    Transnational innovation systems for bioeconomy: insights from cassava value chains in East Africa

    In this study, we analyze the purpose, challenges, and opportunities of transnational innovation systems using cassava as the case study crop in the East Africa region. Cassava scientifically referred to as Manihot esculenta Crantz, is an important food security crop for the poor and vulnerable and a potential building block for advancing the bioeconomy in Africa and the global South. Semi-structured interviews with researchers, government official, and small and medium enterprise representatives from the region were done to determine their level of collaborations with other partners across the region and the opportunities and challenges for transnational innovation systems along the cassava value chain. The selection of interviewees followed a purposive sampling technique according to their knowledge on transnational innovation in the cassava value chain. This was corroborated by a focused literature review on innovation systems concepts gathered from scholarly literature. The findings show that transnational collaborations and innovations in the East African region offer opportunities for expansion of biomass production, bioprocessing, and value addition to the rich bioresources available. Collaboration across borders and transnational innovation systems were found to play an important role for advancing and/or upscaling cassava breeding, growing, value-adding products and novel processing methods and contributing to a growing East African bioeconomy. Other than the regional policy challenges, organizational and cultural barriers were more prominent in venturing, participation, and involvement of parties and users in transnational innovations. These findings draw attention to the often unnoticed, but vital, role that institutional and policy frameworks play in initiating as well as supporting transnational innovation systems that address issues of current and future global concern. Therefore, appropriate policy environment and partnerships that offer opportunities for synergy and complementarity are vital for building effective transnational innovation systems that enhance the sustainability of production, value addition and end-uses of biobased cassava products

    Enhancement of wave damping within metamaterials having embedded negative stiffness inclusions

    The wave dissipation properties of layered periodic structures are modelled by FE as well as analytical approaches. A linear oscillator incorporating a negative stiffness element and having exceptional energy dissipation properties is exhibited and incorporated within the modelled structures. The structural dynamic stability of both the oscillator and the modelled waveguides is discussed and ensured. The numerical results provide evidence of a drastic increase of several orders of magnitude for the damping ratio of the flexural waves propagating within the structures

    Polyfunctional T cell responses in children in early stages of chronic Trypanosoma cruzi infection contrast with monofunctional responses of long-term infected adults

    Background: Adults with chronic Trypanosoma cruzi exhibit a poorly functional T cell compartment, characterized by monofunctional (IFN-γ-only secreting) parasite-specific T cells and increased levels of terminally differentiated T cells. It is possible that persistent infection and/or sustained exposure to parasites antigens may lead to a progressive loss of function of the immune T cells. Methodology/Principal Findings: To test this hypothesis, the quality and magnitude of T. cruzi-specific T cell responses were evaluated in T. cruzi-infected children and compared with long-term T. cruzi-infected adults with no evidence of heart failure. The phenotype of CD4+ T cells was also assessed in T. cruzi-infected children and uninfected controls. Simultaneous secretion of IFN-γ and IL-2 measured by ELISPOT assays in response to T. cruzi antigens was prevalent among T. cruzi-infected children. Flow cytometric analysis of co-expression profiles of CD4+ T cells with the ability to produce IFN-γ, TNF-α, or to express the co-stimulatory molecule CD154 in response to T. cruzi showed polyfunctional T cell responses in most T. cruzi-infected children. Monofunctional T cell responses and an absence of CD4+TNF-α+-secreting T cells were observed in T. cruzi-infected adults. A relatively high degree of activation and differentiation of CD4+ T cells was evident in T. cruzi-infected children. Conclusions/Significance: Our observations are compatible with our initial hypothesis that persistent T. cruzi infection promotes eventual exhaustion of immune system, which might contribute to disease progression in long-term infected subjects.Fil: Albareda, María Cecilia.     Stress-induced decreases in local cerebral glucose utilization in specific regions of the mouse brain

    BACKGROUND: Restraint stress in rodents has been reported to activate the hypothalamic-pituitary-adrenocortical (HPA) axis and to increase c-fos expression in regions that express components of the corticotropin-releasing factor (CRF) system. We have previously reported that acute central administration of CRF increased a measure of relative local cerebral glucose utilization (LCGU), a measure of neuronal activity in specific brain regions, and activated the HPA axis in mice. It was hypothesized that the involvement of the CRF system in the stress response would lead to similar changes in relative LCGU after restraint stress. In the present studies the effect of restraint stress on relative LCGU and on the HPA axis in C57BL/6N mice were examined. FINDINGS: Restraint stress activated the HPA axis in a restraint-duration dependent manner, but in contrast to the reported effects of CRF, significantly decreased relative LCGU in frontal cortical, thalamic, hippocampal and temporal dissected regions. These findings support evidence that stressors enforcing limited physical activity reduce relative LCGU, in contrast to high activity stressors such as swim stress. CONCLUSIONS: In conclusion, the present studies do not support the hypothesis that stress-induced changes in relative LCGU are largely mediated by the CRF system. Further studies will help to delineate the role of the CRF system in the early phases of the relative LCGU response to stress and investigate the role of other neurotransmitter systems in this response

    Functional and structural studies of the vaccinia virus virulence factor N1 reveal a Bcl-2-like anti-apoptotic protein

    Vaccinia virus (VACV) encodes many immunomodulatory proteins, including inhibitors of apoptosis and modulators of innate immune signalling. VACV protein N1 is an intracellular homodimer that contributes to virus virulence and was reported to inhibit nuclear factor (NF)-κB signalling. However, analysis of NF-κB signalling in cells infected with recombinant viruses with or without the N1L gene showed no difference in NF-κB-dependent gene expression. Given that N1 promotes virus virulence, other possible functions of N1 were investigated and this revealed that N1 is an inhibitor of apoptosis in cells transfected with the N1L gene and in the context of VACV infection. In support of this finding virally expressed N1 co-precipitated with endogenous pro-apoptotic Bcl-2 proteins Bid, Bad and Bax as well as with Bad and Bax expressed by transfection. In addition, the crystal structure of N1 was solved to 2.9 Å resolution (0.29 nm). Remarkably, although N1 shows no sequence similarity to cellular proteins, its three-dimensional structure closely resembles Bcl-xL and other members of the Bcl-2 protein family. The structure also reveals that N1 has a constitutively open surface groove similar to the grooves of other anti-apoptotic Bcl-2 proteins, which bind the BH3 motifs of pro-apoptotic Bcl-2 family members. Molecular modelling of BH3 peptides into the N1 surface groove, together with analysis of their physico-chemical properties, suggests a mechanism for the specificity of peptide recognition. This study illustrates the importance of the evolutionary conservation of structure, rather than sequence, in protein function and reveals a novel anti-apoptotic protein from orthopoxviruses
