    Neotropical tadpoles: spatial and temporal distribution and habitat use in a seasonal lake in Veracruz, México

    We studied a tadpole assemblage in a seasonal neotropical lake where 14 species of anurans reproduce. Tadpoles were collected monthly at nine sampling stations at depth intervals of 1 m from the surface to the bottom (13 m). Sufficient numbers of tadpoles of three species were collected to compare habitat use. This three species assemblage breed in the following order (first to last): Smilisca baudinii, Gastrophryne usta, and Rana berlandieri. R. berlandieri had the greatest microhabitat breadth followed by S. baudinii. S. baudinii and G. usta had high microhabitat overlap, but significant differences in microhabitat use were found. S. baudinii tended to occur near the bottom, while G. usta was near the surface. This study shows that temporal and habitat partitioning both occur and depend on the species of tadpole. Dynamic interactions occur between habitat and temporal dimensions. Phenology and habitat selection depend both on the species and on abiotic and biotic factors

    Motor Decline in Clinically Presymptomatic Spinocerebellar Ataxia Type 2 Gene Carriers

    BACKGROUND: Motor deficits are a critical component of the clinical characteristics of patients with spinocerebellar ataxia type 2. However, there is no current information on the preclinical manifestation of those motor deficits in presymptomatic gene carriers. To further understand and characterize the onset of the clinical manifestation in this disease, we tested presymptomatic spinocerebellar ataxia type 2 gene carriers, and volunteers, in a task that evaluates their motor performance and their motor learning capabilities. METHODS AND FINDINGS: 28 presymptomatic spinocerebellar ataxia type 2 gene carriers and an equal number of control volunteers matched for age and gender participated in the study. Both groups were tested in a prism adaptation task known to be sensible to both motor performance and visuomotor learning deficits. Our results clearly show that although motor learning capabilities are intact, motor performance deficits are present even years before the clinical manifestation of the disease start. CONCLUSIONS: The results show a clear deficit in motor performance that can be detected years before the clinical onset of the disease. This motor performance deficit appears before any motor learning or clinical manifestations of the disease. These observations identify the performance coefficient as an objective and quantitative physiological biomarker that could be useful to assess the efficiency of different therapeutic agents

    Systemic lupus erythematosus (SLE), a complex and heterogeneous autoimmune disease, represents a significant challenge for both diagnosis and treatment. Patients with SLE in Latin America face special problems that should be considered when therapeutic guidelines are developed. The objective of the study is to develop clinical practice guidelines for Latin American patients with lupus. Two independent teams (rheumatologists with experience in lupus management and methodologists) had an initial meeting in Panama City, Panama, in April 2016. They selected a list of questions for the clinical problems most commonly seen in Latin American patients with SLE. These were addressed with the best available evidence and summarised in a standardised format following the Grading of Recommendations Assessment, Development and Evaluation approach. All preliminary findings were discussed in a second face-to-face meeting in Washington, DC, in November 2016. As a result, nine organ/system sections are presented with the main findings; an 'overarching' treatment approach was added. Special emphasis was made on regional implementation issues. Best pharmacologic options were examined for musculoskeletal, mucocutaneous, kidney, cardiac, pulmonary, neuropsychiatric, haematological manifestations and the antiphospholipid syndrome. The roles of main therapeutic options (ie, glucocorticoids, antimalarials, immunosuppressant agents, therapeutic plasma exchange, belimumab, rituximab, abatacept, low-dose aspirin and anticoagulants) were summarised in each section. In all cases, benefits and harms, certainty of the evidence, values and preferences, feasibility, acceptability and equity issues were considered to produce a recommendation with special focus on ethnic and socioeconomic aspects.     Analysis of protein-coding genetic variation in 60,706 humans

