a melt blending technique. Transmission electron microscopy (TEM) results indicate the average size of nanoparticles to be approximately 62 nm. Tensile measurement results show an increase in the tensile modulus with CaCO3 nanoparticle loading. Tensile strength and elongation at break show gradual improvement with the addition of up to 1 wt% of nano-sized CaCO3. Decreasing performance of these properties is observed for loading of more than 1 wt% of nano-sized CaCO3. The thermal stability was best enhanced at 1 wt% of CaCO3 nanoparticle loading. The fractured surface morphology of the PCL/chitosan blend becomes more stretched and homogeneous in PCL/chitosan/CaCO3 nanocomposite.     Analysis of the spatio-temporal patterns of dry and wet conditions in the Huai River Basin using the standardized precipitation index

    Located in a transition zone between the northern and southern climates in China, the Huai River Basin is prone to extreme events such as drought and flood. Based on the daily precipitation data at 134 stations between 1961 and 2013, this paper analyzed the spatial and temporal patterns of the dry and wet conditions in the Huai River Basin through the statistical analysis of the rainfall stations' annual and seasonal standard precipitation index (SPI) series. Annual SPI series exhibited a decreasing trend at 86 stations and an increasing trend at the remaining stations. None of the increasing trend was significant, while the decreasing trend was significant at two stations at 5% significance level (α = 0.05) and one station at 10% level. Seasonal-wise, there has been a prevailing trend of drying in spring and autumn, and wetting in summer and winter. The trends in the spring and summer SPI series have been mostly insignificant, while those in autumn and winter, significant (α = 0.10) at over 30 stations. The Pettitt test results indicated that the significant transitions (α = 0.10) in the autumn and winter SPI series mostly occurred in the middle to late 1980s. Comparison of the average number of dry and wet years between the two sub-periods of 1961–1984 and 1990–2013 suggested a significant increase (α = 0.05) in the average number of severely wet years across much of the basin. Overall, significant changes have already occurred in the dry and wet conditions of the Huai River Basin, which could have profound impacts on the food and water safety situation of the region

    Tiny Medicine: Nanomaterial-Based Biosensors

    Tiny medicine refers to the development of small easy to use devices that can help in the early diagnosis and treatment of disease. Early diagnosis is the key to successfully treating many diseases. Nanomaterial-based biosensors utilize the unique properties of biological and physical nanomaterials to recognize a target molecule and effect transduction of an electronic signal. In general, the advantages of nanomaterial-based biosensors are fast response, small size, high sensitivity, and portability compared to existing large electrodes and sensors. Systems integration is the core technology that enables tiny medicine. Integration of nanomaterials, microfluidics, automatic samplers, and transduction devices on a single chip provides many advantages for point of care devices such as biosensors. Biosensors are also being used as new analytical tools to study medicine. Thus this paper reviews how nanomaterials can be used to build biosensors and how these biosensors can help now and in the future to detect disease and monitor therapies

    Acupuncture for the treatment of severe acute pain in Herpes Zoster: results of a nested, open-label, randomized trial in the VZV Pain Study

    <p>Abstract</p> <p>Background</p> <p>Data on the potential efficacy of acupuncture (AC) in controlling intense or very intense pain in patients with Herpes Zoster (HZ) has not been so far adequately assessed in comparison with standard pharmacological treatment (ST) by a controlled trial design.</p> <p>Methods</p> <p>Within the VZV Pescara study, pain was assessed in HZ patients on a Visual Analogue Scale (VAS) and by the McGill Pain Questionnaire (MPQ) both at the beginning and at the end of treatment. Response rates, mean changes in pain intensity, differences in total pain burden with an area-under-the-curve (AUC) method over a 1-year follow-up and differences in the incidence of Post-Herpetic Neuralgia (PHN) were evaluated.</p> <p>Results</p> <p>One hundred and two patients were randomized to receive either AC (n = 52) or ST (n = 50) for 4 weeks. Groups were comparable regarding age, sex, pain intensity at presentation and missed antiviral prescription. Both interventions were largely effective. No significant differences were observed in response rates (81.6% vs 89.2%, p = 0.8), mean reduction of VAS (4.1 +/- 2.3 vs 4.9 +/- 1.9, p = 0.12) and MPQ scores (1.3 +/- 0.9 vs 1.3 +/- 0.9, p = 0.9), incidence of PHN after 3 months (48.4% vs 46.8%, p = 0.5), and mean AUC during follow-up (199 +/- 136 vs 173 +/- 141, p = 0.4). No serious treatment-related adverse event was observed in both groups.</p> <p>Conclusions</p> <p>This controlled and randomized trial provides the first evidence of a potential role of AC for the treatment of acute herpetic pain.</p> <p>Trial registration</p> <p>ChiCTR-TRC-10001146.</p

    Global patient outcomes after elective surgery: prospective cohort study in 27 low-, middle- and high-income countries.

    BACKGROUND: As global initiatives increase patient access to surgical treatments, there remains a need to understand the adverse effects of surgery and define appropriate levels of perioperative care. METHODS: We designed a prospective international 7-day cohort study of outcomes following elective adult inpatient surgery in 27 countries. The primary outcome was in-hospital complications. Secondary outcomes were death following a complication (failure to rescue) and death in hospital. Process measures were admission to critical care immediately after surgery or to treat a complication and duration of hospital stay. A single definition of critical care was used for all countries. RESULTS: A total of 474 hospitals in 19 high-, 7 middle- and 1 low-income country were included in the primary analysis. Data included 44 814 patients with a median hospital stay of 4 (range 2-7) days. A total of 7508 patients (16.8%) developed one or more postoperative complication and 207 died (0.5%). The overall mortality among patients who developed complications was 2.8%. Mortality following complications ranged from 2.4% for pulmonary embolism to 43.9% for cardiac arrest. A total of 4360 (9.7%) patients were admitted to a critical care unit as routine immediately after surgery, of whom 2198 (50.4%) developed a complication, with 105 (2.4%) deaths. A total of 1233 patients (16.4%) were admitted to a critical care unit to treat complications, with 119 (9.7%) deaths. Despite lower baseline risk, outcomes were similar in low- and middle-income compared with high-income countries. CONCLUSIONS: Poor patient outcomes are common after inpatient surgery. Global initiatives to increase access to surgical treatments should also address the need for safe perioperative care. STUDY REGISTRATION: ISRCTN5181700
