    On the evaluation of the conversational speech quality in telecommunications

    International audienceIn this paper we propose an objective method to assess speech quality in the conversational context by taking into account the talking and listening speech qualities and the impact of delay. This approach is applied to the results of four subjective tests on the effects of echo, delay, packet loss and noise. The dataset is divided into training and validation sets. For the training set, a multiple linear regression is applied to determine a relationship between conversational, talking and listening speech qualities and the delay value. The multiple linear regression leads to an accurate estimation of the conversational scores with high correlation and low error between subjective and estimated scores, both on the training and validation sets. In addition, a validation is performed on the data of a subjective test found in the literature which confirms the reliability of the regression. The relationship is then applied to an objective level by replacing talking and listening subjective scores with talking and listening objective scores provided by existing objective models, fed by speech signals recorded during the subjective tests. The conversational model achieves high perfor- mance as revealed by comparison with the test results and with the existing standard methodology “E-model”, presented in the ITU-T (International Telecommunication Union) Recommendation G.107

    Native lysozyme and dry-heated lysozyme interactions with membrane lipid monolayers: lateral reorganization of LPS monolayer, model of the E. coli outer membrane

    International audienceLysozyme is mainly described active against Gram-positive bacteria, but is also efficient against some Gram-negative species. Especially, it was recently demonstrated that lysozyme disrupts Escherichia coli membranes. Moreover, dry-heating changes the physicochemical properties of the protein and increases the membrane activity of lysozyme. In order to elucidate the mode of insertion of lysozyme into the bacterial membrane, the interaction between lysozyme and a LPS monolayer mimicking the E. coli outer membrane has been investigated by tensiometry, ellipsometry, Brewster angle microscopy and atomic force microscopy. It was thus established that lysozyme has a high affinity for the LPS monolayer, and is able to insert into the latter as long as polysaccharide moieties are present, causing reorganization of the LPS monolayer. Dry-heating increases the lysozyme affinity for the LPS monolayer and its insertion capacity; the resulting reorganization of the LPS monolayer is different and more drastic than with the native protein

    Identification of the bacteria and their metabolic activities associated with the microbial spoilage of custard cream desserts

    The famous French dessert “ile flottante” consists of a sweet egg white foam floating on a vanilla custard cream,which contains highly nutritive raw materials, including milk, sugar and egg. Spoilage issues are therefore a keyconcern for the manufacturers. This study explored the bacterial diversity of 64 spoiled custard cream dessertsmanufactured by 2 French companies. B. cereus group bacteria, coagulase negative Staphylococcus, Enterococcus and Leuconostoc spp. were isolated from spoiled products. Thirty-one bacterial isolates representative of the main spoilage species were tested for their spoilage abilities. Significant growth and pH decrease were observed regardless of species. While off-odours were detected with B. cereus group and staphylococci, yoghurt odours were detected with Enterococcus spp. and Leuconostoc spp. B. cereus group bacteria produced various esters and several compounds derived from amino acid and sugar metabolism. Most Staphylococci produced phenolic compounds. Enterococcus spp. and Leuconostoc spp. isolates produced high levels of compounds derived from sugar metabolism. Each type of spoilage bacteria was associated with a specific volatile profile and lactic acid was identified as a potential marker of spoilage of custard cream-based desserts. These findings provide valuable information for manufacturers to improve food spoilage detection and prevention of chilled desserts made with milk and egg

    High quality phased assembly of grape genome offer new opportunities in chimera detection

    In perennial plants and especially those propagated through cuttings, several genotypes can coexist in a single individual, thus leading to chimeras. When the variant induces a noticeable phenotype modification, it can lead to a new cultivar. Viticulture already took economic advantage of this natural phenomenon: for instance, the berry skin of ‘Pinot gris’ derived from ‘Pinot noir’ by the selection of a chimera. Chimeras could also impact other crucial traits without being visually identified. Periclinal chimera where the variant has entirely colonized a cell layer is the most stable and can be propagated through cuttings. In grapevine, two functional cell layers are present in leaves, L1 and L2. However, lateral roots are formed from the L2 cell layer only. Thus, comparing DNA sequences of roots and leaves could allow chimera detection. In this study we used new generation Hifi long reads sequencing and recent bioinformatics tools applied to ‘Merlot’ to detect periclinal chimeras. Sequencing of ‘Magdeleine Noire des Charentes’ and ‘Cabernet franc’, the parents of ‘Merlot’, allowed haplotype resolved assembly. Pseudomolecules were built with few contigs, in some occasions only one per chromosome. This high resolution allowed haplotype comparison. Annotation from PN40024 was transferred to all pseudomolecules. Through variant detection, periclinal chimeras were found on both haplotypes. These results open new perspectives on chimera detection, which is an important resource to improve cultivars through clonal selection or breed new ones.     Production, secretion and purification of a correctly folded staphylococcal antigen in Lactococcus lactis.

