Characterisation of nicotine receptors on human peripheral blood mononuclear cells (PBMC)

Method: PBMC were isolated from human blood buffy coats provided by the blood transfusion service and were used for radioligand binding studies with [3H]-nicotine. RT-PCR experiments were used to determine nAChR subunit expression while immunoblotting experiments were used to confirm that nAChR subunits identified by RT-PCR were translated into protein. Results: Binding studies suggested the presence of one binding site for (-)- nicotine on human peripheral blood lymphocytes. Competition studies showed that only (-)- nicotine, epibatidine and α-bungarotoxin, displaced radiolabelled nicotine from cells. RT-PCR studies demonstrated mRNA for α4, α5, α7, ÎČ1 and ÎČ2 nAChRs subunits in PBMC. Expression of mRNA for the a5 subunit of nAChR was observed in all lymphocyte samples tested. In contrast, the expression pattern of mRNAs for α4, α7, ÎČ1, and ÎČ2 mRNAs subunits of nAChRs, varied between samples. Western blot analysis showed that protein for α4, α5, and α7 and ÎČ2 nAChR subunits was expressed in most, but not all of the PBMC samples tested but some of the bands obtained were faint. Conclusion: The results obtained suggest that human PBMC contain nAChRs containing α4ÎČ2, α4ÎČ2α5, and/or α7 subunits.Peer reviewe

Comparing Formulations of Generalized Quantum Mechanics for Reparametrization-Invariant Systems
A class of decoherence schemes is described for implementing the principles
of generalized quantum theory in reparametrization-invariant `hyperbolic'
models such as minisuperspace quantum cosmology. The connection with
sum-over-histories constructions is exhibited and the physical equivalence or
inequivalence of different such schemes is analyzed. The discussion focuses on
comparing constructions based on the Klein-Gordon product with those based on
the induced (a.k.a. Rieffel, Refined Algebraic, Group Averaging, or Spectral
Analysis) inner product. It is shown that the Klein-Gordon and induced products
can be simply related for the models of interest. This fact is then used to
establish isomorphisms between certain decoherence schemes based on these

Semiclassical Trace Formulas for Noninteracting Identical Particles
We extend the Gutzwiller trace formula to systems of noninteracting identical
particles. The standard relation for isolated orbits does not apply since the
energy of each particle is separately conserved causing the periodic orbits to
occur in continuous families. The identical nature of the particles also
introduces discrete permutational symmetries. We exploit the formalism of
Creagh and Littlejohn [Phys. Rev. A 44, 836 (1991)], who have studied
semiclassical dynamics in the presence of continuous symmetries, to derive
many-body trace formulas for the full and symmetry-reduced densities of states.

Multiresolution analysis of active region magnetic structure and its correlation with the Mt. Wilson classification and flaring activity
Two different multi-resolution analyses are used to decompose the structure
of active region magnetic flux into concentrations of different size scales.
Lines separating these opposite polarity regions of flux at each size scale are
found. These lines are used as a mask on a map of the magnetic field gradient
to sample the local gradient between opposite polarity regions of given scale
sizes. It is shown that the maximum, average and standard deviation of the
magnetic flux gradient for alpha, beta, beta-gamma and beta-gamma-delta active
regions increase in the order listed, and that the order is maintained over all
length-scales. This study demonstrates that, on average, the Mt. Wilson
classification encodes the notion of activity over all length-scales in the
active region, and not just those length-scales at which the strongest flux
gradients are found. Further, it is also shown that the average gradients in
the field, and the average length-scale at which they occur, also increase in
the same order. Finally, there are significant differences in the gradient
distribution, between flaring and non-flaring active regions, which are
maintained over all length-scales. It is also shown that the average gradient
content of active regions that have large flares (GOES class 'M' and above) is
larger than that for active regions containing flares of all flare sizes; this

