    Kinematics of the Southern Rhodope Core Complex (North Greece)

    The Southern Rhodope Core Complex is a wide metamorphic dome exhumed in the northern Aegean as a result of large-scale extension from mid-Eocene to mid-Miocene times. Its roughly triangular shape is bordered on the SW by the Jurassic and Cretaceous metamorphic units of the Serbo-Macedonian in the Chalkidiki peninsula and on the N by the eclogite bearing gneisses of the Sideroneron massif. The main foliation of metamorphic rocks is flat lying up to 100 km core complex width. Most rocks display a stretching lineation trending NEâ SW. The Kerdylion detachment zone located at the SW controlled the exhumation of the core complex from middle Eocene to mid-Oligocene. From late Oligocene to mid-Miocene exhumation is located inside the dome and is accompanied by the emplacement of the synkinematic plutons of Vrondou and Symvolon. Since late Miocene times, extensional basin sediments are deposited on top of the exhumed metamorphic and plutonic rocks and controlled by steep normal faults and flat-ramp-type structures. Evidence from Thassos Island is used to illustrate the sequence of deformation from stacking by thrusting of the metamorphic pile to ductile extension and finally to development of extensional Plio-Pleistocene sedimentary basin. Paleomagnetic data indicate that the core complex exhumation is controlled by a 30� dextral rotation of the Chalkidiki block. Extensional displacements are restored using a pole of rotation deduced from the curvature of stretching lineation trends at core complex scale.     Urban Green Parks for Long-term Subjective Well-being: Empirical Relationships between Personal Characteristics, Park Characteristics, Park Use, Sense of Place, and Satisfaction with Life in the Netherlands

    As our living environment is becoming increasingly urbanized, this puts the livability, health, and quality of life in cities under pressure. Due to the urbanization process, urban green spaces are under threat of becoming scarce, while it is recognized that these green spaces can positively contribute to the subjective well-being of citizens. It is thus important to maximize the use and benefits derived from green spaces by designing them as positively experienced places. The aim of this research is to gain more empirical insights on the relationships between personal and park characteristics, park use behavior, sense of place, and park visitors’ long-term subjective well-being (i.e., life satisfaction). An online questionnaire was administered to participants in two medium-sized cities in The Netherlands, namely Eindhoven and ‘s-Hertogenbosch. Data were analyzed using a structural equation model. The results of this study show that the appreciation of facilities and the absence of disturbances positively influence the use and sense of place of a park. Furthermore, the findings show that sense of place has a positive influence on life satisfaction. The findings can be used by designers and policy-makers as guidelines to improve existing parks or to design new parks that support the subjective well-being of individuals in The Netherlands

    QCD and strongly coupled gauge theories : challenges and perspectives

    We highlight the progress, current status, and open challenges of QCD-driven physics, in theory and in experiment. We discuss how the strong interaction is intimately connected to a broad sweep of physical problems, in settings ranging from astrophysics and cosmology to strongly coupled, complex systems in particle and condensed-matter physics, as well as to searches for physics beyond the Standard Model. We also discuss how success in describing the strong interaction impacts other fields, and, in turn, how such subjects can impact studies of the strong interaction. In the course of the work we offer a perspective on the many research streams which flow into and out of QCD, as well as a vision for future developments.Peer reviewe

    Translation of non-canonical open reading frames as a cancer cell survival mechanism in childhood medulloblastoma

    A hallmark of high-risk childhood medulloblastoma is the dysregulation of RNA translation. Currently, it is unknown whether medulloblastoma dysregulates the translation of putatively oncogenic non-canonical open reading frames (ORFs). To address this question, we performed ribosome profiling of 32 medulloblastoma tissues and cell lines and observed widespread non-canonical ORF translation. We then developed a stepwise approach using multiple CRISPR-Cas9 screens to elucidate non-canonical ORFs and putative microproteins implicated in medulloblastoma cell survival. We determined that multiple lncRNA-ORFs and upstream ORFs (uORFs) exhibited selective functionality independent of main coding sequences. A microprotein encoded by one of these ORFs, ASNSD1-uORF or ASDURF, was upregulated, associated with MYC-family oncogenes, and promoted medulloblastoma cell survival through engagement with the prefoldin-like chaperone complex. Our findings underscore the fundamental importance of non-canonical ORF translation in medulloblastoma and provide a rationale to include these ORFs in future studies seeking to define new cancer targets

    Variation in Cell Wall Composition and Saccharification Potential of Seed‐Based Miscanthus Hybrids Grown on Marginal Lands Across Six European Trial Locations

    Miscanthus breeding programs have focused on developing intraspecific (M. sinensis × M. sinensis) and interspecific (M. sinensis × M. sacchariflorus) seed-based hybrids with distinct cell wall characteristics for different biomass value chains. Here, we evaluated the performance of 13 novel hybrids (including seed-based intraspecific, seed-based interspecific, and one clonally propagated interspecific hybrid) relative to Miscanthus × giganteus (M×g). We compared the cell wall composition, saccharification efficiency, and yield after spring harvests in 2021 and 2022 across six European locations. Cell wall content and composition varied significantly among hybrids and were influenced by environmental conditions, yet differences due to parental background were largely consistent across locations. On average, seed-based interspecific hybrids (80.6%–84.0% neutral detergent fiber) had a lower total cell wall content than the other hybrids evaluated in this study (88.3%–90.8%). In contrast, cellulose was ~5.5% higher in hybrids with an M. sinensis × M. sacchariflorus background relative to the intraspecific hybrids, while hemicellulose averaged above 34% for intraspecific hybrids, 29.4% to 31.8% in the interspecific hybrids, and below 27% for M×g. Lignin content was highest in M×g (~13.8%), intermediate in the interspecific hybrids (11.0%–12.2%), and lowest in the intraspecific hybrids (~10%). These compositional traits translated into saccharification efficiencies that were 32.9% higher for the intraspecific hybrids and 9.8%–13.1% higher for the interspecific hybrids (seed-based and clonally propagated) compared to M×g. Accounting for biomass yield, either several seed-based hybrids or the novel clonally propagated hybrid exceeded the theoretical ethanol potential of M×g at all trial locations, indicating strong potential for their use in lignocellulosic biofuel production

