En el Diagnóstico Situacional se da a conocer los aspectos socio-demográficos de la ciudad de Atuntaqui, así como aspectos históricos, geográficos, económicos y poblacionales del área de estudio, información que sirve para establecer aliados, oponentes, oportunidades y riesgos del sector objeto como una oportunidad para la apertura de un local de Pollos Texas Chicken. El Marco Teórico consta el sustento teórico y de análisis conceptual de los aspectos básicos del presente estudio. El Estudio de Mercado comprende el análisis de las diferentes variables como son el producto, precio, plaza y promoción ya que a través de esto se determina que existen oportunidades de posicionarse en el mercado debido a que hay la necesidad de diversificar y satisfacer la demanda alimenticia. El Estudio Técnico ¿ Ingeniería del Proyecto indica la localización de la microempresa, determinación del lugar idóneo para la apertura del local del Pollo Texas Chicken y requerimientos técnicos para el funcionamiento de la entidad. El Estudio Económico hace referencia a la inversión total requerida para la implementación de este proyecto, se hizo el cálculo de: el costo de oportunidad, tabla de amortización, depreciaciones, elaboración de estados financieros, evaluaciones del VAN, TIR, Periodo de recuperación, punto de equilibrio, indicadores financieros, Análisis EVA y por último el costo beneficio. La Estructura Organizacional comprende la razón social, además de la estructura jerárquica y descripción de cada cargo, reglamento interno de trabajo, código de ética y conducta.     Latino Businesses in Oregon & MWBE Certifications: Understanding the Barriers Faced by Latino Business Owners in Applying for and Obtaining the MWBE Certification

    17 pagesThis report focuses on the state of Latino businesses and the Minority and Women Business Enterprise Certification (MWBE), which is issued to qualifying businesses in the private, for-profit sector in the state of Oregon. Oregon's Latino population has been growing rapidly since the early 1990s, with Latinos now comprising 12.5% of the total state population (U.S. Census Bureau 2016). There has also been a rapid increase in the number of Latino businesses in Oregon, with the number of Latino-owned businesses increasing by 144% between 2002 and 2012 (U.S. Census Bureau 2006, 2015). Despite the growth in Oregon's Latino population and business community, a relatively small number of Latino owned small businesses have the MWBE certification. Currently, only 1.2% of all for-profit businesses owned by Latinos in the state of Oregon have the MBE certification. This report’s goal is to understand the specific barriers encountered by Latino business owners in Oregon when applying for the MBE certification. To do this, we spoke with government and non-government organizations that help business owners with the application process. We asked them where difficulties in the application process most frequently arise. We also conducted preliminary qualitative interviews with a select few Latino business owners that had already earned their MBE certification to help us document their experience with the application process. The report identifies five key areas of opportunity surrounding the MWBE certification process that could be tailored to meeting the needs of Latino business owners: ● Centralizing and simplifying the application process ● Making accessing application assistance more immediate and equitable ● Increasing targeted outreach to the Latino business community in regard to the certification ● Collaborating with government contractors to make the RFP and budget proposal processes for the certified MWBE firms that compete for government contracts less time consuming and more accessible. ● Increasing opportunities for language access and bilingual/bicultural services in English/Spanish at all levels and phases of the MWBE outreach, application, and certification process This report's also aims to create a foundation for further research on this important topic and investigate and develop potential state strategies for overcoming the barriers that Latino business owners face in obtaining their MBE certifications. With the ultimate goal of increasing their accessibility to resources that make the application process less daunting and navigable for more Latino businesses to earn the MBE certification. Further research will include conducting more qualitative interviews with Latino business owners from various sectors who have their MWBE certifications and solicit input on specific ways to improve and simplify the application process. We will also speak to staff at COBID Oregon and solicit suggestions for improving and simplifying the application process. Finally, we will speak with other state departments such as ODOT and the Department of Education to understand their experiences with issuing contracts to MWBEs and get feedback on ways to improve and streamline RFPs and budget requests received from competing for MWBE-certified firms

    System size and centrality dependence of the balance function in A + A collisions at sqrt s NN = 17.2 GeV

