    Investigation of Deterioration of Joints in Concrete Pavements: Field Study of Penetrating Sealers

    The objective of this research was to assess the efficacy of various waterproofing sealers applied to pavement joints with respect to limiting water ingress. The measure of water ingress was the chloride concentration profile as measured by scanning electron microscopy and energy dispersive spectroscopy. The fieldwork was conducted at the MnROAD facility. Cores were retrieved from pavements to assess the before condition. Various silane- and siloxane-based sealers were applied in 2013, and the pavements were exposed to service for two years. After two years, cores were retrieved. The chloride profiles for various pavement sites were compared before and after application of the sealer. No appreciable differences were noted. The lack of measured differences is attributed to the short time span allowed for ingress. The older pavements measured had a considerable degree of chloride ingress and, as such, small changes were difficult to detect. The newer pavement analyzed also showed no appreciable change, but it would be worth re-analyzing after more time has elapsed (e.g., two more years)

    On-line analysis of ash containing slurries

    The ash content of coal suspended in a slurry is determined by bombarding a sample of the slurry flowing past a window of a measuring chamber with radiation from an annular nucleonic source, such as Cm-244, for emitting radiation within the range of about 7 to about 30 KeV and causing the sample to emit both backscat- tered and iron fluorescent x-rays. These x-rays are detected by a radiation detector which produces first and second electrical signals representative of the intensity of each. The density of the sample flowing from the measuring chamber is measured, such as by a nucelonic density gauge, to produce an electrical signal representative of the density and the ash content is determined from the detected intensities of the backscattered and iron fluorescent x-rays and the sample density.https://digitalcommons.mtu.edu/patents/1093/thumbnail.jp

    Neuraminidase-deficient Sendai virus HN mutants provide protection from homologous superinfection

    Binding of hemagglutinin-neuraminidase proteins (HN) to sialylated receptors initiates the infection process of several paramyxoviruses, whereas later in the viral life cycle, the neuramindase (NA) activity of newly synthesized HN destroys all receptors. Prior to NA action, expressed HN has to bind the receptor. To evaluate this HN–receptor complex with respect to receptor inactivation, three temperature-sensitive Sendai virus HN mutants carrying amino acid exchanges at positions 262, 264 and/or 461 were created that uncoupled NA activity from receptor binding at 39°C. Interestingly, at elevated temperature, when there is no detectable neuramindase activity, all infected cells are protected against homologous superinfection. Mutated HN protein on the cell surface is mainly bound to sialylated cell-surface components but can be released by treatment with NA. Thus, continuous binding to HN already inactivates the receptors quantitatively. Furthermore, mutant HN bound to receptors is prevented from being incorporated into virus particles in the absence of NA. It is shown here for the first time that during paramyxoviral infection, quantitative receptor inactivation already occurs due to binding of receptors to expressed HN protein without involvement of NA and is independent of NA activity of viral progeny. NA subsequently functions in the release of HN from the complex, coupled with desialysation of receptors. These findings could have implications for further antiviral drug development

    Taxation and market power

    "We analyze the incidence and welfare effects of unit sales taxes in experimental monopoly and Bertrand markets. We find, in line with economic theory, that firms with no market power are able to shift a high share of a tax burden on to consumers, independent of whether buyers are automated or human players. In monopoly markets, a monopolist bears a large share of the burden of a tax increase. With human buyers, however, this share is smaller than with automated buyers as the presence of human buyers constrains the pricing behavior of a monopolist."     Gene Expression Profiles of Sporadic Canine Hemangiosarcoma Are Uniquely Associated with Breed

    The role an individual's genetic background plays on phenotype and biological behavior of sporadic tumors remains incompletely understood. We showed previously that lymphomas from Golden Retrievers harbor defined, recurrent chromosomal aberrations that occur less frequently in lymphomas from other dog breeds, suggesting spontaneous canine tumors provide suitable models to define how heritable traits influence cancer genotypes. Here, we report a complementary approach using gene expression profiling in a naturally occurring endothelial sarcoma of dogs (hemangiosarcoma). Naturally occurring hemangiosarcomas of Golden Retrievers clustered separately from those of non-Golden Retrievers, with contributions from transcription factors, survival factors, and from pro-inflammatory and angiogenic genes, and which were exclusively present in hemangiosarcoma and not in other tumors or normal cells (i.e., they were not due simply to variation in these genes among breeds). Vascular Endothelial Growth Factor Receptor 1 (VEGFR1) was among genes preferentially enriched within known pathways derived from gene set enrichment analysis when characterizing tumors from Golden Retrievers versus other breeds. Heightened VEGFR1 expression in these tumors also was apparent at the protein level and targeted inhibition of VEGFR1 increased proliferation of hemangiosarcoma cells derived from tumors of Golden Retrievers, but not from other breeds. Our results suggest heritable factors mold gene expression phenotypes, and consequently biological behavior in sporadic, naturally occurring tumors

    Endocytic regulation of alkali metal transport proteins in mammals, yeast and plants

    The relative concentrations of ions and solutes inside cells are actively maintained by several classes of transport proteins, in many cases against their concentration gradient. These transport processes, which consume a large portion of cellular energy, must be constantly regulated. Many structurally distinct families of channels, carriers, and pumps have been characterized in considerable detail during the past decades and defects in the function of some of these proteins have been linked to a growing list of human diseases. The dynamic regulation of the transport proteins present at the cell surface is vital for both normal cellular function and for the successful adaptation to changing environments. The composition of proteins present at the cell surface is controlled on both the transcriptional and post-translational level. Post-translational regulation involves highly conserved mechanisms of phosphorylation- and ubiquitylation-dependent signal transduction routes used to modify the cohort of receptors and transport proteins present under any given circumstances. In this review, we will summarize what is currently known about one facet of this regulatory process: the endocytic regulation of alkali metal transport proteins.
