Galectin-12 colocalizes with splicing factor-rich speckles and shuttles between the nucleus and cytoplasm in colon cancer cells
Several members of the glycan-binding family of galectins have been linked to colon cancer initiation and progression while the role of galectin-12 in this malignancy is largely unexplored. In previous studies we observed expression of galectin-12 in normal colon epithelium in contrast to its lack of expression in colorectal cancer tissues. In order to gain insight into its molecular function we established a genetically engineered colon cancer model cell line, which facilitates inducible and reconstituted expression of LGALS12 transgene at physiological levels in an isogenic background. Regulation of transgene expression by doxycycline was confirmed at the transcript- and protein level in a time- and dose-dependent manner for two independent clones. Reconstituted galectin-12 expression did not affect cell morphology and proliferation. Analysis of its subcellular distribution showed that galectin-12 resides in and shuttles between the nucleus and cytoplasm. More detailed analysis by double-immunofluorescence and confocal microscopy revealed colocalization of galectin-12 with splicing-factor rich nuclear speckles, hinting towards a potential role of galectin-12 in pre-mRNA splicing and processing. Therefore, the established model cell line represents a powerful tool to investigate the functional impact of galectin-12 reconstitution on colon cancer tumorigenesis

Organotypic cocultures as skin equivalents: A complex and sophisticated in vitro system
To assess the role of genes required for skin organogenesis, tissue regeneration and homeostasis, we have established in vitro skin equivalents composed of primary cells or cell lines, respectively. In these organotypic cocultures keratinocytes generate a normal epidermis irrespective of the species and tissue origin of fibroblasts. The combination of cells derived from mouse and human tissues facilitates the identification of the origin of compounds involved in epidermal tissue reconstitution and thus the precise analysis of growth regulatory mechanisms
Which conditions should be considered as disorders in the International Classification of Diseases (ICD-11) designation of “other specified disorders due to addictive behaviors”?

Other problematic behaviors may be considered as “other specified disorders due to addictive behaviors (6C5Y).” Methods: Narrative review, experts’ opinions. Results: We suggest the following meta-level criteria for considering potential addictive behaviors as fulfilling the category of “other specified disorders due to addictive behaviors”:1. Clinical relevance: Empirical evidence from multiple scientific studies demonstrates that the specific potential addictive behavior is clinically relevant and individuals experience negative consequences and functional impairments in daily life due to the problematic and potentially addictive behavior.2. Theoretical embedding: Current theories and theoretical models belonging to the field of research on addictive behaviors describe and explain most appropriately the candidate phenomenon of a potential addictive behavior.3. Empirical evidence: Data based on self-reports, clinical interviews, surveys, behavioral experiments, and, if available, biological investigations (neural, physiological, genetic) suggest that psychological (and neurobiological) mechanisms involved in other addictive behaviors are also valid for the candidate phenomenon.Varying degrees of support for problematic forms of pornography use, buying and shopping, and use of social networks are available.These conditions may fit the category of “other specified disorders due to addictive behaviors”. Conclusion: It is important not to overpathologize everyday-life behavior while concurrently not trivializing conditions that are of clinical importance and that deserve public health considerations. The proposed meta-level-criteria mayhelp guide both research efforts and clinical practice.Peer reviewe
Soil respiratory quotient determined via barometric process separation combined with nitrogen-15 labeling
The barometric process separation (BaPS) and ¹⁵N dilution techniques were used to determine gross nitrification rates on the same soil cores from an old grassland soil. The BaPS-technique separates the O₂ consumption into that from nitrification and that from soil organic matter (SOM) respiration. The most sensitive parameter for the calculations via the BaPS technique is the respiratory quotient (RQ = ∆CO₂/∆O₂) for SOM turnover (RQSOM). Combining both methods (BaPS–¹⁵N ) allowed the determination of the RQSOM. The RQ value determined in such a way is adjusted for the influence of nitrification and denitrification, which are both characterized by totally different RQ values. The results for the grassland soil showed that 6 to 10% of O₂ was consumed by nitrification when incubated at 20°C and 0.49 g H₂O g⁻¹ soil. A set of BaPS measurements with the same soil at various temperature and moisture contents showed that up to 49% of the total O₂ consumption was due to nitrification. The calculated RQSOM values via the BaPS–¹⁵N technique presented here are more closely associated with the overall SOM turnover than the usual net RQ reported in the literature. Furthermore, the RQSOM value provides an overall indication of the decomposability and chemical characteristics of the respired organic material. Hence, it has the potential to serve as a single state index for SOM quality and therefore be a useful index for SOM turnover models based on substrate quality

New genetic loci link adipose and insulin biology to body fat distribution.
Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms
The Science Performance of JWST as Characterized in Commissioning
This paper characterizes the actual science performance of the James Webb
Space Telescope (JWST), as determined from the six month commissioning period.
We summarize the performance of the spacecraft, telescope, science instruments,
and ground system, with an emphasis on differences from pre-launch
expectations. Commissioning has made clear that JWST is fully capable of
achieving the discoveries for which it was built. Moreover, almost across the
board, the science performance of JWST is better than expected; in most cases,
JWST will go deeper faster than expected. The telescope and instrument suite
have demonstrated the sensitivity, stability, image quality, and spectral range
that are necessary to transform our understanding of the cosmos through
