    Removal of spermatozoa with externalized phosphatidylserine from sperm preparation in human assisted medical procreation: effects on viability, motility and mitochondrial membrane potential

    <p>Abstract</p> <p>Background</p> <p>Externalization of phosphatidylserine (EPS) occurs in apoptotic-like spermatozoa and could be used to remove them from sperm preparations to enhance sperm quality for assisted medical procreation. We first characterized EPS in sperms from infertile patients in terms of frequency of EPS spermatozoa as well as localization of phosphatidylserine (PS) on spermatozoa. Subsequently, we determined the impact of depleting EPS spermatozoa on sperm quality.</p> <p>Methods</p> <p>EPS were visualized by fluorescently-labeled annexin V binding assay. Double staining with annexin V and Hoechst differentiates apoptotic from necrotic spermatozoa. We used magnetic-activated cell sorting using annexin V-conjugated microbeads (MACS-ANMB) technique to remove EPS spermatozoa from sperm prepared by density gradient centrifugation (DGC). The impact of this technique on sperm quality was evaluated by measuring progressive motility, viability, and the integrity of the mitochondrial membrane potential (MMP) by Rhodamine 123.</p> <p>Results</p> <p>Mean percentages of EPS spermatozoa were 14% in DGC sperm. Four subpopulations of spermatozoa were identified: 70% alive, 3% early apoptotic, 16% necrotic and 11% late apoptotic or necrotic. PS were localized on head and/or midpiece or on the whole spermatozoa. MACS efficiently eliminates EPS spermatozoa. MACS combined with DGC allows a mean reduction of 70% in EPS and of 60% in MMP-disrupted spermatozoa with a mean increase of 50% in sperm survival at 24 h.</p> <p>Conclusion</p> <p>Human ejaculates contain EPS spermatozoa which can mostly be eliminated by DGC plus MACS resulting in improved sperm long term viability, motility and MMP integrity. EPS may be used as an indicator of sperm quality and removal of EPS spermatozoa may enhance fertility potential in assisted medical procreation.</p

    Implicit GPS-based bicycle route choice model using clustering methods and a LSTM network

    International audienceBiking is gaining in popularity all around the world as a healthy and environmentally friendly mode of transportation. Urban policies tend to encourage citizens to use bicycles. This can be done by creating new cycling infrastructures, the renovation of old ones or the deployment of bike-sharing systems (BSS). These policies having a cost, understanding and predicting the behavior of cyclists has become a necessity in order to optimize them. Classical methods analyzing cyclists’ route choices use external factors and generated choice sets of paths along with a logit model to create a discrete route choice model. Nevertheless, few studies focus on the predictive capacity that this type of model can offer. In this paper, we developed a prediction-centered bicycle route choice model. Our model is created without using external factors or choice sets of paths as in the more classical methods. The idea of our method is to use deep and machine learning algorithms on GPS tracks. These algorithms learn representations from the data which replace explicit factors. To build the model, we clustered the GPS tracks using DBSCAN. The clusters allow to identify the cyclists’ preferred road segments and are used to create paths using them. A method weighting the road graph weights is developed to create paths passing through the preferred road segments of a given cluster. A LSTM is finally trained in order to retrieve a cluster from a shortest path between an origin/destination pair. Tracks created by our model are more similar to the original GPS tracks than the shortest paths or tracks generated by a prominent path computation service

    Genetic diversity and structure in Arapaima gigas populations from Amazon and Araguaia-Tocantins river basins

    Background Arapaima gigas (Schinz, 1822) is the largest freshwater scaled fish in the world, and an emerging species for tropical aquaculture development. Conservation of the species, and the expansion of aquaculture requires the development of genetic tools to study polymorphism, differentiation, and stock structure. This study aimed to investigate genomic polymorphism through ddRAD sequencing, in order to identify a panel of single nucleotide polymorphisms (SNPs) and to simultaneously assess genetic diversity and structure in wild (from rivers Amazon, Solimões, Tocantins and Araguaia) and captive populations. Results Compared to many other teleosts, the degree of polymorphism in A. gigas was low with only 2.3% of identified RAD-tags (135 bases long) containing SNPs. A panel of 393 informative SNPs was identified and screened across the five populations. Higher genetic diversity indices (number of polymorphic loci and private alleles, Shannon’s Index and HO) were found in populations from the Amazon and Solimões, intermediate levels in Tocantins and Captive, and very low levels in the Araguaia population. These results likely reflect larger population sizes from less urbanized environments in the Amazon basin compared to Araguaia. Populations were significantly differentiated with pairwise FST values ranging from 0.086 (Amazon × Solimões) to 0.556 (Amazon × Araguaia).     Endothelial Secreted Factors Suppress Mitogen Deprivation-Induced Autophagy and Apoptosis in Glioblastoma Stem-Like Cells

