    Long-term acclimation might enhance the growth and competitive ability of Microcystis aeruginosa in warm environments

The positive effect of global warming on the growth of cyanobacteria has been widely predicted, but long-term studies targeting their adaptive potential to higher temperature have not been carried out so far. Predicting the magnitude and impact of cyanobacterial blooms in the future as a response to global warming requires an understanding of how cyanobacteria might change in the long term due to climate change. 2. Here we examined the effect of exposing three Microcystis aeruginosa strains isolated in Romania to ambient (22°C) and high (26°C) temperature for 6 months. Then, the competitive ability of the strains after heat acclimation was evaluated, by analysing their impact on plankton community composition. 3. One of the three strains displayed significantly higher growth rates after 6 months of cultivation at higher temperatures. Following inoculation into a natural plankton community, the overall cyanobacterial abundance significantly increased in the cultures inoculated with this heat-acclimated strain of M. aeruginosa as compared to the ambient-acclimated version. The structure of eukaryotic communities was impacted by both inoculated cyanobacteria and temperature during the experiments. 4. The results of this study emphasise the high potential of cyanobacteria to respond to stressors, and highlight the fact that previous acclimation to warming is a critical factor in shaping the overall structure of plankton communities. 5. Our study strongly advocates for including a step of culture acclimation to future experimental conditions in research programmes aiming to better understand the long-term impact of climate change on aquatic ecosystems

    The combined impact of low temperatures and shifting phosphorus availability on the competitive ability of cyanobacteria

    In freshwater systems, cyanobacteria are strong competitors under enhanced temperature and eutrophic conditions. Understanding their adaptive and evolutionary potential to multiple environmental states allows us to accurately predict their response to future conditions. To better understand if the combined impacts of temperature and nutrient limitation could suppress the cyanobacterial blooms, a single strain of Microcystis aeruginosa was inoculated into natural phytoplankton communities with different nutrient conditions: oligotrophic, eutrophic and eutrophic with the addition of bentophos. We found that the use of the bentophos treatment causes significant differences in prokaryotic and eukaryotic communities. This resulted in reduced biodiversity among the eukaryotes and a decline in cyanobacterial abundance suggesting phosphorus limitation had a strong impact on the community structure. The low temperature during the experiment lead to the disappearance of M. aeruginosa in all treatments and gave other phytoplankton groups a competitive advantage leading to the dominance of the eukaryotic families that have diverse morphologies and nutritional modes. These results show cyanobacteria have a reduced competitive advantage under certain temperature and nutrient limiting conditions and therefore, controlling phosphorus concentrations could be a possible mitigation strategy for managing harmful cyanobacterial blooms in a future warmer climate

    Understanding the long-term response of key marine phytoplankton species to ongoing global changes is pivotal in determining how oceanic community composition will respond over the coming decades. To better understand the impact of ocean acidification and warming, we acclimated two strains of Skeletonema marinoi isolated from natural communities to three pCO2 (400 μatm, 600 μatm and 1000 μatm) for 8 months and five temperature conditions (7°C, 10°C, 13°C, 16°C and 19°C) for 11 months. These strains were then tested in natural microbial communities, exposed to three pCO2 treatments (400 μatm, 600 μatm and 1000 μatm). DNA metabarcoding of the 16S and 18S gene for prokaryotes and eukaryotes respectively was used to show differences in abundance and diversity between the three CO2 treatments. We found there were no significant differences in acclimated S. marinoi concentrations between the three pCO2 treatments, most likely due to the high variability these strains experience in their natural environment. There were significant compositional differences between the pCO2 treatments for prokaryotes suggesting that indirect changes to phytoplankton-bacteria interactions could be a possible driver of bacterial community composition. Yet, there were no differences for eukaryotic community composition, with all treatments dominated by diatoms (but not the acclimated S. marinoi) resulting in similar biodiversity. Furthermore, strain-specific differences in community composition suggests interactions between prokaryotic and eukaryotic taxa could play a role in determining future community composition.publishedVersio

    Use of thyroid hormones in hypothyroid and euthyroid patients: a 2022 THESIS questionnaire survey of members of the Latin American Thyroid Society (LATS)

