Expressão e distribuição da conexina 32 em fígados de ratos com fibrose induzida experimentalmente
The connexin 32 (Cx32) is a protein that forms the channels that promote the gap junction intercellular communication (GJIC) in the liver, allowing the diffusion of small molecules through cytosol from cell-to-cell. Hepatic fibrosis is characterized by a disruption of normal tissue architeture by cellular lesions, and may alter the GJIC. This work aimed to study the expression and distribution of Cx32 in liver fibrosis induced by the oral administration of dimethylnitrosamine in female Wistar rats. The necropsy of the rats was carried out after five weeks of drug administration. They presented a hepatic fibrosis state. Sections from livers with fibrosis and from control livers were submitted to immunohistochemical, Real Time-PCR and Western-Blot analysis to Cx32. In fibrotic livers the Cxs were diffusely scattered in the cytoplasm, contrasting with the control livers, where the Cx32 formed junction plaques at the cell membrane. Also it was found a decrease in the gene expression of Cx32 without reduction in the protein quantity when compared with controls. These results suggest that there the mechanism of intercellular communication between hepatocytes was reduced by the fibrotic process, which may predispose to the occurrence of a neoplastic process, taken in account that connexins are considered tumor suppressing genes.A conexina 32 (Cx32) é uma proteína que constitui os canais que promovem as comunicações intercelulares via junções comunicantes (CIJC) no fígado, permitindo difusão de pequenas moléculas citoplasmáticas de uma célula à outra. A fibrose hepática caracteriza-se pela alteração da arquitetura normal do fígado e podem alterar as CIJCs. O objetivo deste trabalho foi estudar a expressão e distribuição de Cx32 na fibrose hepática. O objetivo do presente trabalho foi estudar a expressão e distribuição da Cx32 em fígados com fibrose induzida pela administração oral de dimetilnitrosamina em fêmeas de ratos Wistar. A necropsia foi realizada após cinco semanas da última administração da droga e observou-se um quadro de fibrose hepática. Amostras dos fígados com fibrose e de animais controle foram submetidas à análise imunoistoquímica, por Real Time-PCR e por Western-Blot verificando-se a presença de Cx32 difusa e dispersa no citoplasma dos fígados com fibrose. No grupo controle a Cx32 localizou-se na membrana citoplasmática com a formação de placas juncionais. O fígado com fibrose também revelou diminuição da expressão gênica de Cx32, embora sem a redução da quantidade do produto protéico, quando comparado ao grupo controle. Estes resultados sugerem que o mecanismo de comunicação intercelular entre os hepatócitos reduziu-se durante o processo fibrótico, o que pode predispor a ocorrência de processos neoplásicos, uma vez que as conexinas são consideradas genes supressores de tumores.FAPESPCNP
Ocean-floor sediments as a resource of rare earth elements: an overview of recently studied sites
The rare earth elements (REE), comprising 15 elements of the lanthanum series (La-Lu) together with yttrium (Y) and scandium (Sc), have become of particular interest because of their use, for example, in modern communications, renewable energy generation, and the electrification of transport. However, the security of supply of REE is considered to be at risk due to the limited number of sources, with dependence largely on one supplier that produced approximately 63% of all REE in 2019. As a result, there is a growing need to diversify supply. This has resulted in the drive to seek new resources elsewhere, and particularly on the deep-ocean floor. Here, we give a summary of REE distribution in minerals, versatile applications, and an update of their economic value. We present the most typical onshore methods for the determination of REE and examine methods for their offshore exploration in near real time. The motivation for this comes from recent studies over the past decade that showed ΣREE concentrations as high as 22,000 ppm in ocean-floor sediments in the Pacific Ocean. The ocean-floor sediments are evaluated in terms of their potential as resources of REE, while the likely economic cost and environmental impacts of deep-sea mining these are also considered
Joint toxicity of cadmium and ionizing radiation on zooplankton carbon incorporation, growth and mobility
The risk of exposure to radioactive elements is seldom assessed considering mixture toxicity, potentially over- or underestimating biological and ecological effects on ecosystems. This study investigated how three end points, carbon transfer between phytoplankton and Daphnia magna, D. magna mobility and growth, responded to exposure to γ-radiation in combination with the heavy metal cadmium (Cd), using the MIXTOX approach. Observed effects were compared with mixture effects predicted by concentration addition (CA) and independent action (IA) models and with deviations for synergistic/antagonistic (S/A), dose-level (DL), and dose-ratio (DR) dependency interactions. Several patterns of response were observed depending on the end point tested. DL-dependent deviation from the IA model was observed for carbon incorporation with antagonism switching to synergism at higher doses, while the CA model indicated synergism, mainly driven by effects at high doses of γ-radiation. CA detected antagonism regarding acute immobilization, while IA predicted DR-dependency. Both CA and IA also identified antagonism for daphnid growth. In general, effects of combinations of γ-radiation and Cd seem to be antagonistic at lower doses, but synergistic at the higher range of the doses tested. Our results highlight the importance of investigating the effects of exposure to γ-radiation in a multistressor context

