    Involving Immigrant Latino Farmers in Local Food Systems A Community Capitals Approach

    Presentation made at Latinos in the Heartland (10th : 2012 : Kansas City, Mo.) and published in the annual conference proceedings.Using the community capital framework, this study analyzes two projects that engage immigrants (Latinos) in gardening. The first project focuses on creation and implementation of a farm incubator program for both immigrant Latino farmers and beginning native (Anglo) farmers and the subsequent organization of a local foods group in a non-metropolitan Iowa community. The second project revitalizes a community garden by involving immigrants (Latinos). The paper uses multiple sources and methods to document the successes and shortcomings in building a multicultural food system. These include focus groups with Latino/a farmers and organizers of the program, participant observation, notes from steering committee meetings, analysis of participatory evaluations, and content analysis of documents from meetings and classes. Human, social, and cultural capitals are essential elements for these programs to succeed. The interaction among these three capitals mobilizes other community capitals for program improvement. However, in a multicultural situation, these capitals can inadvertently challenge continuity and success. The intergroup relations that emerge from the interaction among these capitals can be unpredictable. Recommendations center on how to reduce risk in mobilizing the most critical community capitals

    Exploring the meanings of food and agriculture for Latino youth through the use of PhotoVoice in three Iowa cities

    Presentation made at Latinos in the Heartland (11th : 2013 : Columbia, Mo.) and published in the annual conference proceedings.Using PhotoVoice as a tool for participatory action research, 10 Latino high school students, from three different cities of Iowa, Des Moines, Council Bluffs and Ottumwa, completed a project developed by Iowa State University Sociology Extension. The students explored the cultural meanings of food and agriculture within their families and in their communities. Participants were asked to reflect on and describe their pictures, incorporating their points of view on the different elements that integrated local food systems. We coded their final essays in NVIVO using the Community Capital Framework (CCF) to evaluate students' experiences and perceptions of food-value chains in their communities. The results showed cultural capital and bonding social capital to be the dominant themes, followed by health and nutrition (human capital) and natural capital (emphasis on home gardens and fresh vegetables). Financial capital, built and political capital indicators were mentioned incidentally to other capitals. Recommendations in this paper center on improving the use of PhotoVoice among Latino youth

    Druggable Nucleolin Identifies Breast Tumours Associated with Poor Prognosis That Exhibit Different Biological Processes

    Background: Nucleolin (NCL) is a multifunctional protein with oncogenic properties. Anti-NCL drugs show strong cytotoxic effects, including in triple-negative breast cancer (TNBC) models, and are currently being evaluated in phase II clinical trials. However, few studies have investigated the clinical value of NCL and whether NCL stratified cancer patients. Here, we have investigated for the first time the association of NCL with clinical characteristics in breast cancers independently of the different subtypes. Methods: Using two independent series (n = 216; n = 661), we evaluated the prognostic value of NCL in non-metastatic breast cancers using univariate and/or multivariate Cox-regression analyses. Results: We reported that NCL mRNA expression levels are markers of poor survivals independently of tumour size and lymph node invasion status (n = 216). In addition, an association of NCL expression levels with poor survival was observed in TNBC (n = 40, overall survival (OS) p = 0.0287, disease-free survival (DFS) p = 0.0194). Transcriptomic analyses issued from The Cancer Genome Atlas (TCGA) database (n = 661) revealed that breast tumours expressing either low or high NCL mRNA expression levels exhibit different gene expression profiles. These data suggest that tumours expressing high NCL mRNA levels are different from those expressing low NCL mRNA levels. Conclusions: NCL is an independent marker of prognosis in breast cancers. We anticipated that anti-NCL is a promising therapeutic strategy that could rapidly be evaluated in high NCL-expressing tumours to improve breast cancer management

    Hunter-gatherer environments at the Late Pleistocene sites of Mwanganda's Village and Bruce, northern Malawi

