    The effect of pressure on the electrical conductivity of sea-water

    Values of the pressure coefficient of electrical conductivity of sea-water (cblorinity 19.7‰) at different temperatures are presented. The pressure coefficients vary from 1.50 × 10-5 decibar-1 at 0.5°C to 0.82 × 10-5 decibar-1 at 19.5°C. The accuracy is estimated to be ±5%. The results have been confirmed by tests at sea

    Practical application of the Exceedance Probability Method of Middleton and Thompson (J. Geophys. Res., V ol.91, 1986, pp. 11707-11716) to the estimation of extreme sea levels is considered in detail, for ports where tidal amplitudes dominate surge. Data from Sydney (Australia) are used to illustrate the method. Direct use of the histogram of tidal residuals, instead of fitting a model to it, is discussed, and the limitations are pointed out. Graphical fitting of the sum of two normal distributions to the histogram is also discussed. A method is given for including a trend in mean sea level. A simplified method of checking if tidal amplitudes dominate the surge is given in an appendix

    Donor IFNL4 Genotype Is Associated with Early Post-Transplant Fibrosis in Recipients with Hepatitis C

    Background and Aims Early post-transplant hepatic fibrosis is associated with poor outcomes and may be influenced by donor/recipient genetic factors. The rs368234815 IFNL4 polymorphism is related to the previously described IL28B polymorphism, which predicts etiology-independent hepatic fibrosis. The aim of this study was to identify the impact of donor and/or recipient IFNL4 genotype on early fibrosis among patients transplanted for hepatitis C (HCV). Methods Clinical data were collected for 302 consecutive patients transplanted for HCV. 116 patients who had available liver biopsies and donor/recipient DNA were included. 28% of these patients with stage 2 fibrosis or greater were compared to patients without significant post-transplant fibrosis with respect to clinical features as well as donor/recipient IFNL4 genotype. Results The IFNL4 TT/TT genotype was found in 26.0% of recipients and 38.6% of donors. Patients who developed early post-transplant fibrosis had a 3.45 adjusted odds of having donor IFNL4 TT/TT genotype (p = 0.012). Donor IFNL4 TT/TT genotype also predicted decreased overall survival compared to non-TT/TT genotypes (p = 0.016). Conclusions Donor IFNL4 TT/TT genotype, a favorable predictor of spontaneous HCV clearance pre-transplant, is associated with increased early post-transplant fibrosis and decreased survival

    Gene flow estimation with microsatellites in a Malagasy seed orchard of Eucalyptus grandis

    International audienceEucalyptus grandis has a mixed-mating repro- ductive system. Malagasy Eucalyptus seed orchards were established 15 years ago with two aims both based on panmixia: open-pollinated seed production and genetic improvement. The panmixia hypothesis has never been confirmed in the seed orchard. From a seedling seed- orchard stand comprising 349 trees and using data obtained with six selected microsatellite markers, pater- nity analysis was performed for 724 offspring collected on 30 adult trees. Paternity assignment, based on exclu- sion procedures and likelihood-ratio method, was achieved with high accuracy; the exclusion probability value was 0.997. The outcrossing rate was very high (96.7%). More than 50% of potential male trees (199 out of 349) in the seed orchard contributed to pollination for 440 offspring of 30 progenies (8.6% of the basic population). The pollination rate from outside the seed orchard was high (39.2%), but might be due to the small size of this seed orchard. This study showed that "panmixia-like pollination" can be assume

    Phosphorylation by serine/threonine kinases has been described as a new mechanism for regulating the effects of insecticides on insect neuronal receptors and channels. Although insect GABA receptors are commercially important targets for insecticides (e.g. fipronil), their modulation by kinases is poorly understood and the influence of phosphorylation on insecticide sensitivity is unknown. Using the whole-cell patch-clamp technique, we investigated the modulatory effect of PKC and CaMKinase II on GABA receptor subtypes (GABAR1 and GABAR2) in DUM neurons isolated from the terminal abdominal ganglion (TAG) of Periplaneta americana. Chloride currents through GABAR2 were selectively abolished by PMA and PDBu (the PKC activators) and potentiated by Gö6983, an inhibitor of PKC. Furthermore, using KN-62, a specific CaMKinase II inhibitor, we demonstrated that CaMKinase II activation was also involved in the regulation of GABAR2 function. In addition, using CdCl(2) (the calcium channel blocker) and LOE-908, a blocker of TRPγ, we revealed that calcium influx through TRPγ played an important role in kinase activations. Comparative studies performed with CACA, a selective agonist of GABAR1 in DUM neurons confirmed the involvement of these kinases in the specific regulation of GABAR2. Furthermore, our study reported that GABAR1 was less sensitive than GABAR2 to fipronil. This was demonstrated by the biphasic concentration-response curve and the current-voltage relationship established with both GABA and CACA. Finally, we demonstrated that GABAR2 was 10-fold less sensitive to fipronil following inhibition of PKC, whereas inhibition of CaMKinase II did not alter the effect of fipronil