    Electric charge correlations were studied for p+p, C+C, Si+Si and centrality selected Pb+Pb collisions at sqrt s_NN = 17.2$ GeV with the NA49 large acceptance detector at the CERN-SPS. In particular, long range pseudo-rapidity correlations of oppositely charged particles were measured using the Balance Function method. The width of the Balance Function decreases with increasing system size and centrality of the reactions. This decrease could be related to an increasing delay of hadronization in central Pb+Pb collisions

    Effectiveness of EDACS Versus ADAPT Accelerated Diagnostic Pathways for Chest Pain: A Pragmatic Randomized Controlled Trial Embedded Within Practice

    Study objective A 2-hour accelerated diagnostic pathway based on the Thrombolysis in Myocardial Infarction score, ECG, and troponin measures (ADAPT-ADP) increased early discharge of patients with suspected acute myocardial infarction presenting to the emergency department compared with standard care (from 11% to 19.3%). Observational studies suggest that an accelerated diagnostic pathway using the Emergency Department Assessment of Chest Pain Score (EDACS-ADP) may further increase this proportion. This trial tests for the existence and size of any beneficial effect of using the EDACS-ADP in routine clinical care. Methods This was a pragmatic randomized controlled trial of adults with suspected acute myocardial infarction, comparing the ADAPT-ADP and the EDACS-ADP. The primary outcome was the proportion of patients discharged to outpatient care within 6 hours of attendance, without subsequent major adverse cardiac event within 30 days. Results Five hundred fifty-eight patients were recruited, 279 in each arm. Sixty-six patients (11.8%) had a major adverse cardiac event within 30 days (ADAPT-ADP 29; EDACS-ADP 37); 11.1% more patients (95% confidence interval 2.8% to 19.4%) were identified as low risk in EDACS-ADP (41.6%) than in ADAPT-ADP (30.5%). No low-risk patients had a major adverse cardiac event within 30 days (0.0% [0.0% to 1.9%]). There was no difference in the primary outcome of proportion discharged within 6 hours (EDACS-ADP 32.3%; ADAPT-ADP 34.4%; difference −2.1% [−10.3% to 6.0%], P=.65). Conclusion There was no difference in the proportion of patients discharged early despite more patients being classified as low risk by the EDACS-ADP than the ADAPT-ADP. Both accelerated diagnostic pathways are effective strategies for chest pain assessment and resulted in an increased rate of early discharges compared with previously reported rates

    Lysine harvesting is an antioxidant strategy and triggers underground polyamine metabolism

    Both single and multicellular organisms depend on anti-stress mechanisms that enable them to deal with sudden changes in the environment, including exposure to heat and oxidants. Central to the stress response are dynamic changes in metabolism, such as the transition from the glycolysis to the pentose phosphate pathway—a conserved first-line response to oxidative insults1,2. Here we report a second metabolic adaptation that protects microbial cells in stress situations. The role of the yeast polyamine transporter Tpo1p3,4,5 in maintaining oxidant resistance is unknown6. However, a proteomic time-course experiment suggests a link to lysine metabolism. We reveal a connection between polyamine and lysine metabolism during stress situations, in the form of a promiscuous enzymatic reaction in which the first enzyme of the polyamine pathway, Spe1p, decarboxylates lysine and forms an alternative polyamine, cadaverine. The reaction proceeds in the presence of extracellular lysine, which is taken up by cells to reach concentrations up to one hundred times higher than those required for growth. Such extensive harvest is not observed for the other amino acids, is dependent on the polyamine pathway and triggers a reprogramming of redox metabolism. As a result, NADPH—which would otherwise be required for lysine biosynthesis—is channelled into glutathione metabolism, leading to a large increase in glutathione concentrations, lower levels of reactive oxygen species and increased oxidant tolerance. Our results show that nutrient uptake occurs not only to enable cell growth, but when the nutrient availability is favourable it also enables cells to reconfigure their metabolism to preventatively mount stress protection