Temporal changes in the gut microbiota in farmed Atlantic cod (Gadus morhua) outweigh the response to diet supplementation with macroalgae
Background:
Aquaculture successfully meets global food demands for many fish species. However, aquaculture production of Atlantic cod (Gadus morhua) is just 2.5% of total market production. For cod farming to be a viable economic venture specific challenges on how to increase growth, health and farming productivity need to be addressed. Feed ingredients play a key role here. Macroalgae (seaweeds) have been suggested as a functional feed supplement with both health and economic benefits for terrestrial farmed animals and fish. The impact of such dietary supplements to cod gut integrity and microbiota, which contribute to overall fish robustness is unknown. The objective of this study was to supplement the diet of juvenile Atlantic cod with macroalgae and determine the impacts on fish condition and growth, gut morphology and hindgut microbiota composition (16S rRNA amplicon sequencing). Fish were fed one of three diets: control (no macroalgal inclusion), 10% inclusion of either egg wrack (Ascophyllum nodosum) or sea lettuce (Ulva rigida) macroalgae in a 12-week trial.
Results:
The results demonstrated there was no significant difference in fish condition, gut morphology or hindgut microbiota between the U. rigida supplemented fish group and the control group at any time-point. This trend was not observed with the A. nodosum treatment. Fish within this group were further categorised as either âNormalâ or âLower Growthâ. âLower Growthâ individuals found the diet unpalatable resulting in reduced weight and condition factor combined with an altered gut morphology and microbiome relative to the other treatments. Excluding this group, our results show that the hindgut microbiota was largely driven by temporal pressures with the microbial communities becoming more similar over time irrespective of dietary treatment. The core microbiome at the final time-point consisted of the orders Vibrionales (Vibrio and Photobacterium), Bacteroidales (Bacteroidetes and Macellibacteroides) and Clostridiales (Lachnoclostridium).
Conclusions:
Our study indicates that U. rigida macroalgae can be supplemented at 10% inclusion levels in the diet of juvenile farmed Atlantic cod without any impact on fish condition or hindgut microbial community structure. We also conclude that 10% dietary inclusion of A. nodosum is not a suitable feed supplement in a farmed cod diet
Opsin evolution in the Ambulacraria
Opsins - G-protein coupled receptors involved in photoreception - have been extensively studied in the animal kingdom. The present work provides new insights into opsin-based photoreception and photoreceptor cell evolution with a first analysis of opsin sequence data for a major deuterostome clade, the Ambulacraria. Systematic data analysis, including for the first time hemichordate opsin sequences and an expanded echinoderm dataset, led to a robust opsin phylogeny for this cornerstone superphylum. Multiple genomic and transcriptomic resources were surveyed to cover each class of Hemichordata and Echinodermata. In total, 119 ambulacrarian opsin sequences were found, 22 new sequences in hemichordates and 97 in echinoderms (including 67 new sequences). We framed the ambulacrarian opsin repertoire within eumetazoan diversity by including selected reference opsins from non-ambulacrarians. Our findings corroborate the presence of all major ancestral bilaterian opsin groups in Ambulacraria. Furthermore, we identified two opsin groups specific to echinoderms. In conclusion, a molecular phylogenetic framework for investigating light-perception and photobiological behaviors in marine deuterostomes has been obtained
Detector Description and Performance for the First Coincidence Observations between LIGO and GEO
For 17 days in August and September 2002, the LIGO and GEO interferometer
gravitational wave detectors were operated in coincidence to produce their
first data for scientific analysis. Although the detectors were still far from
their design sensitivity levels, the data can be used to place better upper
limits on the flux of gravitational waves incident on the earth than previous
direct measurements. This paper describes the instruments and the data in some

Search for Gravitational Waves from Primordial Black Hole Binary Coalescences in the Galactic Halo
We use data from the second science run of the LIGO gravitational-wave
detectors to search for the gravitational waves from primordial black hole
(PBH) binary coalescence with component masses in the range 0.2--.
The analysis requires a signal to be found in the data from both LIGO
observatories, according to a set of coincidence criteria. No inspiral signals
were found. Assuming a spherical halo with core radius 5 kpc extending to 50
kpc containing non-spinning black holes with masses in the range 0.2--, we place an observational upper limit on the rate of PBH coalescence
of 63 per year per Milky Way halo (MWH) with 90% confidence.Comment: 7 pages, 4 figures, to be submitted to Phys. Rev.
Update on the correlation of the highest energy cosmic rays with nearby extragalactic matter
Data collected by the Pierre Auger Observatory through 31 August 2007 showed
evidence for anisotropy in the arrival directions of cosmic rays above the
Greisen-Zatsepin-Kuz'min energy threshold, \nobreak{eV}. The
anisotropy was measured by the fraction of arrival directions that are less
than from the position of an active galactic nucleus within 75 Mpc
(using the V\'eron-Cetty and V\'eron catalog). An updated
measurement of this fraction is reported here using the arrival directions of
cosmic rays recorded above the same energy threshold through 31 December 2009.
The number of arrival directions has increased from 27 to 69, allowing a more
precise measurement. The correlating fraction is , compared
with expected for isotropic cosmic rays. This is down from the early
estimate of . The enlarged set of arrival directions is
examined also in relation to other populations of nearby extragalactic objects:
galaxies in the 2 Microns All Sky Survey and active galactic nuclei detected in
hard X-rays by the Swift Burst Alert Telescope. A celestial region around the
position of the radiogalaxy Cen A has the largest excess of arrival directions
relative to isotropic expectations. The 2-point autocorrelation function is
shown for the enlarged set of arrival directions and compared to the isotropic
