Antagonistic Interaction of Selenium and Cadmium in Human Hepatic Cells Through Selenoproteins
Cadmium (Cd) is a highly toxic heavy metal for humans and animals, which is associated with acute hepatotoxicity. Selenium (Se) confers protection against Cd-induced toxicity in cells, diminishing the levels of ROS and increasing the activity of antioxidant selenoproteins such as glutathione peroxidase (GPx). The aim of this study was to evaluate the antagonistic effect of selenomethionine (SeMet) against Cd toxicity in HepG2 cells, through the modulation of selenoproteins. To this end, the cells were cultured in the presence of 100 ”M SeMet and 5 ”M, 15 ”M, and 25 ”M CdCl2 and a combination of both species for 24 h. At the end of the experiment, cell viability was determined by MTT assay. The total metal content of Cd and Se was analyzed by triple-quadrupole inductively coupled plasmaâmass spectrometry (ICP-QqQ-MS). To quantify the concentration of three selenoproteins [GPx, selenoprotein P (SELENOP), and selenoalbumin (SeAlb)] and selenometabolites, an analytical methodology based on column switching and a species-unspecific isotopic dilution approach using two-dimensional size exclusion and affinity chromatography coupled to ICP-QqQ-MS was applied. The co-exposure of SeMet and Cd in HepG2 cells enhanced the cell viability and diminished the Cd accumulation in cells. Se supplementation increased the levels of selenometabolites, GPx, SELENOP, and SeAlb; however, the presence of Cd resulted in a significant diminution of selenometabolites and SELENOP. These results suggested that SeMet may affect the accumulation of Cd in cells, as well as the suppression of selenoprotein synthesis induced by Cd.This work was supported by the projects PG2018-096608-B-C21

Malaria surveillance from both ends: concurrent detection of Plasmodium falciparum in saliva and excreta harvested from Anopheles mosquitoes
Background: Malaria is the most important vector-borne disease in the world. Epidemiological and ecological studies of malaria traditionally utilize detection of Plasmodium sporozoites in whole mosquitoes or salivary glands by microscopy or serological or molecular assays. However, these methods are labor-intensive, and can over- or underestimate mosquito transmission potential. To overcome these limitations, alternative sample types have been evaluated for the study of malaria. It was recently shown that Plasmodium could be detected in saliva expectorated on honey-soaked cards by Anopheles stephensi, providing a better estimate of transmission risk. We evaluated whether excretion of Plasmodium falciparum nucleic acid by An. stephensi correlates with expectoration of parasites in saliva, thus providing an additional sample type for estimating transmission potential. Mosquitoes were exposed to infectious blood meals containing cultured gametocytes, and excreta collected at different time points post-exposure. Saliva was collected on honey-soaked filter paper cards, and salivary glands were dissected and examined microscopically for sporozoites. Excreta and saliva samples were tested by real time polymerase chain reaction (RT-rtPCR).
Results: Plasmodium falciparum RNA was detected in mosquito excreta as early as four days after ingesting a bloodmeal containing gametocytes. Once sporogony (the development of sporozoites) occurred, P. falciparum RNA was detected concurrently in both excreta and saliva samples. In the majority of cases, no difference was observed between the Ct values obtained from matched excreta and saliva samples, suggesting that both samples provide equally sensitive results. A positive association was observed between the molecular detection of the parasites in both samples and the proportion of mosquitoes with sporozoites in their salivary glands from each container. No distinguishable parasites were observed when excreta samples were stained and microscopically analyzed.
Conclusions: Mosquito saliva and excreta are easily collected and are promising for surveillance of malaria-causing parasites, especially in low transmission settings or in places where arboviruses co-circulate