    International audienceLactococcus lactis, a lactic acid bacterium traditionally used to ferment milk and manufacture cheeses, is also, in the biotechnology field, an interesting host to produce proteins of medical interest, as it is "Generally Recognized As Safe". Furthermore, as L. lactis naturally secretes only one major endogenous protein (Usp45), the secretion of heterologous proteins in this species facilitates their purification from a protein-poor culture medium. Here, we developed and optimized protein production and secretion in L. lactis to obtain proteins of high quality, both correctly folded and pure to a high extent. As proteins to be produced, we chose the two transmembrane members of the HtrA protease family in Staphylococcus aureus, an important extra-cellular pathogen, as these putative surface-exposed antigens could constitute good targets for vaccine development. A recombinant ORF encoding a C-terminal, soluble, proteolytically inactive and tagged form of each staphylococcal HtrA protein was cloned into a lactococcal expression-secretion vector. After growth and induction of recombinant gene expression, L. lactis was able to produce and secrete each recombinant rHtrA protein as a stable form that accumulated in the culture medium in similar amounts as the naturally secreted endogenous protein, Usp45. L. lactis growth in fermenters, in particular in a rich optimized medium, led to higher yields for each rHtrA protein. Protein purification from the lactococcal culture medium was easily achieved in one step and allowed recovery of highly pure and stable proteins whose identity was confirmed by mass spectrometry. Although rHtrA proteins were monomeric, they displayed the same secondary structure content, thermal stability and chaperone activity as many other HtrA family members, indicating that they were correctly folded. rHtrA protein immunogenicity was established in mice. The raised polyclonal antibodies allowed studying the expression and subcellular localization of wild type proteins in S. aureus: although both proteins were expressed, only HtrA1 was found to be, as predicted, exposed at the staphylococcal cell surface suggesting that it could be a better candidate for vaccine development. In this study, an efficient process was developed to produce and secrete putative staphylococcal surface antigens in L. lactis and to purify them to homogeneity in one step from the culture supernatant. This allowed recovering fully folded, stable and pure proteins which constitute promising vaccine candidates to be tested for protection against staphylococcal infection. L. lactis thus proved to be an efficient and competitive cell factory to produce proteins of high quality for medical applications

    Coeval extensional shearing and lateral underflow during Late Cretaceous core complex development in the Niğde Massif, Central Anatolia, Turkey

    27 p.International audienceThe Nig¢de Massif, at the southern tip of the Central Anatolian Crystalline Complex, consists of two structural units. Foliations in the lower unit define a dome cored by migmatites. The contact between the two units bears all the elements of a ductile to brittle extensional detachment. Hence the Nig¢de Massif represents an extensional metamorphic core complex. Top-to-NE/ENE shearing at higher levels of the lower unit relates to displacement along the detachment. Deeper levels of the lower unit display hightemperature top-to-SSW ductile shearing. The two shearing deformations show a difference in the mean trend of stretching lineations of up to 58. New 40Ar/39Ar ages combined with previously published data enable us to infer that the two shears were contemporaneous. In our favored interpretation, oblique shearing in the core of the dome reflects lateral underflow, i.e., horizontal flowing of the lower crust in a direction highly oblique to the direction of extension. As a result of the interaction between lateral underflow and downdip shearing along the overlying detachment, distinct structural domains are expected to exist within the migmatitic part of the core complex, with observed counterparts in the Nig¢de dome. Lateral underflow may reflect ''inward'' flow on the scale of the core complex. Regional-scale channel flow is an alternative that would better account for the record of non-coaxial deformation in the core of the dome. More generally, we suspect that the development of lateral underflow in a metamorphic core complex more likely reflects regional channel flow, rather than local inward flow
