    A comparison of Rosseland-mean opacities from OP and OPAL

    Monochromatic opacities from the Opacity Project (OP) (Seaton et al.) have been augmented by hitherto missing inner-shell contributions (Badnell & Seaton). OP Rosseland-mean opacities are compared with results from OPAL (Iglesias & Rogers) for the elements H, He, C, O, S and Fe. Overall, there is good agreement between OP and OPAL Rosseland-mean opacities for the 6-elements, but there are some differences. In particular, recent work (Bahcall et al.) has shown that helioseismology measurements give a very accurate value for the depth of the solar convection zone and that solar models give agreement with that value only if opacities there are about 7% larger than OPAL values.     Conditional gene deletion with DiCre demonstrates an essential role for CRK3 in Leishmania mexicana cell cycle regulation

    Leishmania mexicana has a large family of cyclin-dependent kinases (CDKs) that reflect the complex interplay between cell cycle and life cycle progression. Evidence from previous studies indicated that Cdc2 related kinase 3 (CRK3) in complex with the cyclin CYC6 is a functional homologue of the major cell cycle regulator CDK1, yet definitive genetic evidence for an essential role in parasite proliferation is lacking. To address this, we have implemented an inducible gene deletion system based on a dimerised Cre recombinase (diCre) to target CRK3 and elucidate its role in the cell cycle of L. mexicana. Induction of diCre activity in promastigotes with rapamycin resulted in efficient deletion of floxed CRK3, resulting in G2/M growth arrest. Co-expression of a CRK3 transgene during rapamycin-induced deletion of CRK3 resulted in complementation of growth, whereas expression of an active site CRK3T178E mutant did not, showing that protein kinase activity is crucial for CRK3 function. Inducible deletion of CRK3 in stationary phase promastigotes resulted in attenuated growth in mice, thereby confirming CRK3 as a useful therapeutic target and diCre as a valuable new tool for analysing essential genes in Leishmania

    Evolutionary genomics of epidemic visceral leishmaniasis in the Indian subcontinent

    Leishmania donovani causes visceral leishmaniasis (VL), the second most deadly vector-borne parasitic disease. A recent epidemic in the Indian subcontinent (ISC) caused up to 80% ofglobal VL and over 30,000 deaths per year. Resistance against antimonial drugs has probably been a contributing factor in the persistence of this epidemic. Here we use whole genome sequences from 204 clinical isolates to track the evolution and epidemiology of L. donovani from the ISC. We identify independent radiations that have emerged since a bottleneck coincident with 1960s DDT spraying campaigns. A genetically distinct population frequently resistant to antimonials has a two base-pair insertion in the aquaglyceroporin gene LdAQP1 that prevents the transport of trivalent antimonials. We find evidence of genetic exchange between ISC populations, and show that the mutation in LdAQP1 has spread by recombination. Our results reveal the complexity of L. donovani evolution in the ISC in response to drug treatment

    Eclipsing binaries in open clusters. II. V453 Cyg in NGC 6871

    We derive absolute dimensions of the early B-type detached eclipsing binary V453 Cygni (B0.4 IV + B0.7 IV, P=3.89d), a member of the open cluster NGC 6871. From the analysis of new, high-resolution, spectroscopy and the UBV light curves of Cohen (1974) we find the masses to be 14.36 +/- 0.20 and 11.11 +/- 0.13 Msun, the radii to be 8.55 +/- 0.06 and 5.49 +/- 0.06 Rsun, and the effective temperatures to be 26600 +/- 500 and 25 500 +/- 800 K for the primary and secondary stars, respectively. The surface gravities of 3.731 +/- 0.012 and 4.005 +/- 0.015 indicate that V453 Cyg is reaching the end of its main sequence lifetime. We have determined the apsidal motion period of the system to be 66.4 +/- 1.8 yr using the technique of Lacy (1992) extended to include spectroscopic data as well as times of minimum light, giving a density concentration coefficient of log(k_2) = -2.226 +/- 0.024. Contaminating (third) light has been detected for the first time in the light curve of V453 Cyg; previous analyses without this effect systematically underestimate the ratio of the radii of the two stars. The absolute dimensions of the system have been compared to the stellar evolution models of the Granada, Geneva, Padova and Cambridge groups. All model sets fit the data on V453 Cyg for solar helium and metal abundances and an age of 10.0 +/- 0.2 Myr. The Granada models also agree fully with the observed log(k_2) once general relativistic effects have been accounted for. The Cambridge models with convective core overshooting fit V453 Cyg better than those without. Given this success of the theoretical predictions, we briefly discuss which eclipsing binaries should be studied in order to further challenge the models.Comment: Accepted for publication in MNRAS (14 pages, 5 figures, Fig.4 reduced in size