Late Miocene to Quaternary Contourites Depositional Systems in the Gulf of Cadiz and West Portugal related to the Mediterranean - Atlantic exchange evolution: decoding bottom currents behaviour and oceanographic processes associated with gateways
Trabajo presentado en European Geophysical Union EGU General Assembly 2023, celebrada en Viena (Austria) entre el 23 y el 28 de mayo de 2023.Contourite depositional systems (CDS) represent the sedimentary records of paleoceanographic circulation and paleoclimatic changes throughout the geological timescale. These records offer expanded but contingent information relative to their adjacent marine gateways, documenting changes in the intensity and the direction of modern-day and paleo-current pathways on multicentennial, millennial and million-year timescales. This study investigates the late Miocene to Quaternary CDSs from the Gulf of Cadiz towards the West Iberian margin after the exit of the past Betic and Rifian corridors and most recent Strait of Gibraltar, the key gateways for the Mediterranean – Atlantic exchange trough time. Identifying new isatin derivatives with GSK-3 inhibition capacity through molecular docking and bioassays
The semi-synthesis of 11 isatin derivatives was achieved through bimolecular nucleophilic substitution and click chemistry. Seven new compounds were obtained. All chemical structures were determined by infrared spectroscopy (IR), nuclear magnetic resonance spectrometry (NMR) and high-resolution mass spectrometry (HRMS) data. These derivatives were evaluated for their anti-GSK-3 activity and all isatin derivatives (N-alkyl and 1,2,3-triazolic) exhibited strong inhibitory activity, with 2b and 4h exhibiting remarkable potency. In addition, docking studies were performed with 2b and 2e models to unravel the molecular mechanism underlying the polar interactions on the GSK-3 ATP-binding site.This study was funded by the Coordenação de Aperfeiçoamento de Pessoal de Nível Superior, Brasil

Chemical Constituents and Evaluation of Antimicrobial and Cytotoxic Activities of Kielmeyera coriacea
Many essential oils (EOs) of different plant species possess interesting antimicrobial effects on buccal microorganisms and cytotoxic properties. EOs of Kielmeyera coriacea Mart. & Zucc. were analyzed by gas chromatography coupled to mass spectrometry (GC-MS). The EO from leaves is rich in sesquiterpenes hydrocarbons and oxygenated sesquiterpenes. The three major compounds identified were germacrene-D (24.2%), (E)-caryophyllene (15.5%), and bicyclogermacrene (11.6%). The inner bark EO is composed mainly of sesquiterpenes hydrocarbons and the major components are alpha-copaene (14.9%) and alpha-(E)-bergamotene (13.0%). The outer bark EO is composed mainly of oxygenated sesquiterpenes and long-chain alkanes, and the major components are alpha-eudesmol (4.2%) and nonacosane (5.8%). The wood EO is mainly composed of long-chain alkanes and fatty acids, and the major components are nonacosane (9.7%) and palmitic acid (16.2%). The inner bark EO showed the strongest antimicrobial activity against the anaerobic bacteria Prevotella nigrescens (minimum inhibitory concentration-MIC of 50 µg mL−1). The outer bark and wood EOs showed MICs of 100 µg mL−1 for all aerobic microorganisms tested. The EOs presented low toxicity to Vero cells. These results suggest that K. coriacea, a Brazilian plant, provide initial evidence of a new and alternative source of substances with medicinal interest
Measurement of the mass and lifetime of the baryon
A proton-proton collision data sample, corresponding to an integrated
luminosity of 3 fb collected by LHCb at and 8 TeV, is used
to reconstruct , decays. Using the , decay mode for calibration, the lifetime ratio and absolute
lifetime of the baryon are measured to be \begin{align*}
\frac{\tau_{\Omega_b^-}}{\tau_{\Xi_b^-}} &= 1.11\pm0.16\pm0.03, \\
\tau_{\Omega_b^-} &= 1.78\pm0.26\pm0.05\pm0.06~{\rm ps}, \end{align*} where the

Constraints on the unitarity triangle angle from Dalitz plot analysis of decays
The first study is presented of CP violation with an amplitude analysis of
the Dalitz plot of decays, with , and . The analysis is based on a data sample corresponding to
of collisions collected with the LHCb detector. No
significant CP violation effect is seen, and constraints are placed on the
angle of the unitarity triangle formed from elements of the
Cabibbo-Kobayashi-Maskawa quark mixing matrix. Hadronic parameters associated
with the decay are determined for the first time. These
measurements can be used to improve the sensitivity to of existing and
