    Characterization of Genotype by Planting Date Effects on Runner-Type Peanut Seed Germination and Vigor Response to Temperature

    Experiments evaluated the genotype by environment effects on seed germination and vigor of the peanut runner-type cultivars ‘Georgia Green’, ‘AT3085R0’, ‘AT271516’, ‘Georgia 03L’, and ‘FR458’ grown under similar production practices, for three planting dates: April, May, and June in Georgia and Alabama. Objectives were to determine if time of planting and harvest dates would subsequently affect germination and vigor when tested using a thermal gradient devise (temperature range14 to 35 °C). Runner-type peanut seed grown in Dawson Georgia in 2008 had the strongest seed vigor with Germ80 of 22 to 40 growing degree days (GDD), and maximum incidence of germination rate 84.8-95.7% when planted April, May, and June 2008 across 15 seed lots. In contrast, seed harvested from plantings of May 2009 at Dawson Georgia exhibited Germ80 of 24 to 40 GDD with maximum incidence of germination rate 79.8-93.6%, but seed from April 2009 plantings had poor vigor of 56.8-72.8% and no amount of GDD could achieve Germ80, with similar results for June 2009 plantings for this location. For Headland April, May, and June 2009 plantings of the same cultivars, all seed had poor vigor, ≤75.6% maximum incidence for germination rate, and none obtained a measurable Germ80

    IRE1β negatively regulates IRE1α signaling in response to endoplasmic reticulum stress

    IRE1β is an ER stress sensor uniquely expressed in epithelial cells lining mucosal surfaces. Here, we show that intestinal epithelial cells expressing IRE1β have an attenuated unfolded protein response to ER stress. When modeled in HEK293 cells and with purified protein, IRE1β diminishes expression and inhibits signaling by the closely related stress sensor IRE1α. IRE1β can assemble with and inhibit IRE1α to suppress stress-induced XBP1 splicing, a key mediator of the unfolded protein response. In comparison to IRE1α, IRE1β has relatively weak XBP1 splicing activity, largely explained by a nonconserved amino acid in the kinase domain active site that impairs its phosphorylation and restricts oligomerization. This enables IRE1β to act as a dominant-negative suppressor of IRE1α and affect how barrier epithelial cells manage the response to stress at the host–environment interface

    Geographically widespread C-13-depletion of grazing caddis larvae:a third way of fuelling stream food webs?

    Stream ecosystems are supported by both green (i.e. based on grazing) and brown (i.e. detritus) food webs, whereas methane-derived carbon is not considered generally to be important; here, we add circumstantial evidence for this potential third way. Grazing cased-caddis (Trichoptera) larvae in the family Glossosomatidae can be very abundant in springs and headwaters and frequently have much lower stable carbon isotope ratios (i.e. they are depleted in the heavier C-13 stable isotope) than the biofilm (epilithon) on the upper surfaces of the stones on which they live, and which is their presumed diet. Evidence for similar isotopic depletion in other lotic invertebrates is currently limited, however; even for glossosomatids it has been observed so far only in some streams draining the southern English cretaceous chalk and in a few headwaters in northern California. If this phenomenon proves to be more widespread, among streams or taxa, it could imply a more general underpinning of stream food webs by isotopically light carbon derived from methane and accessed via consumers feeding on methanotrophic bacteria. Here, we sampled 58 stream sites to examine whether caddis larvae are also C-13-depleted in streams draining other geologies. We focused mainly on carboniferous limestone and sandstone, as well as on further chalk streams representative of most of the British chalk aquifer: together, these new sites covered an area of almost 90,000 km(2), around three times greater than that surveyed previously. At all 58 sites methane gas was supersaturated relative to the atmospheric equilibrium, and at 49 of them larvae were conspicuously C-13-depleted (from -17.5 parts per thousand to -3.6 parts per thousand) relative to the bulk epilithon (components of which we know can oxidise methane). Although still most pronounced on chalk, this phenomenon was geographically and geologically much more widespread than shown previously and suggests methane-derived carbon could indeed play a prominent role in stream food webs (i.e. the third way)

    Geobiology of the late Paleoproterozoic Duck Creek Formation, Western Australia

    The ca. 1.8 Ga Duck Creek Formation, Western Australia, preserves 1000 m of carbonates and minor iron formation that accumulated along a late Paleoproterozoic ocean margin. Two upward-deepening stratigraphic packages are preserved, each characterized by peritidal precipitates at the base and iron formation and carbonate turbidites in its upper part. Consistent with recent studies of Neoarchean basins, carbon isotope ratios of Duck Creek carbonates show no evidence for a strong isotopic depth gradient, but carbonate minerals in iron formations can be markedly depleted in C-13. In contrast, oxygen isotopes covary strongly with depth; delta O-18 values as positive as 2%. VPDB in peritidal facies systematically decline to values of 6 to 16% in basinal rocks, reflecting, we posit, the timing of diagenetic closure. The Duck Creek Formation contains microfossils similar to those of the Gunflint Formation, Canada; they are restricted to early diagenetic cherts developed in basinal facies, strengthening the hypothesis that such fossils capture communities driven by iron metabolism. Indeed, X-ray diffraction data indicate that the Duck Creek basin was ferruginous throughout its history. The persistence of ferruginous waters and iron formation deposition in Western Australia for at least several tens of millions of years after the transition to sulfidic conditions in Laurentia suggests that the late Paleoproterozoic expansion of sulfidic subsurface waters was globally asynchronous

    Translation of the International Physical Activity Questionnaire to Maltese and reliability testing.

    The International Physical Activity Questionnaire (IPAQ) is a widely used self-reported physical activity (PA) measure developed to allow for international cross-country comparisons. Due to its unavailability, the aim of this study was to translate the IPAQ-long to Maltese and undertake reliability testing. The IPAQ-long English version was translated into Maltese following the IPAQ guidelines, which included backwards translation. Maltese-speaking participants, aged between 18 and 69 years, were recruited through convenience sampling (n = 170). Participants completed the IPAQ-long twice within an 8- to 48-hr period. PA was calculated in MET minutes per week, and reliability was calculated using the Spearman correlation, intraclass correlation coefficient, concordance correlation coefficient, and Bland–Altman plots. A total of 155 participants completed the questionnaire at two time points. Spearman correlation was .83 (.76–.88) for total PA and .84 (.77–.89) for total sitting time. The intraclass correlation coefficient was .83 (.76–.88) and the concordance correlation coefficient was .75–.87 for total PA. The lowest reliability was for total transport, with a concordance correlation coefficient of .21−.45. Bland–Altman plots highlight that 95% of the differences fell within 2 SDs from the mean. Since the Maltese IPAQ-long has similar reliability to the English version, the authors recommend that health care professionals and PA practitioners use this tool when examining population-level PA among Maltese-speaking individual