    Large-scale reference data sets of human genetic variation are critical for the medical and functional interpretation of DNA sequence changes. We describe the aggregation and analysis of high-quality exome (protein-coding region) sequence data for 60,706 individuals of diverse ethnicities generated as part of the Exome Aggregation Consortium (ExAC). This catalogue of human genetic diversity contains an average of one variant every eight bases of the exome, and provides direct evidence for the presence of widespread mutational recurrence. We have used this catalogue to calculate objective metrics of pathogenicity for sequence variants, and to identify genes subject to strong selection against various classes of mutation; identifying 3,230 genes with near-complete depletion of truncating variants with 72% having no currently established human disease phenotype. Finally, we demonstrate that these data can be used for the efficient filtering of candidate disease-causing variants, and for the discovery of human “knockout” variants in protein-coding genes

    Consensus guidelines for the use and interpretation of angiogenesis assays

    The formation of new blood vessels, or angiogenesis, is a complex process that plays important roles in growth and development, tissue and organ regeneration, as well as numerous pathological conditions. Angiogenesis undergoes multiple discrete steps that can be individually evaluated and quantified by a large number of bioassays. These independent assessments hold advantages but also have limitations. This article describes in vivo, ex vivo, and in vitro bioassays that are available for the evaluation of angiogenesis and highlights critical aspects that are relevant for their execution and proper interpretation. As such, this collaborative work is the first edition of consensus guidelines on angiogenesis bioassays to serve for current and future reference

    Albiglutide and cardiovascular outcomes in patients with type 2 diabetes and cardiovascular disease (Harmony Outcomes): a double-blind, randomised placebo-controlled trial