Metabolites produced by antagonistic microbes inhibit the principal avocado pathogens in vitro
The demand for Hass avocado in the global market exceeds the supply by over 50%. Colombia has a remarkable advantage as a producer in the region due to its high yields. However, the productivity of this crop can be seriously affected by diseases such as root rot, caused by Phytophthora cinnamomi, postharvest body rot and stem end rot, caused by Colletotrichum sp. and Phomopsis sp., respectively. The potential of 76 bacterial isolates obtained from avocado rhizosphere to produce inhibitory metabolites against avocadoŽs pathogens was evaluated. The antagonistic effect of the rhizobacteria against P. cinnamomi, Colletotrichum sp. and Phomopsis sp. was tested through dual cultures. Thirty-six percent of the tested isolates presented inhibition halos against P. cinnamomi, 36% against Colletotrichum sp. and 67% against Phomopsis sp. Additionally, three isolates were selected for fermentation tests using different broth cultures. The extracts obtained from fermentations in the minimal medium of isolates ARP5.1 and AED06 showed inhibitory activity against the evaluated pathogens, but this effect was not observed with the AED26 extract. The media supplemented with copper chloride did not enhance activity of the extracts. These results suggest that using microbial metabolic extracts is a viable alternative for controlling avocado pathogens in vitro
The nature of the Cygnus extreme B supergiant 2MASS J20395358+4222505
2MASS J20395358+4222505 is an obscured early B supergiant near the massive OB star association Cygnus OB2. Despite its bright infrared magnitude (Ks = 5.82) it has remained largely ignored because of its dim optical magnitude (B = 16.63, V = 13.68). In a previous paper, we classified it as a highly reddened, potentially extremely luminous, early B-type supergiant. We obtained its spectrum in the U, B and R spectral bands during commissioning observations with the instrument MEGARA at the Gran Telescopio CANARIAS. It displays a particularly strong Hα emission for its spectral type, B1âIa. The star seems to be in an intermediate phase between supergiant and hypergiant, a group that it will probably join in the near (astronomical) future. We observe a radial velocity difference between individual observations and determine the stellar parameters, obtaining Teff = 24â000 K and logâgc = 2.88 ± 0.15. The rotational velocity found is large for a B supergiant, vâsinâi = 110 ± 25 kmsâ1â . The abundance pattern is consistent with solar, with a mild Câunderabundance (based on a single line). Assuming that J20395358+4222505 is at the distance of Cyg OB2, we derive the radius from infrared photometry, finding R = 41.2 ± 4.0 Râ, log(L/Lâ) = 5.71 ± 0.04 and a spectroscopic mass of 46.5 ± 15.0 Mâ. The clumped mass-loss rate (clumping factor 10) is very high for the spectral type, MË = 2.4 Ă 10â6 Mâ aâ1. The high rotational velocity and mass-loss rate place the star at the hot side of the bi-stability jump. Stellar Archaeology: a Keck Pilot Program on Extremely Metal- Poor Stars From the Hamburg/ESO Survey. III. The Lead (Pb) Star HE 0024-2523
We present a detailed abundance analysis, including spectral syntheses, of a
very metal-poor ([Fe/H]= -2.7), peculiar main sequence star, HE0024-2523
detected during the course of the Keck Pilot Program. Radial velocities of this
star were obtained during four different observing runs over a time span of 1.1
years, and demonstrate that it is clearly a short period spectroscopic binary.
An orbital solution was obtained, and orbital parameters were determined with
high precision. The rotational velocity was also measured (vsin i=9.71.5
kms); rotation appears likely to be synchronous with the orbit. The abundance
analysis and spectral syntheses indicate that the object is a CH star
characterized by extreme s-process enrichment, likely due to mass accretion
from an evolved companion which has now probably become a white dwarf. The lead
(Pb) abundance of HE0024-2523 is very high, the same as that of the recently
discovered lead-rich metal-poor star CS 29526-110, [Pb/Fe]=+3.3. The abundance
ratio of the heavy-s to light-s elements, as characterized by Pb and Ba,
[Pb/Ba]=+1.9, is the highest yet found for any metal-poor star, and is about
0.7 dex higher than that of CS29526-110. On the basis of the measured isotopic
ratio of carbon (12C/13C about 6) we argue that the mass donor must have had an
original mass of at least 3 Msun. The unusually short period of this CH star
suggests that it underwent a past common-envelope phase with its evolved
companion. Our results are compared to the latest available models for AGB
yields and s-process nucleosynthesis. We also discuss the possible connection
between HE0024-2523 the lithium depletion of halo stars, and halo blue

The state of the Martian climate
60°N was +2.0°C, relative to the 1981â2010 average value (Fig. 5.1). This marks a new high for the record. The average annual surface air temperature (SAT) anomaly for 2016 for land stations north of starting in 1900, and is a significant increase over the previous highest value of +1.2°C, which was observed in 2007, 2011, and 2015. Average global annual temperatures also showed record values in 2015 and 2016. Currently, the Arctic is warming at more than twice the rate of lower latitudes
Mitochondrial cristae-remodeling protein OPA1 in POMC neurons couples Ca2+ homeostasis with adipose tissue lipolysis
© 2021 The Authors.Appropriate cristae remodeling is a determinant of mitochondrial function and bioenergetics and thus represents a crucial process for cellular metabolic adaptations. Here, we show that mitochondrial cristae architecture and expression of the master cristae-remodeling protein OPA1 in proopiomelanocortin (POMC) neurons, which are key metabolic sensors implicated in energy balance control, is affected by fluctuations in nutrient availability. Genetic inactivation of OPA1 in POMC neurons causes dramatic alterations in cristae topology, mitochondrial Ca2+ handling, reduction in alpha-melanocyte stimulating hormone (α-MSH) in target areas, hyperphagia, and attenuated white adipose tissue (WAT) lipolysis resulting in obesity. Pharmacological blockade of mitochondrial Ca2+ influx restores α-MSH and the lipolytic program, while improving the metabolic defects of mutant mice. Chemogenetic manipulation of POMC neurons confirms a role in lipolysis control. Chemical Tagging in the Sdss-Iii/Apogee Survey: New Identifications of Halo Stars with Globular Cluster Origins
We present new identifications of five red giant stars in the Galactic halo with chemical abundance patterns that indicate they originally formed in globular clusters. Using data from the Apache Point Observatory Galactic Evolution Experiment (APOGEE) Survey available through Sloan Digital Sky Survey (SDSS) Data Release 12, we first identify likely halo giants, and then search those for the well-known chemical tags associated with globular clusters, specifically enrichment in nitrogen and aluminum. We find that 2% of the halo giants in our sample have this chemical signature, in agreement with previous results. Following the interpretation in our previous work on this topic, this would imply that at least 13% of halo stars originally formed in globular clusters. Recent developments in the theoretical understanding of globular cluster formation raise questions about that interpretation, and we concede the possibility that these migrants represent a small fraction of the halo field. There are roughly as many stars with the chemical tags of globular clusters in the halo field as there are in globular clusters, whether or not they are accompanied by a much larger chemically untaggable population of former globular cluster stars