    Resistance to paclitaxel in a cisplatin-resistant ovarian cancer cell line is mediated by P-glycoprotein

    The IGROVCDDP cisplatin-resistant ovarian cancer cell line is also resistant to paclitaxel and models the resistance phenotype of relapsed ovarian cancer patients after first-line platinum/taxane chemotherapy. A TaqMan low-density array (TLDA) was used to characterise the expression of 380 genes associated with chemotherapy resistance in IGROVCDDP cells. Paclitaxel resistance in IGROVCDDP is mediated by gene and protein overexpression of P-glycoprotein and the protein is functionally active. Cisplatin resistance was not reversed by elacridar, confirming that cisplatin is not a P-glycoprotein substrate. Cisplatin resistance in IGROVCDDP is multifactorial and is mediated in part by the glutathione pathway and decreased accumulation of drug. Total cellular glutathione was not increased. However, the enzyme activity of GSR and GGT1 were up-regulated. The cellular localisation of copper transporter CTR1 changed from membrane associated in IGROV-1 to cytoplasmic in IGROVCDDP. This may mediate the previously reported accumulation defect. There was decreased expression of the sodium potassium pump (ATP1A), MRP1 and FBP which all have been previously associated with platinum accumulation defects in platinum-resistant cell lines. Cellular localisation of MRP1 was also altered in IGROVCDDP shifting basolaterally, compared to IGROV-1. BRCA1 was also up-regulated at the gene and protein level. The overexpression of P-glycoprotein in a resistant model developed with cisplatin is unusual. This demonstrates that P-glycoprotein can be up-regulated as a generalised stress response rather than as a specific response to a substrate. Mechanisms characterised in IGROVCDDP cells may be applicable to relapsed ovarian cancer patients treated with frontline platinum/taxane chemotherapy

    End-to-End Evaluation in Simultaneous Translation

    This paper presents the end-to-end evaluation of an automatic simultaneous translation system, built with state-of-the-art components. It shows whether, and for which situations, such a system might be advantageous when compared to a human interpreter. Using speeches in English translated into Spanish, we resent the evaluation procedure and we discuss the results both for the recognition and translation components as well as for the overall system. Even if the translation process remains the Achilles’ heel of the system, the results show that the system can keep at least half of the information, becoming potentially useful for final users

    Evidence of triggered star formation in G327.3-0.6. Dust-continuum mapping of an infrared dark cloud with P-ArT\'eMiS

    Aims. Expanding HII regions and propagating shocks are common in the environment of young high-mass star-forming complexes. They can compress a pre-existing molecular cloud and trigger the formation of dense cores. We investigate whether these phenomena can explain the formation of high-mass protostars within an infrared dark cloud located at the position of G327.3-0.6 in the Galactic plane, in between two large infrared bubbles and two HII regions. Methods: The region of G327.3-0.6 was imaged at 450 ? m with the CEA P-ArT\'eMiS bolometer array on the Atacama Pathfinder EXperiment telescope in Chile. APEX/LABOCA and APEX-2A, and Spitzer/IRAC and MIPS archives data were used in this study. Results: Ten massive cores were detected in the P-ArT\'eMiS image, embedded within the infrared dark cloud seen in absorption at both 8 and 24 ?m. Their luminosities and masses indicate that they form high-mass stars. The kinematical study of the region suggests that the infrared bubbles expand toward the infrared dark cloud. Conclusions: Under the influence of expanding bubbles, star formation occurs in the infrared dark areas at the border of HII regions and infrared bubbles.Comment: 4 page

    Assessment of musical noise using localization of isolated peaks in time-frequency domain

    Musical noise is a recurrent issue that appears in spectral techniques for denoising or blind source separation. Due to localised errors of estimation, isolated peaks may appear in the processed spectrograms, resulting in annoying tonal sounds after synthesis known as “musical noise”. In this paper, we propose a method to assess the amount of musical noise in an audio signal, by characterising the impact of these artificial isolated peaks on the processed sound. It turns out that because of the constraints between STFT coefficients, the isolated peaks are described as time-frequency “spots” in the spectrogram of the processed audio signal. The quantification of these “spots”, achieved through the adaptation of a method for localisation of significant STFT regions, allows for an evaluation of the amount of musical noise. We believe that this will pave the way to an objective measure and a better understanding of this phenomenon