    Evolution of fluid expulsion and concentrated hydrate zones across the southern Hikurangi subduction margin, New Zealand: An analysis from depth migrated seismic data

    Identification of methane sources controlling hydrate distribution and concentrations in continental margins remains a major challenge in gas hydrate research. Lack of deep fluid samples and high quality regional scale seismic reflection data may lead to underestimation of the significance of fluid escape from subducting and compacting sediments in the global inventory of methane reaching the hydrate zone, the water column and the atmosphere. The distribution of concentrated hydrate zones in relation to focused fluid flow across the southern Hikurangi subduction margin was investigated using high quality, long offset (10 km streamer), pre-stack depth migrated multichannel seismic data. Analysis of low P wave velocity zones, bright-reverse polarity reflections and dim-amplitude anomalies reveals pathways for gas escape and zones of gas accumulation. The study shows the structural and stratigraphic settings of three main areas of concentrated hydrates: (1) the Opouawe Bank, dominated by focused periodic fluid input along thrust faults sustaining dynamic hydrate concentrations and gas chimneys development; (2) the frontal anticline, with a basal set of protothrusts controlling permeability for fluids from deeply buried and subducted sediments sustaining hydrate concentrations at the crest of the anticline; and (3) the Hikurangi Channel, with buried sand dominated channels hosting significant amounts of gas beneath the base of the hydrate zone. In sand dominated channels gas injection into the hydrate zone favors highly concentrated hydrate accumulations.     Cell polarity genes including Crumbs (Crb) and Par complexes are essential for controlling photoreceptor morphogenesis. Among the Crb and Par complexes, Bazooka (Baz, Par-3 homolog) acts as a nodal component for other cell polarity proteins. Therefore, finding other genes interacting with Baz will help us to understand the cell polarity genes' role in photoreceptor morphogenesis. mutation on developing eyes to determine its role in photoreceptor morphogenesis. We found that Cnn is dispensable for retinal differentiation in eye imaginal discs during the larval stage. However, photoreceptors deficient in Cnn display dramatic morphogenesis defects including the mislocalization of Crumbs (Crb) and Bazooka (Baz) during mid-stage pupal eye development, suggesting that Cnn is specifically required for photoreceptor morphogenesis during pupal eye development. This role of Cnn in apical domain modulation was further supported by Cnn's gain-of-function phenotype.     C9ORF72 interaction with cofilin modulates actin dynamics in motor neurons.

    Intronic hexanucleotide expansions in C9ORF72 are common in amyotrophic lateral sclerosis (ALS) and frontotemporal dementia, but it is unknown whether loss of function, toxicity by the expanded RNA or dipeptides from non-ATG-initiated translation are responsible for the pathophysiology. We determined the interactome of C9ORF72 in motor neurons and found that C9ORF72 was present in a complex with cofilin and other actin binding proteins. Phosphorylation of cofilin was enhanced in C9ORF72-depleted motor neurons, in patient-derived lymphoblastoid cells, induced pluripotent stem cell-derived motor neurons and post-mortem brain samples from ALS patients. C9ORF72 modulates the activity of the small GTPases Arf6 and Rac1, resulting in enhanced activity of LIM-kinases 1 and 2 (LIMK1/2). This results in reduced axonal actin dynamics in C9ORF72-depleted motor neurons. Dominant negative Arf6 rescues this defect, suggesting that C9ORF72 acts as a modulator of small GTPases in a pathway that regulates axonal actin dynamics

    Diversity of Antibiotic-Active Bacteria Associated with the Brown Alga Laminaria saccharina from the Baltic Sea

    Bacteria associated with the marine macroalga Laminaria saccharina, collected from the Kiel Fjord (Baltic Sea, Germany), were isolated and tested for antimicrobial activity. From a total of 210 isolates, 103 strains inhibited the growth of at least one microorganism from the test panel including Gram-negative and Gram-positive bacteria as well as a yeast. Most common profiles were the inhibition of Bacillus subtilis only (30%), B. subtilis and Staphylococcus lentus (25%), and B. subtilis, S. lentus, and Candida albicans (11%). In summary, the antibiotic-active isolates covered 15 different activity patterns suggesting various modes of action. On the basis of 16S rRNA gene sequence similarities >99%, 45 phylotypes were defined, which were classified into 21 genera belonging to Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Bacteroidetes, Firmicutes, and Actinobacteria. Phylogenetic analysis of 16S rRNA gene sequences revealed that four isolates possibly represent novel species or even genera. In conclusion, L. saccharina represents a promising source for the isolation of new bacterial taxa and antimicrobially active bacteria