    Mwanganda's Village (MGD) and Bruce (BRU) are two open-air site complexes in northern Malawi with deposits dating to between 15 and 58 thousand years ago (ka) and containing Middle Stone Age (MSA) lithic assemblages. The sites have been known since 1966 and 1965, respectively, but lacked chronometric and site formation data necessary for their interpretation. The area hosts a rich stone artifact record, eroding from and found within alluvial fan deposits exhibiting poor preservation of organic materials. Although this generally limits opportunities for site-based environmental reconstructions, MGD and BRU are located at the distal margins of the alluvial fan, where lacustrine lagoonal deposits were overprinted by a calcrete paleosol. This has created locally improved organic preservation and allowed us to obtain ecological data from pollen, phytoliths, and pedogenic carbonates, producing a regional- to site-scale environmental context for periods of site use and abandonment. Here, we integrate the ecological data into a detailed site formation history, based on field observations and micromorphology, supplemented by cathodoluminescence microscopy and Ό-XRF. By comparing local, on-site environmental proxies with more regional indicators, we can better evaluate how MSA hunter-gatherers made decisions about the use of resources across the landscape. Our data indicate that while tree cover similar to modern miombo woodland and evergreen gallery forest prevailed at most times, MSA hunter-gatherers chose more locally open environments for activities that resulted in a lithic artifact record at multiple locations between 51 and 15 ka.publishedVersio

    Factor Structure of the CES-D and Measurement Invariance Across Gender in Mainland Chinese Adolescents

    ObjectivesThe primary aim was to examine the depressive symptom structure of Mainland China adolescents using the Center for Epidemiologic Studies Depression Scale (CES-D). DesignExploratory factor analysis (EFA) and confirmatory factor analysis (CFA) were simultaneously conducted to determine the structure of the CES-D in a large scale, representative adolescent samples recruited from Mainland China. Multigroup CFA (N=5059, 48% boys, mean=16.551.06) was utilized to test the factorial invariance of the depressive symptom structure, which was generated by EFA and confirmed by CFA across gender. ResultsThe CES-D can be interpreted in terms of 3 symptom dimensions. Additionally, factorial invariance of the new proposed model across gender was supported at all assuming different degrees of invariance.     Ribosomal RNA 2â€ČO-methylation as a novel layer of inter-tumour heterogeneity in breast cancer

    International audienceRecent epitranscriptomics studies unravelled that ribosomal RNA (rRNA) 2â€ČO-methylation is an additional layer of gene expression regulation highlighting the ribosome as a novel actor of translation control. However, this major finding lies on evidences coming mainly, if not exclusively, from cellular models. Using the innovative next-generation RiboMeth-seq technology, we established the first rRNA 2â€ČO-methylation landscape in 195 primary human breast tumours. We uncovered the existence of compulsory/stable sites, which show limited inter-patient variability in their 2â€ČO-methylation level, which map on functionally important sites of the human ribosome structure and which are surrounded by variable sites found from the second nucleotide layers. Our data demonstrate that some positions within the rRNA molecules can tolerate absence of 2â€ČO-methylation in tumoral and healthy tissues. We also reveal that rRNA 2â€ČO-methylation exhibits intra- and inter-patient variability in breast tumours. Its level is indeed differentially associated with breast cancer subtype and tumour grade. Altogether, our rRNA 2â€ČO-methylation profiling of a large-scale human sample collection provides the first compelling evidence that ribosome variability occurs in humans and suggests that rRNA 2â€ČO-methylation might represent a relevant element of tumour biology useful in clinic. This novel variability at molecular level offers an additional layer to capture the cancer heterogeneity and associates with specific features of tumour biology thus offering a novel targetable molecular signature in cancer