    International audienceRapidly growing and highly vascularized tumors, such as glioblastoma multiforme, contain heterogeneous areas within the tumor mass, some of which are inefficiently supplied with nutrients and oxygen. While the cell death rate is elevated in such zones, tumor cells are still suspected to grow and survive independently of extracellular growth factors. In line with this, glioblastoma stem-like cells (GSCs) are found closely associated with brain vasculature in situ, and as such are most likely in a protected microenvironment. However, the behavior of GSCs under deprived conditions has not been explored in detail. Using a panel of 14 patient-derived GSCs, we report that ex vivo mitogen deprivation impaired self-renewal capability, abolished constitutive activation of the mTor pathway, and impinged on GSC survival via the engagement of autophagic and apoptotic cascades. Moreover, pharmacological inhibition of the mTor pathway recapitulated the mitogen deprivation scenario. In contrast, blocking either apoptosis or autophagy, or culturing GSCs with endothelial-secreted factors partly restored mTor pathway activation and rescued GSC survival. Overall, our data suggest that GSCs are addicted to mTor, as their survival and self-renewal are profoundly dependent on this signaling axis. Thus, as mTor governs the fate of GSCs under both deprivation conditions and in the presence of endothelial factors, it could be a key target for therapeutic purposes

    Massive open star clusters using the VVV survey II. Discovery of six clusters with Wolf-Rayet stars

    Context: The ESO Public Survey "VISTA Variables in the V\'ia L\'actea" (VVV) provides deep multi-epoch infrared observations for an unprecedented 562 sq. degrees of the Galactic bulge, and adjacent regions of the disk. In this survey nearly 150 new open clusters and cluster candidates have been discovered. Aims: This is the second in a series of papers about young, massive open clusters observed using the VVV survey. We present the first study of six recently discovered clusters. These clusters contain at least one newly discovered Wolf-Rayet (WR) star. Methods: Following the methodology presented in the first paper of the series, wide-field, deep JHKs VVV observations, combined with new infrared spectroscopy, are employed to constrain fundamental parameters for a subset of clusters. Results: We affirm that the six studied stellar groups are real young (2-7 Myr) and massive (between 0.8 and 2.2 10^3 Msol) clusters. They are highly obscured (Av ~ 5-24 mag) and compact (1-2 pc). In addition to WR stars, two of the six clusters also contain at least one red supergiant star. We claim the discovery of 8 new WR stars, and 3 stars showing WR-like emission lines which could be classified WR or OIf. Preliminary analysis provides initial masses of ~30-50 Msol for the WR stars.     Typologie des plantations villageoises de palmier à huile (Elaeis guineensis Jacq.) dans le département du Plateau au Bénin

    Une typologie des palmeraies villageoises du département du Plateau au sud-est du Bénin a été établie à travers : i) une étude cartographique de la couverture pédologique et d'occupation des sols, ii) des entretiens semi-structurés en assemblée générale avec des groupes de producteurs choisis de façon aléatoire dans chaque commune, iii) des entretiens semi-structurés individuels avec quatre vingt dix producteurs dont les palmeraies sont installées sur les terres de barre et étudiées par une analyse factorielle des correspondances multiples. Les terres de barre sont les sols les plus plantés avec le palmier à huile et Adja-Ouèrè et Sakété constituent les communes de grosse production. Les palmeraies du département présentent i) des caractéristiques très peu variables : la densité de plantation, le précédent cultural, les pratiques d'association des cultures vivrières et d'utilisation des engrais et ii) des caractéristiques variables et spécifiques : la restitution des sous-produits de transformation et le recyclage des feuilles d'élagage des palmiers. Parmi les cinq modalités de gestion des feuilles d'élagage identifiées, l'exportation des feuilles est une pratique dominante dans le département. Sur les terres de barre, on retrouve quatre types de palmeraies qui se différencient par la superficie, les pratiques culturales et l'âge des palmeraies.Mots clés: Palmeraies villageoises, sols, sud-Bénin, pratiques culturales, typologie, palmier à huile

We consider pure-exchange economies with infinite horizon, discrete time, uncertainty with a continuum of possible shocks at every date. The evolution of shocks follows a Markov process, and fundamentals depend continuously on shocks.