    Gene and cell therapy for cystic fibrosis: From bench to bedside

    Clinical trials in cystic fibrosis (CF) patients established proof-of-principle for transfer of the wild-type cystic fibrosis transmembrane conductance regulator (CFTR) gene to airway epithelial cells. However, the limited efficacy of gene transfer vectors as well as extra- and intracellular barriers have prevented the development of a gene therapy-based treatment for CF. Here, we review the use of new viral and nonviral gene therapy vectors, as well as human artificial chromosomes, to overcome barriers to successful CFTR expression. Pre-clinical studies will surely benefit from novel animal models, such as CF pigs and ferrets. Prenatal gene therapy is a potential alternative to gene transfer to fully developed lungs. However, unresolved issues, including the possibility of adverse effects on pre- and postnatal development, the risk of initiating oncogenic or degenerative processes and germ line transmission require further investigation.     Network analysis of sea turtle movements and connectivity: A tool for conservation prioritization

    Aim: Understanding the spatial ecology of animal movements is a critical element in conserving long-lived, highly mobile marine species. Analyzing networks developed from movements of six sea turtle species reveals marine connectivity and can help prioritize conservation efforts. Location: Global. Methods: We collated telemetry data from 1235 individuals and reviewed the literature to determine our dataset's representativeness. We used the telemetry data to develop spatial networks at different scales to examine areas, connections, and their geographic arrangement. We used graph theory metrics to compare networks across regions and species and to identify the role of important areas and connections. Results: Relevant literature and citations for data used in this study had very little overlap. Network analysis showed that sampling effort influenced network structure, and the arrangement of areas and connections for most networks was complex. However, important areas and connections identified by graph theory metrics can be different than areas of high data density. For the global network, marine regions in the Mediterranean had high closeness, while links with high betweenness among marine regions in the South Atlantic were critical for maintaining connectivity. Comparisons among species-specific networks showed that functional connectivity was related to movement ecology, resulting in networks composed of different areas and links. Main conclusions: Network analysis identified the structure and functional connectivity of the sea turtles in our sample at multiple scales. These network characteristics could help guide the coordination of management strategies for wide-ranging animals throughout their geographic extent. Most networks had complex structures that can contribute to greater robustness but may be more difficult to manage changes when compared to simpler forms. Area-based conservation measures would benefit sea turtle populations when directed toward areas with high closeness dominating network function.     Molecular Determinants of Survival Motor Neuron (SMN) Protein Cleavage by the Calcium-Activated Protease, Calpain

    Spinal muscular atrophy (SMA) is a leading genetic cause of childhood mortality, caused by reduced levels of survival motor neuron (SMN) protein. SMN functions as part of a large complex in the biogenesis of small nuclear ribonucleoproteins (snRNPs). It is not clear if defects in snRNP biogenesis cause SMA or if loss of some tissue-specific function causes disease. We recently demonstrated that the SMN complex localizes to the Z-discs of skeletal and cardiac muscle sarcomeres, and that SMN is a proteolytic target of calpain. Calpains are implicated in muscle and neurodegenerative disorders, although their relationship to SMA is unclear. Using mass spectrometry, we identified two adjacent calpain cleavage sites in SMN, S192 and F193. Deletion of small motifs in the region surrounding these sites inhibited cleavage. Patient-derived SMA mutations within SMN reduced calpain cleavage. SMN(D44V), reported to impair Gemin2 binding and amino-terminal SMN association, drastically inhibited cleavage, suggesting a role for these interactions in regulating calpain cleavage. Deletion of A188, a residue mutated in SMA type I (A188S), abrogated calpain cleavage, highlighting the importance of this region. Conversely, SMA mutations that interfere with self-oligomerization of SMN, Y272C and SMNΔ7, had no effect on cleavage. Removal of the recently-identified SMN degron (Δ268-294) resulted in increased calpain sensitivity, suggesting that the C-terminus of SMN is important in dictating availability of the cleavage site. Investigation into the spatial determinants of SMN cleavage revealed that endogenous calpains can cleave cytosolic, but not nuclear, SMN. Collectively, the results provide insight into a novel aspect of the post-translation regulation of SMN