    Concerns about failed and fragile states have put state- and nation-building firmly on the academic and policy agenda, but the crucial role of public services in this process has remained underexplored. The 1960s and '70s generated a substantial set of literature that is largely missing from current writing. It identified state penetration, standardisation and accommodation as key processes in the state- and nation-building sequence. This article analyses these three processes in Western Europe in the seventeenth to nineteenth centuries, and the role of public services therein, to explore how they may help us to understand the success and failure of state- and nation-building in developing countries and fragile states. © The Authors 2011.     Low level of Fibrillarin, a ribosome biogenesis factor, is a new independent marker of poor outcome in breast cancer

    International audienceBackground: A current critical need remains in the identification of prognostic and predictive markers in early breast cancer. It appears that a distinctive trait of cancer cells is their addiction to hyperactivation of ribosome biogenesis. Thus, ribosome biogenesis might be an innovative source of biomarkers that remains to be evaluated. Methods: Here, fibrillarin (FBL) was used as a surrogate marker of ribosome biogenesis due to its essential role in the early steps of ribosome biogenesis and its association with poor prognosis in breast cancer when overexpressed. Using 3,275 non-metastatic primary breast tumors, we analysed FBL mRNA expression levels and protein nucleolar organisation. Usage of TCGA dataset allowed transcriptomic comparison between the different FBL expression levelsrelated breast tumours. Results: We unexpectedly discovered that in addition to breast tumours expressing high level of FBL, about 10% of the breast tumors express low level of FBL. A correlation between low FBL mRNA level and lack of FBL detection at protein level using immunohistochemistry was observed. Interestingly, multivariate analyses revealed that these low FBL tumors displayed poor outcome compared to current clinical gold standards. Transcriptomic data revealed that FBL expression is proportionally associated with distinct amount of ribosomes, low FBL level being associated with low amount of ribosomes. Moreover, the molecular programs supported by low and high FBL expressing tumors were distinct. Conclusion: Altogether, we identified FBL as a powerful ribosome biogenesis-related independent marker of breast cancer outcome. Surprisingly we unveil a dual association of the ribosome biogenesis FBL factor with prognosis. These data suggest that hyper-but also hypo-activation of ribosome biogenesis are molecular traits of distinct tumors

    Early human impacts and ecosystem reorganization in southern-central Africa

    Modern Homo sapiens engage in substantial ecosystem modification, but it is difficult to detect the origins or early consequences of these behaviors. Archaeological, geochronological, geomorphological, and paleoenvironmental data from northern Malawi document a changing relationship between forager presence, ecosystem organization, and alluvial fan formation in the Late Pleistocene. Dense concentrations of Middle Stone Age artifacts and alluvial fan systems formed after ca. 92 thousand years ago, within a paleoecological context with no analog in the preceding half-million-year record. Archaeological data and principal coordinates analysis indicate that early anthropogenic fire relaxed seasonal constraints on ignitions, influencing vegetation composition and erosion. This operated in tandem with climate-driven changes in precipitation to culminate in an ecological transition to an early, pre-agricultural anthropogenic landscape.info:eu-repo/semantics/publishedVersio

    The spectrum of ATM missense variants and their contribution to contralateral breast cancer

    Heterozygous carriers of ATM mutations are at increased risk of breast cancer. In this case-control study, we evaluated the significance of germline ATM missense variants to the risk of contralateral breast cancer (CBC). We have determined the spectrum and frequency of ATM missense variants in 443 breast cancer patients diagnosed before age 50, including 247 patients who subsequently developed CBC. Twenty-one per cent of the women with unilateral breast cancer and 17% of the women with CBC had at least one ATM germline missense variant, indicating no significant difference in variant frequency between these two groups. We have found that carriers of an ATM missense mutation, who were treated with radiotherapy for the first breast tumour, developed their second tumour on average in a 92-month interval compared to a 136-month mean interval for those CBC patients who neither received RT nor carried a germline variant, (p = 0.029). Our results indicate that the presence of ATM variants does not have a major impact on the overall risk of CBC. However, the combination of RT and (certain) ATM missense variants seems to accelerate tumour development