    Field Intercomparison of Radiometers Used for Satellite Validation in the 400–900 nm Range

    An intercomparison of radiance and irradiance ocean color radiometers (the second laboratory comparison exercise—LCE-2) was organized within the frame of the European Space Agency funded project Fiducial Reference Measurements for Satellite Ocean Color (FRM4SOC) May 8–13, 2017 at Tartu Observatory, Estonia. LCE-2 consisted of three sub-tasks: (1) SI-traceable radiometric calibration of all the participating radiance and irradiance radiometers at the Tartu Observatory just before the comparisons; (2) indoor, laboratory intercomparison using stable radiance and irradiance sources in a controlled environment; (3) outdoor, field intercomparison of natural radiation sources over a natural water surface. The aim of the experiment was to provide a link in the chain of traceability from field measurements of water reflectance to the uniform SI-traceable calibration, and after calibration to verify whether different instruments measuring the same object provide results consistent within the expected uncertainty limits. This paper describes the third phase of LCE-2: The results of the field experiment. The calibration of radiometers and laboratory comparison experiment are presented in a related paper of the same journal issue. Compared to the laboratory comparison, the field intercomparison has demonstrated substantially larger variability between freshly calibrated sensors, because the targets and environmental conditions during radiometric calibration were different, both spectrally and spatially. Major differences were found for radiance sensors measuring a sunlit water target at viewing zenith angle of 139° because of the different fields of view. Major differences were found for irradiance sensors because of imperfect cosine response of diffusers. Variability between individual radiometers did depend significantly also on the type of the sensor and on the specific measurement target. Uniform SI traceable radiometric calibration ensuring fairly good consistency for indoor, laboratory measurements is insufficient for outdoor, field measurements, mainly due to the different angular variability of illumination. More stringent specifications and individual testing of radiometers for all relevant systematic effects (temperature, nonlinearity, spectral stray light, etc.) are needed to reduce biases between instruments and better quantify measurement uncertainties

The indoor experiment of the LCE-2 has proven that uniform calibration just before the use of radiometers is highly effective. Distinct radiometers from different manufacturers operated by different scientists can yield quite close radiance and irradiance results (standard deviation s < 1%) under defined conditions. This holds when measuring stable lamp-based targets under stationary laboratory conditions with all the radiometers uniformly calibrated against the same standards just prior to the experiment. In addition, some unification of measurement and data processing must be settled. Uncertainty of radiance and irradiance measurement under these conditions largely consists of the sensor’s calibration uncertainty and of the spread of results obtained by individual sensors measuring the same object

    Application of IVDr NMR spectroscopy to stratify Parkinson's disease with absolute quantitation of blood serum metabolites and lipoproteins

    The challenge of early detection and stratification in Parkinson’s disease (PD) is urgent due to the current emergence of mechanism-based disease-modifying treatments. In here, metabolomic and lipidomic parameters obtained by a standardized and targeted in vitro diagnostic research (IVDr) platform have a significant potential to address therapy-related questions and generate improved biomarker panels. Our study aimed to use IVDr nuclear magnetic resonance (NMR) spectroscopy to quantify metabolites and lipoproteins in PD blood serum from different cohorts to stratify metabolically driven subtypes of idiopathic and genetic PD. Serum aliquots from three neurodegeneration biobank cohorts (287 samples in total, including 62 PD patient samples with GBA mutation, 98/43 PD patient samples of early/late stages of disease duration, 20 PD samples from patients with mutations in recessive PD genes and some smaller subgroups of mitochondrial and double mutation cases) were prepared and analyzed with IVDr NMR spectroscopy, covering 39 blood serum metabolites and 112 lipoprotein parameters. Uni- and multivariate statistics were used to identify metabolism-driven changes under consideration of typical confounders such as age, sex and disease duration and set into context with clinical biomarkers such as CSF concentrations of alpha-synuclein, neurofilament light chain, and tau protein. Based on the different PD subgroups we performed a total of eight different comparisons. Highlights from these comparisons include increased citrate and dimethylglycine with a decrease of creatinine and methionine in healthy controls and early PD group compared to GBA, PD late and recessive PD. We furthermore identified decreased HDL-3 free cholesterol in genetic PD cases compared to sporadic subject samples (sum of the PD early and PD late groups). Considering medication, we found that the levodopa equivalent daily dose (LEDD) is mostly positively correlated with tyrosine and citrate in sporadic PD compared to pyruvate and phenylalanine in genetic PD. Cerebrospinal fluid levels of alpha-synuclein were negatively correlated with alanine. Further metabolites and lipoproteins with discriminatory power for double mutation PD cases involved ornithine, 2-aminobutyrate and 2-hydroxybutyrate as well as for mitochondrial phenotypes via LDL phospholipid, apolipoprotein and cholesterol subfractions. Quantitative IVDr NMR serum spectroscopy is able to stratify PD patient samples of different etiology and can contribute to a wider understanding of the underlying metabolism-driven alterations e.g. in energy, amino acid, and lipoprotein metabolism. Though our overall cohort was large, major confounders such as age, sex and medication have a strong impact. That is why absolute quantification and detailed patient knowledge about metabolic confounders, is a premise for future translation of NMR serum spectroscopy to routine PD diagnostics
