    Desferrioxamine decreases NAD redox potential of intact red blood cells: evidence for desferrioxamine as an inducer of oxidant stress in red blood cells

    BACKGROUND: Desferrioxamine (DFO) is an important iron chelating agent. It has also been thought of as an agent with anti-oxidant potential as it chelates ferric iron in various parts of the body. However, there is evidence suggesting that it may paradoxically affect red blood cells (RBC) by inducing intracellular oxidant stress. To further understand the mechanism of DFO's interaction with RBC, we conducted a study to determine the effect of DFO upon RBC's redox status. METHODS: We examined NAD redox potential in intact RBC (N = 5) incubated with DFO. RBC were incubated with 6 mM DFO for 2 hours. RESULTS: Significant decreases in NAD redox potential were observed after incubation of RBC with 6 mM DFO. The mean decrease was 10.01 ± 1.98% (p < 0.0004). CONCLUSIONS: The data confirm the oxidant effect of DFO on RBC

    Identification of genes preferentially expressed in wheat egg cells and zygotes

    Wheat genes differentially expressed in the egg cell before and after fertilization were identified. The data support zygotic gene activation before the first cell division in wheat. To have an insight into fertilization-induced gene expression, cDNA libraries have been prepared from isolated wheat egg cells and one-celled zygotes. Two-hundred and twenty-six egg cell and 253 zygote-expressed EST sequences were determined. Most of the represented transcripts were detected in the wheat egg cell or zygote transcriptome at the first time. Expression analysis of fourteen of the identified genes and three controls was carried out by real-time quantitative PCR. The preferential expression of all investigated genes in the female gametophyte-derived samples (egg cells, zygotes, two-celled proembryos, and basal ovule parts with synergids) in comparison to the anthers, and the leaves were verified. Three genes with putative signaling/regulatory functions were expressed at a low level in the egg cell but exhibited increased (2-to-33-fold) relative expression in the zygote and the proembryo. Genes with high EST abundance in cDNA libraries exhibited strong expression in the egg cell and the zygote, while the ones coding for unknown or hypothetical proteins exhibited differential expression patterns with preferential transcript accumulation in egg cells and/or zygotes. The obtained data support the activation of the zygotic genome before the first cell division in wheat

    Texting with touchscreen and keypad phones - A comparison of thumb kinematics, upper limb muscle activity, exertion, discomfort, and performance

    This study aimed to compare thumb kinematics and upper limb muscle activity, and the influence of hand size, when texting on a keypad smartphone and a touchscreen smartphone. Furthermore, the study compared exertion, discomfort, and performance when texting on the two phones. The thumb kinematics were tracked using a 3D motion analysis system and muscle activity was registered in six upper limb muscles using surface electromyography in 19 participants. When texting on the touchscreen phone compared to the keypad phone thumb flexion (p = 0.008) and flexion/extension range of motion were smaller (p = 0.02), the thumb was on average less internally rotated (p = 0.02), and activity (50th and 90th percentile) of the thumb and forearm muscles was lower (p = 0.05). The differences in thumb flexion were found only in the group with shorter hands and the differences in muscle activity was found only in the group with longer hands. These findings suggest there are differences in risks for developing musculoskeletal disorders during smartphone use with different key activation mechanisms and different hand sizes

    Conventional anticancer therapies, such as chemotherapy, radiotherapy, and targeted therapy, are designed to kill cancer cells. However, the efficacy of anticancer therapies is not only determined by their direct effects on cancer cells but also by off-target effects within the host immune system. Cytotoxic treatment regimens elicit several changes in immune-related parameters including the composition, phenotype, and function of immune cells. Here we discuss the impact of innate and adaptive immune cells on the success of anticancer therapy. In this context we examine the opportunities to exploit host immune responses to boost tumor clearing, and highlight the challenges facing the treatment of advanced metastatic disease

    Opposing Roles of Membrane and Soluble Forms of the Receptor for Advanced Glycation End Products in Primary Respiratory Syncytial Virus Infection

    Respiratory syncytial virus (RSV), a common respiratory pathogen in infants and the older population, causes pulmonary inflammation and airway occlusion that leads to impairment of lung function. Here, we have established a role for receptor for advanced glycation end products (RAGE) in RSV infection. RAGE-deficient (ager−/−) mice were protected from RSV-induced weight loss and inflammation. This protection correlated with an early increase in type I interferons, later decreases in proinflammatory cytokines, and a reduction in viral load. To assess the contribution of soluble RAGE (sRAGE) to RSV-induced disease, wild-type and ager−/− mice were given doses of sRAGE following RSV infection. Of interest, sRAGE treatment prevented RSV-induced weight loss and neutrophilic inflammation to a degree similar to that observed in ager−/− mice. Our work further elucidates the roles of RAGE in the pathogenesis of respiratory infections and highlights the opposing roles of membrane and sRAGE in modulating the host response to RSV infection

    The role of conscious control in maintaining stable posture

This study aimed to examine the relationship between conscious control of movements, as defined by the Theory of Reinvestment (Masters & Maxwell, 2008; Masters, Polman, & Hammond, 1993), and both traditional and complexity-based COP measures. Fifty-three young adults (mean age = 20.93 ± 2.53 years), 39 older adults with a history of falling (mean age = 69.23 ± 3.84 years) and 39 older adults without a history of falling (mean age = 69.00 ± 3.72 years) were asked to perform quiet standing balance in single- and dual-task conditions. The results showed that higher scores on the Movement Specific Reinvestment Scale (MSRS; Masters, Eves, & Maxwell, 2005; Masters & Maxwell, 2008), a psychometric measure of the propensity for conscious involvement in movement, were associated with larger sway amplitude and a more constrained (less complex) mode of balancing in the medial–lateral direction for young adults only. Scores on MSRS explained approximately 10% of total variation in the medial–lateral sway measures. This association was not apparent under dual-task conditions, during which a secondary task was used to limit the amount of cognitive resources available for conscious processing. No relationship between postural control and score on the MSRS was found for either older adult fallers or non-fallers. Possible explanations for these results are discussed

    Constraints on the χ_(c1) versus χ_(c2) polarizations in proton-proton collisions at √s = 8 TeV

    The polarizations of promptly produced χ_(c1) and χ_(c2) mesons are studied using data collected by the CMS experiment at the LHC, in proton-proton collisions at √s=8  TeV. The χ_c states are reconstructed via their radiative decays χ_c → J/ψγ, with the photons being measured through conversions to e⁺e⁻, which allows the two states to be well resolved. The polarizations are measured in the helicity frame, through the analysis of the χ_(c2) to χ_(c1) yield ratio as a function of the polar or azimuthal angle of the positive muon emitted in the J/ψ → μ⁺μ⁻ decay, in three bins of J/ψ transverse momentum. While no differences are seen between the two states in terms of azimuthal decay angle distributions, they are observed to have significantly different polar anisotropies. The measurement favors a scenario where at least one of the two states is strongly polarized along the helicity quantization axis, in agreement with nonrelativistic quantum chromodynamics predictions. This is the first measurement of significantly polarized quarkonia produced at high transverse momentum