    Background: Glucagon-like peptide 1 receptor agonists differ in chemical structure, duration of action, and in their effects on clinical outcomes. The cardiovascular effects of once-weekly albiglutide in type 2 diabetes are unknown. We aimed to determine the safety and efficacy of albiglutide in preventing cardiovascular death, myocardial infarction, or stroke. Methods: We did a double-blind, randomised, placebo-controlled trial in 610 sites across 28 countries. We randomly assigned patients aged 40 years and older with type 2 diabetes and cardiovascular disease (at a 1:1 ratio) to groups that either received a subcutaneous injection of albiglutide (30–50 mg, based on glycaemic response and tolerability) or of a matched volume of placebo once a week, in addition to their standard care. Investigators used an interactive voice or web response system to obtain treatment assignment, and patients and all study investigators were masked to their treatment allocation. We hypothesised that albiglutide would be non-inferior to placebo for the primary outcome of the first occurrence of cardiovascular death, myocardial infarction, or stroke, which was assessed in the intention-to-treat population. If non-inferiority was confirmed by an upper limit of the 95% CI for a hazard ratio of less than 1·30, closed testing for superiority was prespecified. This study is registered with ClinicalTrials.gov, number NCT02465515. Findings: Patients were screened between July 1, 2015, and Nov 24, 2016. 10 793 patients were screened and 9463 participants were enrolled and randomly assigned to groups: 4731 patients were assigned to receive albiglutide and 4732 patients to receive placebo. On Nov 8, 2017, it was determined that 611 primary endpoints and a median follow-up of at least 1·5 years had accrued, and participants returned for a final visit and discontinuation from study treatment; the last patient visit was on March 12, 2018. These 9463 patients, the intention-to-treat population, were evaluated for a median duration of 1·6 years and were assessed for the primary outcome. The primary composite outcome occurred in 338 (7%) of 4731 patients at an incidence rate of 4·6 events per 100 person-years in the albiglutide group and in 428 (9%) of 4732 patients at an incidence rate of 5·9 events per 100 person-years in the placebo group (hazard ratio 0·78, 95% CI 0·68–0·90), which indicated that albiglutide was superior to placebo (p<0·0001 for non-inferiority; p=0·0006 for superiority). The incidence of acute pancreatitis (ten patients in the albiglutide group and seven patients in the placebo group), pancreatic cancer (six patients in the albiglutide group and five patients in the placebo group), medullary thyroid carcinoma (zero patients in both groups), and other serious adverse events did not differ between the two groups. There were three (<1%) deaths in the placebo group that were assessed by investigators, who were masked to study drug assignment, to be treatment-related and two (<1%) deaths in the albiglutide group. Interpretation: In patients with type 2 diabetes and cardiovascular disease, albiglutide was superior to placebo with respect to major adverse cardiovascular events. Evidence-based glucagon-like peptide 1 receptor agonists should therefore be considered as part of a comprehensive strategy to reduce the risk of cardiovascular events in patients with type 2 diabetes.     From April 1992 lo May 1993, zooplankton samples were collected monthly by means of horizontal tows in nine sites of the lake. Prior to the towing, temperature of surface water, transparency (Secchi), pH and dissolved oxygen were evaluated. A total of 31 zooplankton forrns, inc ing 14 species of rotifers, tbree copepods, five cladocerans and one ostracod, as well as protozoans (mainly vorticellíds and ciliates), were detected. Rotifers were the dominant organisms, mainly Brachionus havanaensis (27.6 ¡nd ¡-I), B. angularis (6.9 ind ¡-I), Keratella cochlearis (4.9 ¡nd ¡-I), Conochilus unicornis (10.8 ¡nd 1-1) and C. dossuarius ( 3.1 ¡nd ¡-I).Entre abril de 1992 y mayo de 1993, se realizaron mensualmente recolectas subsuperficiales de zooplancton, con red, en nueve localidades del lago. en donde también se determinaron la temperatura superficial, la visibilidad del disco de Secchi, el pH y la concentraci6n de oxígeno disuelto. Entre las 31 formas de plancton detectadas en las distintas submuestras se registraron 14 especies de rotíferos, tres de copépodos, cinco de cJad6ceros y un ostrácodo, además de protozoarios, principalmente vorticélidos y ciliados.     We analyze the origin of knowledge about fish species richness in the Tuxpan-Tampamachoco estuarine system, in Veracuz, México. A complete inventory of the fish species known to date for this system (N=179) was elaborated from published lists and from sampling seagrass meadows of Tampamachoco Lagoon, which yielded 14 previously unknown species. W hen compared, the different Iists showed a low similarity that may reflect differences in sampling methods and collecting strategies. Current data suggest that fish species richness in Mexican coastal lagoons (Gulf of Mexico) is not related with ¡agoon surface area, as has been suggested, but with the number of inventories available for each lagoon, being these a reflection of the sampling effort. A sampling design for the assessment of fish species richness in estuarine systems should consíder: a) using the highest possíble varíety of samplíng fishing gears, b) col!ecting in al! microhabitat types and c) the preference of bimonthly or quarterly samplings for two or more years over monthly samplings in a single yearSe analiza la manera como se ha conformado el conocimiento de la ictiofauna del sistema estuarino-Iagunar de Tuxpan-Tampamachoco, en el estado mexicano de Veracruz. Se elaboró un inventario de los peces conocidos hasta la fecha en dicho sistema (N= 179), a partir de listas previamente publicadas y de muestreos propios en las áreas someras de la laguna cubiertas por pastos marinos, que nos permitieron detectar 14 especies antes desconocidas en el área. Al comparar los distintos listados se observa una escasa similitud, la cual se atribuye a las diferencias en las estrategias de recolecta empleadas. Al menos con los datos actualmente disponibles, la riqueza íctica registrada en las lagunas costeras del Golfo de México no está relacionada con su área, como se ha sugerido, sino con el número de inventarios disponibles para cada laguna, pues éste involucra importantes diferencias en el esfuerzo de muestreo y en las estrategias de recolecta. Se recomienda un diseño de muestreo que contemple: a) emplear la mayor variedad posible de artes de pesca, b) recolectar en todos los tipos de microhábitat y, c) preferir los muestreos cada dos o tres meses por dos o más años sobre los muestreos mensuales durante un sólo año
