    Test of prototype liquid-water-content meter for aircraft use

    This report describes the effort undertaken to meet the objectives of National Science Foundation Grant ATM-9207345 titled 'Test of Prototype Liquid-Water-Content Meter for Aircraft Use.' Three activities were proposed for testing the new aircraft instrument, PVM-100A: (1) Calibrate the PVM-100A in a facility where the liquid-water-content (LWC) channel, and the integrated surface area channel (PSA) could be compared to standard means for LWC and PSA measurements. Scaling constant for the channels were to be determined in this facility. The fog/wind tunnel at ECN, Petten, The Netherlands was judged the most suitable facility for this effort. (2) Expose the PVM-100A to high wind speeds similar to those expected on research aircraft, and test the anti-icing heaters on the PVM-100A under typical icing conditions expected in atmospheric clouds. The high-speed icing tunnel at NRC, Ottawa, Canada was to be utilized. (3) Operate the PVM-100A on an aircraft during cloud penetrations to determine its stability and practicality for such measurements. The C-131A aircraft of the University of Washington was the aircraft of opportunity for these-tests, which were to be conducted during the 4-week Atlantic Stratocumulus Transition Experiment (ASTEX) in June of 1992

    Atomic force microscopy-based mechanobiology

    Mechanobiology emerges at the crossroads of medicine, biology, biophysics and engineering and describes how the responses of proteins, cells, tissues and organs to mechanical cues contribute to development, differentiation, physiology and disease. The grand challenge in mechanobiology is to quantify how biological systems sense, transduce, respond and apply mechanical signals. Over the past three decades, atomic force microscopy (AFM) has emerged as a key platform enabling the simultaneous morphological and mechanical characterization of living biological systems. In this Review, we survey the basic principles, advantages and limitations of the most common AFM modalities used to map the dynamic mechanical properties of complex biological samples to their morphology. We discuss how mechanical properties can be directly linked to function, which has remained a poorly addressed issue. We outline the potential of combining AFM with complementary techniques, including optical microscopy and spectroscopy of mechanosensitive fluorescent constructs, super-resolution microscopy, the patch clamp technique and the use of microstructured and fluidic devices to characterize the 3D distribution of mechanical responses within biological systems and to track their morphology and functional state.Peer ReviewedPostprint (published version

    Loss of Akt activity increases circulating soluble endoglin release in preeclampsia:identification of inter-dependency between Akt-1 and heme oxygenase-1

    Aims - Endothelial dysfunction is a hallmark of preeclampsia. Desensitization of the phosphoinositide 3-kinase (PI3K)/Akt pathway underlies endothelial dysfunction and haeme oxygenase-1 (HO-1) is decreased in preeclampsia. To identify therapeutic targets, we sought to assess whether these two regulators act to suppress soluble endoglin (sEng), an antagonist of transforming growth factor-ß (TGF-ß) signalling, which is known to be elevated in preeclampsia. Methods and results - Vascular endothelial growth factor-A (VEGF-A), fibroblast growth factor (FGF-2), angiopoietin-1 (Ang-1), and insulin, which all activate the PI3K/Akt pathway, inhibited the release of sEng from endothelial cells. Inhibition of the PI3K/Akt pathway, by overexpression of phosphatase and tensin homolog (PTEN) or a dominant-negative isoform of Akt (Aktdn) induced sEng release from endothelial cells and prevented the inhibitory effect of VEGF-A. Conversely, overexpression of a constitutively active Akt (Aktmyr) inhibited PTEN and cytokine-induced sEng release. Systemic delivery of Aktmyr to mice significantly reduced circulating sEng, whereas Aktdn promoted sEng release. Phosphorylation of Akt was reduced in preeclamptic placenta and this correlated with the elevated level of circulating sEng. Knock-down of Akt using siRNA prevented HO-1-mediated inhibition of sEng release and reduced HO-1 expression. Furthermore, HO-1 null mice have reduced phosphorylated Akt in their organs and overexpression of Aktmyr failed to suppress the elevated levels of sEng detected in HO-1 null mice, indicating that HO-1 is required for the Akt-mediated inhibition of sEng. Conclusion - The loss of PI3K/Akt and/or HO-1 activity promotes sEng release and positive manipulation of these pathways offers a strategy to circumvent endothelial dysfunction

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.     Parameterization of the Vertical Variability of Tropical Cirrus Cloud Microphysical and Optical Properties

    Cloud Integrating Nephelometers (CIN) were flown on the U. North Dakota Citation aircraft and the NASA WB-57 aircraft for the purpose of measuring in-situ the optical extinction coefficient and the asymmetry parameter (g) at a wavelength of 635 nm of primarily ice particles encountered during the NASA CRYSTAL-FACE study of large cumulus clouds (Cu) and their anvils found in the southern Florida region. The probes performance was largely successful and produced archived data for vertical profiles of extinction, asymmetry parameter, and effective radius (Re), the latter being obtained by combining CIN and CVI (total water; Oregon State U.) measurements. Composites of the CIN and CVI data describing the average microphysical and optical behavior of the Cu and their anvils showed the following: The extinction increases with height as a result of the size of the particles also decreasing with height as shown by the Re measurements; near the top of anvils the size of the primary ice particles is about 10-um radius; and the value of g does not vary significantly with height and has a mean value of about 0.73 consistent with the idea that ambient ice crystals are primarily of complex shape and reflect solar radiation more efficiently than particles of pristine crystal shape. Other observations include: The g measurements were found to be an indicator of the phase of the cloud permitting identification of the clouds with water droplets, rain, and ice; visual ranges as small as several tens of meters were occasionally found in "extinction cores" that coincided with strong updraft cores; and comparison of the cloud probes on the Citation showed significant disagreement