    Abstract Purpose Inconsistencies in the medical management of hypothyroidism have been reported between endocrinologists in different countries. This study aimed to identify the attitudes of Latin America thyroid specialists towards the use of thyroid hormones. Methods Online survey of members of the Latin America Thyroid Society. Results 81/446 (18.2%) completed the questionnaire. Levothyroxine (LT4) was the initial treatment of choice for all respondents. 56.8% would consider LT4 use in biochemically euthyroid patients: infertile women with elevated anti-thyroid antibodies (46.9%), resistant depression (17.3%) and growing goiter (12%). Most respondents preferred tablets (39.5%) over liquid formulations (21.0%) or soft gel capsules (22.2%) and would not consider switching formulations in patients with persistent symptoms. 39.5% would never use LT4 + liothyronine (LT3) combination therapy in symptomatic euthyroid patients, due to low quality evidence for benefit. 60.5% reported that persistence of symptoms despite normal TSH is rare (below 5% of patients) and its prevalence has been stable over the last five years. Psychosocial factors (84.0%), comorbidities (86.4%) and the patient unrealistic expectation (72.8%) were considered the top three explanations for this phenomenon. Conclusion LT4 tablets is the treatment of choice for hypothyroidism. A significant proportion of respondents would use LT4 in some groups of euthyroid individuals, contrasting the recommendations of the major clinical practice guideline indications. LT4 + LT3 combination treatment in euthyroid symptomatic patients was considered by nearly 50%. Practices based on weak or absent evidence included use of thyroid hormones for euthyroid subjects by 56.8% of respondents and use of LT4 + LT3 treatment by 60.5% of respondents for patients with persistent symptoms. In contrast to many European countries, LATS respondents report a low and unchanged proportion of dissatisfied patients over the last five years

    Importância da ecogenicidade da tireóide no diagnóstico da tireoidite crônica auto-imune Value of thyroid echogenicity in the diagnosis of chronic autoimmune thyroiditis

    A tireoidite crônica auto-imune é, atualmente, a principal causa de hipotireoidismo e seu diagnóstico baseia-se nas manifestações clínico-laboratoriais. O marcador laboratorial mais importante é a presença de anticorpos antitireoglobulina e antiperoxidase, sendo este último o teste mais sensível. A biópsia aspirativa apresenta alta sensibilidade e especificidade, porém, é um método invasivo e, por isso, reservado para quando há presença de nódulo ou bócio de crescimento rápido. A cintilografia é desnecessária para o diagnóstico, já que apresenta baixa sensibilidade e especificidade. A ultra-sonografia, tanto ao modo B como ao dúplex-Doppler colorido, evoluiu de forma muito rápida e tornou-se um método simples, não-invasivo, reprodutível e com alta sensibilidade para o diagnóstico da tireoidite crônica auto-imune. Ao modo B, a ecogenicidade é um parâmetro de extrema importância, já que, além de apresentar alta correlação com o quadro citopatológico, também apresenta alta sensibilidade para o diagnóstico da tireoidite crônica auto-imune. Embora este parâmetro não seja específico da tireoidite crônica auto-imune, pois também pode estar presente na doença de Graves, na tireoidite pós-parto e na tireoidite subaguda, tais desordens podem ser facilmente diferenciadas tanto pelo quadro clínico-laboratorial quanto pelo dúplex-Doppler colorido. Assim, este artigo tem o objetivo de revisar a importância do estudo da ecogenicidade no diagnóstico da tireoidite crônica auto-imune.<br>Chronic autoimmune thyroiditis is currently considered as the main cause for hypothyroidism and its diagnosis is based on clinical manifestations and laboratory tests results. The most significant laboratory marker for this disease is the presence of anti-thyroperoxidase and anti-thyroglobulin antibodies, the latter being the most sensitive one. Aspiration biopsy shows high sensitivity and specificity but, considering the invasiveness of this method it is reserved for cases of suspected nodules or fast growing goiter. Scintigraphy is unnecessary for the diagnosis, considering its low specificity and sensitivity. Both B-mode and duplex color Doppler ultrasonography have rapidly evolved, becoming a simple, noninvasive and reproducible method with high sensitivity for the diagnosis of chronic autoimmune thyroiditis. Echogenicity is a relevant parameter at B-mode ultrasonography because of the close correlation with cytopathological findings besides the high sensitivity in the diagnosis of the disease. Although this is not a specific parameter for chronic autoimmune thyroiditis, considering that it may also be found in Graves' disease, postpartum thyroiditis and subacute thyroiditis, such disorders can be easily differentiated both by clinical-laboratory and duplex color Doppler findings.     Descrição do método de elaboração das evidênciasOs integrantes da Comissão de Esclerose Sistêmica da Sociedade Brasileira de Reumatologia (biênio 2010-2012) participaram do Curso de Elaboração de Evidências da Associação Médica Brasileira, em São Paulo, durante o primeiro semestre de 2011. As questões foram propostas e discutidas pela internet, no segundo semestre de 2011. As 15 questões clínicas consideradas relevantes foram estruturadas por meio da estratégia do P.I.C.O. (Paciente; Intervenção ou Indicador; Comparação; Outcome). As estratégias de busca avaliaram as bases de dados MEDLINE, EMBASE, Scielo/Lilacs, Cochrane Library até setembro de 2012 (Apêndice). Os artigos selecionados na primeira estratégia de busca foram submetidos à avaliação crítica das evidências, utilizando-se o escore de Jadad. Foram considerados também estudos observacionais e séries de casos na ausência de ensaios clínicos randomizados. Foi realizada inserção de estudos relevantes obtidos por busca manual. Posteriormente, foram elaboradas as respostas às perguntas das Recomendações, em que cada referência bibliográfica selecionada apresentava o correspondente grau de recomendação e força de evidência científica.
