    Role of ‘One Stop Crisis Centre’ in Identifying and Assisting Victims of Violence in an Indian Health Care Setup

    Introduction: Cases of female targeted violence often go uncounted in India. To identify the unreported cases of violence, Sukoon was established in 2014 as a hospital-based ‘One Stop Crisis Centre’ (OSCC). Sukoon provides counselling, police assistance and legal aid to the victims. The aim of the present study was to recognize the role of Sukoon in preventing violence against women (VAW) in the region.Methods: Secondary data was extracted from 430 victims who approached Sukoon from August 2014 through January 2017. Data was collected on different variables: age, marital status, nature of violence, medium through which victims approached Sukoon and type of assistance provided. Significance of association of studied factors with the type of assault was investigated using ?2 test.Results: Age of study-victims ranged from 4 to 75 years with a median age of 26 years and mean age of 27.61 years with standard deviation of 10.56 years. Major types of VAW (96.51%) were domestic violence, sexual assault, physical assault and poisoning. The types of violences were significantly associated with victims’ age (?2 =5.76, d.f.=1, p<0.05) and marital status (?2 = 98.23, d.f=4, p<0.001). About 78% of victims were identified from Sukoon through screening and counseling. Around 69% of the cases were resolved directly by Sukoon or through police assistance.Conclusion: The above results indicate a significant role of Sukoon in screening victims of violence and providing them required assistance within the hospital environment in one location. Such centers should be further promoted by the government to address the issues of VAW

    Análise comparativa das atividades antioxidantes e anti-inflamatórias dos chás vermelho, azul e preto para benefícios à saúde

    The current COVID-19 predicament necessitates a greater emphasis on developing immunity. Herbal teas are abundant in antioxidants which are important for strengthening the immune system. Hot water decoction of Hibiscus rosa sinensis flowers (red tea), Clitoria ternatea flowers (blue tea) and commercially available black tea were evaluated by comparing for in vitro antioxidant and anti-inflammatory properties. Anthocyanin pigment in red, blue, and black tea demonstrated Rf values of 0.52, 0.86 and 0.78 respectively. Blue and black teas exhibited dominance of polyphenols, flavonoids, tannins, glycosides, terpenoids, saponins as compared to red tea. The highest total phenolic (12.25 ± 0.245 mg GAE/gm extract-1) and flavonoid (15.84 ± 0.268mg QE/gm extract-1) content were observed with black and blue tea respectively. Blue tea, and black tea extracts exhibited FRPA values of 1.81 ± 0.413 mg and 1.93 ± 0.178 mg AAE per gram extract-1 respectively. Black tea exhibits the highest antioxidant capacity in reducing molybdate ions (1.94 ± 0.354 mg AAE per gram extract-1) followed by blue tea (1.56 ± 0.199 mg AAE per gram extract-1). Blue tea extract at a very low concentration showed highest percentage hemolytic inhibition (57.14 ± 0.567%). According to the study, blue tea is a rich source of antioxidants with significant anti-inflammatory properties. The research may offer a valuable supplementary strategyfor its therapeutic applications.La situación actual de COVID-19 requiere un mayor énfasis en desarrollar inmunidad. Los tés de hierbas son ricos en antioxidantes, que son importantes para fortalecer el sistema inmunológico. Se evaluó la decocción en agua caliente de flores de Hibiscus rosa sinensis (té rojo), flores de Clitoria ternatea (té azul) y té negro comercialmente disponible para determinar sus propiedades antioxidantes y antiinflamatorias in vitro. El pigmento de antocianina en el té rojo, azul y negro demostró valores de Rf de 0,52, 0,86 y 0,78, respectivamente. Los tés azul y negro exhibieron un predominio de polifenoles, flavonoides, taninos, glucósidos, terpenoides y saponinas en comparación con el té rojo. Los niveles más altos de fenoles totales (12,25 ± 0,245 mg EAG/g de extracto-1) y flavonoides (15,84 ± 0,268 mg QE/g de extracto-1) se observaron en los tés negro y azul, respectivamente. Los extractos de té azul y té negro exhibieron valores de FRPA de 1,81 ± 0,413 mg y 1,93 ± 0,178 mg de AAE por gramo de extracto-1, respectivamente. El té negro tiene la mayor capacidad antioxidante en la reducción de iones molibdato (1,94 ± 0,354 mg AAE por gramo de extracto-1), seguido del té azul (1,56 ± 0,199 mg AAE por gramo de extracto-1). El extracto de té azul a muy baja concentración presentó el mayor porcentaje de inhibición hemolítica (57,14 ± 0,567%). Según el estudio, el té azul es una rica fuente de antioxidantes con importantes propiedades antiinflamatorias.     Biochemical and molecular characterization of adult patients with type I Gaucher disease and carrier frequency analysis of Leu444Pro - a common Gaucher disease mutation in India

    Abstract Background Gaucher disease is a rare pan-ethnic disorder which occurs due to an increased accumulation of undegraded glycolipid glucocerebroside inside the cells’ lysosomes. A beta-Glucosidase (GBA) gene defect results in glucocerebrosidase enzyme deficiency. Though the disease is mainly diagnosed in childhood, the adult manifestation is often missed or identified late due to the failure to recognize the heterogeneous clinical presentation. The present study includes seven unrelated Indian adult patients (age range: 20–40 years) having splenomegaly, with or without hepatomegaly, cytopenia and bone abnormality. Methods The biochemical investigation implicated measuring plasma chitotriosidase enzyme activity followed by confirmatory test of β-Glucosidase enzyme activity from the leukocytes. The molecular characterization involved patients’ initial screening for the common Gaucher mutation (Leu444Pro). Later, all patients were subjected to whole GBA gene coding region study using bidirectional Sanger sequencing. The population screening for common Gaucher disease mutation (Leu444Pro) was executed in 1200 unrelated and healthy Indian subjects by Restriction Fragment Length Polymorphism-Polymerase Chain Reaction technique. The allele frequency was calculated using Hardy-Weinberg formula. Results The biochemical analysis revealed a significant reduction in the β-Glucosidase activity in all the patients. Also, an elevated level of plasma Chitotriosidase activity in five patients supported their diagnosis of Gaucher disease. Sanger sequencing established four patients with homozygous variation and three patients with compound heterozygous variation in GBA gene. This study uncovers two missense variants (Ala448Thr and Val17Gly) not previously reported in Gaucher disease patients. Also the known mutations like Leu444Pro, Arg329Cys, Asp315Asn, Ser125Arg, and Arg395Cys were identified in these patients. The homology modeling suggested the destabilization of the protein structure due to novel variants. The Leu444Pro mutation screening in the Indian population spotted two people as a carrier. This emerged the carrier frequency of 1:600 along with wild-type allele frequency 0.97113 and mutant allele frequency 0.02887. Conclusions The study reports novel and known variants identified in the GBA gene in seven adult patients. The given study is the first report on the carrier frequency of the Leu444Pro mutant allele in an Indian population which will help understanding the burden and susceptibility of Gaucher disease to affect next generation in India
